Subclinical mastitis (SCM) causes economic losses for dairy producers by reducing milk production and leading to higher incidence of clinical mastitis and premature culling. The prevalence of SCM in first-lactation heifers is highest during early lactation. The objective of this study was to estimate genetic parameters for SCM in early lactation in first-parity Holsteins. Somatic cell count test-day records were collected monthly in 91 Canadian herds participating in the National Cohort of Dairy Farms of the Canadian Bovine Mastitis Research Network. Only the first test-day record available between 5 and 30 d in milk was considered for analysis. The final data set contained 8,518 records from first lactation Holstein heifers. Six alternative traits were defined as indicators of SCM, using various cutoff values of SCC, ranging from 150,000 to 400,000 cells/mL. Both linear and threshold animal models were used. Overall prevalence of SCM using the 6 traits ranged from 13 to 24%. Heritability estimates (standard error) from linear and threshold models ranged from 0.037 to 0.057 (0.015 to 0.018) and from 0.040 to 0.051 (0.017 to 0.020), respectively. We found strong genetic correlations (standard error) among alternative SCC traits, ranging from 0.90 to 0.99 (0.013 to 0.069), indicating that these 6 traits were genetically similar. Despite low heritability, based on estimated breeding values (EBV) predicted from both models, we noted exploitable genetic variation among sires. Higher EBV of SCM resistance corresponded to sires with a higher percentage of daughters without SCM. Based on a linear model (all 6 traits), percentage of daughters with SCM ranged from 5 to 13% and from 19 to 33% for the top 10% and worst 10% of 69 sires with minimum 20 daughters in at least 5 herds, respectively. Spearman's rank correlations among EBV of sires predicted from linear (from 0.75 to 0.95) and threshold (from 0.74 to 0.95) models were moderate to high, respectively. Very high rank correlations (0.98 to 0.99) between EBV predicted for the same trait from linear and threshold model indicated that reranking of sires based on model used was minimal. In conclusion, despite low heritability, we found utilizable genetic variation in early lactation of heifers. Hence, genetic selection to improve genetic resistance to SCM in early lactation of heifers was deemed possible.  相似文献

An LC-MS/MS method for screening 14 aminoglycosides in foodstuffs of animal origin is presented. Its scope includes raw materials and processed ingredients but also finished products composed of milk, meat, fish, egg or fat. Aminoglycosides are extracted in an acidic aqueous solution, which is first recovered after centrifugation, then diluted with a basic buffer and finally purified by molecularly imprinted polymer-solid phase extraction (MIP-SPE). Analytes are detected within 8 min by ion-pair reversed phase LC-MS/MS. Due to the large range of foodstuffs involved, the variability of matrix effects led to significant MS signal variations. This was circumvented by systematically extracting each sample twice, i.e. ‘unspiked’ and ‘spiked’ at the screening target concentration of 50 µg kg?1. The method was validated according to the European Community Reference Laboratories Residues Guidelines giving false-negative and false-positive rates ≤3% for all compounds. Ruggedness of the method was further demonstrated in quality control operations by a second laboratory. The 14 aminoglycosides in water-based standard solutions were stable for up to 6 months when stored at either ?80°C, ?20°C or at 4°C storage temperatures.  相似文献

Over the past 100 yr, the range of traits considered for genetic selection in dairy cattle populations has progressed to meet the demands of both industry and society. At the turn of the 20th century, dairy farmers were interested in increasing milk production; however, a systematic strategy for selection was not available. Organized milk performance recording took shape, followed quickly by conformation scoring. Methodological advances in both genetic theory and statistics around the middle of the century, together with technological innovations in computing, paved the way for powerful multitrait analyses. As more sophisticated analytical techniques for traits were developed and incorporated into selection programs, production began to increase rapidly, and the wheels of genetic progress began to turn. By the end of the century, the focus of selection had moved away from being purely production oriented toward a more balanced breeding goal. This shift occurred partly due to increasing health and fertility issues and partly due to societal pressure and welfare concerns. Traits encompassing longevity, fertility, calving, health, and workability have now been integrated into selection indices. Current research focuses on fitness, health, welfare, milk quality, and environmental sustainability, underlying the concentrated emphasis on a more comprehensive breeding goal. In the future, on-farm sensors, data loggers, precision measurement techniques, and other technological aids will provide even more data for use in selection, and the difficulty will lie not in measuring phenotypes but rather in choosing which traits to select for.  相似文献

This study was conducted to examine the effect of active dry yeast (ADY) supplementation on lactation performance, ruminal fermentation patterns, and CH4 emissions and to determine an optimal ADY dose. Sixty Holstein dairy cows in early lactation (52 ± 1.2 DIM) were used in a randomized complete design. Cows were blocked by parity (2.1 ± 0.2), milk production (35 ± 4.6 kg/d), and body weight (642 ± 53 kg) and assigned to 1 of 4 treatments. Cows were fed ADY at doses of 0, 10, 20, or 30 g/d per head for 91 d, with 84 d for adaptation and 7 d for sampling. Although dry matter intake was not affected by ADY supplementation, the yield of actual milk, 4% fat-corrected milk, milk fat yield, and feed efficiency increased quadratically with increasing ADY supplementation. Yields of milk protein and lactose increased linearly with increasing ADY doses, whereas milk urea nitrogen concentration and somatic cell count decreased quadratically. Ruminal pH and ammonia concentration were not affected by ADY supplementation, whereas ruminal concentration of total volatile fatty acid increased quadratically. Digestibility of dry matter, organic matter, neutral detergent fiber, acid detergent fiber, nonfiber carbohydrate, and crude protein increased quadratically with increasing ADY supplementation. Supplementation of ADY did not affect blood concentration of total protein, triglyceride, aspartate aminotransferase, and alanine aminotransferase, whereas blood urea nitrogen, cholesterol, and nonesterified fatty acid concentrations decreased quadratically with increasing ADY supplementation. Methane production was not affected by ADY supplementation when expressed as grams per day or per kilogram of actual milk yield, dry matter intake, digested organic matter, and digested nonfiber carbohydrate, whereas a trend of linear and quadratic decrease of CH4 production was observed when expressed as grams per kilogram of fat-corrected milk and digested neutral detergent fiber. In conclusion, feeding ADY to early-lactating cows improved lactation performance by increasing nutrient digestibility. The optimal ADY dose should be 20 g/d per head.  相似文献

Enterobacter sakazakii has emerged as a rare cause of neonatal meningitis, septicemia and enterocolitis. Contaminated infant milk formula (IMF) has been identified as one infection route. A small number of clinical outbreaks have been epidemiologically linked to IMF contaminated post-pasteurization during manufacture and/or mishandled when reconstituted. Currently no agreed standardized typing protocol has been developed to trace E. sakazakii. The objectives of this study were to apply biochemical and genetic methods to characterize 51 environmental and food E. sakazakii isolates and 6 E. sakazakii type strains. Isolates were presumptively identified using biochemical profiles based on API 20E and ID32E methods and by culture on differential selective Druggan Forsythe Iversen (DFI) agar. Identification was subsequently confirmed by real time polymerase chain reaction (PCR). All but one of the isolates was identified as E. sakazakii by biochemical profiling. One isolate was identified as Escherichia vulneris by ID 32E and as Pantoea agglomerans by API 20E. All isolates produced green/blue colonies on DFI medium characteristic of this organism. Real time PCR could differentiate between E. sakazakii, Enterobacter spp. and other Enterobacteriacae. Analysis of RAPD banding patterns revealed 3 major clusters of E. sakazakii. There was a large degree of diversity noted amongst the remaining isolates. Our findings indicate that RAPD may be applied as a useful and reliable tool for direct comparison of E. sakazakii isolates providing traceability through the infant formula food chain.  相似文献

A thermophilic mediatorless microbial fuel cell (ML-MFC) was developed for continuous electricity production while treating artificial wastewater concurrently. A maximum power density of 1030 +/- 340 mW/m2 was generated continuously at 55 degrees C with an anode retention time of 27 min (11 mL h(-1)) and continuous pumping of air-saturated phosphate buffer into the cathode compartment at the retention time of 0.7 min (450 mL h(-1)). Meanwhile, about 80% of the electrons available from acetate oxidation were recovered as current. Denaturing gradient gel electrophoresis (DGGE) and direct 16S-rRNA gene analysis revealed that the bacterial diversity in this ML-MFC system was lower than the inoculum. Direct 16S rDNA analysis showed that the dominant bacteria representing 57.8% of total population in anode compartment was phylogenetically very closely related to an uncultured clone, clone E4. Two sheets of graphite used as the anode showed different dominant bacterial population. For the first time, it is shown that thermophilic electrochemically active bacteria can be enriched to concurrently generate electricity and treat artificial wastewater in a thermophilic ML-MFC.  相似文献

Classical treatments of shoulder instability are associated with recurrence. To determine whether the modification of the capsule properties may be an alternative procedure, the effect of crosslinking treatment on the structure and mechanical properties of diseased human shoulder capsules was investigated. Joint capsules harvested from patients during shoulder surgery (n = 5) were treated or not with UV and/or riboflavin (0.1%, 1.0% and 2.5%). The structure and the mechanical properties of the capsules were determined by atomic force microscopy. The effect of treatments on cell death was investigated. Collagen fibrils were well-aligned and adjacent to each other with a D-periodicity of 66.9 ± 3.2 nm and a diameter of 71.8 ± 15.4 nm in control untreated capsules. No effect of treatments was observed on the organization of the collagen fibrils nor on their intrinsic characteristics, including D-periodicity or their mean diameter. The treatments also did not induce cell death. In contrast, UV + 2.5% riboflavin induced capsule stiffness, as revealed by the increased Young’s modulus values (p < 0.0001 for each patient). Our results showed that the crosslinking procedure changed the biomechanics of diseased capsules, while keeping their structural organisation unchanged at the single fibril level. The UV/riboflavin crosslinking procedure may be a promising way to preserve the functions of collagen-based tissues and tune their elasticity for clinically relevant treatments.  相似文献

To control the spread of an infectious disease over a large network, the optimal allocation by a social planner of a limited resource is a fundamental and difficult problem. We address this problem for a livestock disease that propagates on an animal trade network according to an epidemiological–demographic model based on animal demographics and trade data. We assume that the resource is dynamically allocated following a certain score, up to the limit of resource availability. We adapt a greedy approach to the metapopulation framework, obtaining new scores that minimize approximations of two different objective functions, for two control measures: vaccination and treatment. Through intensive simulations, we compare the greedy scores with several heuristics. Although topology-based scores can limit the spread of the disease, information on herd health status seems crucial to eradicating the disease. In particular, greedy scores are among the most effective in reducing disease prevalence, even though they do not always perform the best. However, some scores may be preferred in real life because they are easier to calculate or because they use a smaller amount of resources. The developed approach could be adapted to other epidemiological models or to other control measures in the metapopulation setting.  相似文献

Two diets containing reduced levels of crude protein for both growing (35–65 kg) and finishing (65–95 kg) pigs were tested against standard commercial diets to determine changes in the odorants produced in fresh slurry. The experimental diets were formulated on a least cost basis using either a full commercial database or with restriction mainly to home-grown (UK) cereals and pulses. Synthetic essential amino acids were included in both formulations to obtain essential amino acids in an ideal protein ratio, with the crude protein being as low as possible. Nitrogen excretion by both growing and finishing pigs offered the low crude protein diets was significantly lower (P<0·001) than that of pigs offered the commercial diets. Major odorants responsible for pig odour from slurries were identified and their concentrations determined using gas chromatography–mass spectrometry. Five out of 10 odorants from slurries produced by growing pigs were significantly lower (P<0·05) in concentration for each of the low crude protein diets compared with the commercial diet. For the finishing pigs, nine out of ten odorants from slurries for the least cost and four odorants for the home grown diets were significantly lower (P<0·05) than slurry produced from the commercial diet. Reducing dietary nitrogen and providing essential amino acids in an ideal protein ratio not only decreases the nitrogen excreted by the pig, but also reduces odorants produced in the resultant slurries.  相似文献
