Safety analysis can be labour intensive and error prone for system designers. Moreover, even a relatively minor change to a system’s design can necessitate a complete reworking of the system safety analysis. This paper proposes the use of Behavior Trees and model checking to automate Cut Set Analysis (CSA) : that is, the identification of combinations of component failures that can lead to hazardous system failures. We demonstrate an automated incremental approach to CSA, in which models are extended incrementally and previous results incorporated in such a way as to significantly reduce the time and effort required for the new analysis. The approach is demonstrated on a case study concerning the hydraulics systems for the Airbus A320 aircraft.  相似文献

Melanoma is a lethal form of skin cancer triggered by genetic and environmental factors. Excision of early-stage, poorly aggressive melanoma often leads to a successful outcome; however, left undiagnosed these lesions can progress to metastatic disease. This research investigates whether the exposure of poorly aggressive melanoma to certain normal skin cells can explain how non-metastatic melanoma becomes more aggressive while still confined to the skin. To this end, we used a serial co-culture approach to sequentially expose cells from two different, poorly aggressive human melanoma cell lines against normal cells of the skin beginning with normal melanocytes, then epidermal keratinocytes, and finally dermal fibroblasts. Protein extraction of melanoma cells occurred at each step of the co-culture sequence for western blot (WB) analysis. In addition, morphological and functional changes were assessed to detect differences between the serially co-cultured melanoma cells and non-co-cultured cells. Results show that the co-cultured melanoma cells assumed a more mesenchymal morphology and displayed a significant increase in proliferation and invasiveness compared to control or reference cells. WB analysis of protein from the co-cultured melanoma cells showed increased expression of Snail and decreased levels of E-cadherin suggesting that epithelial-to-mesenchymal transition (EMT) is occurring in these co-cultured cells. Additional WB analysis showed increased levels of Nodal protein and signaling and signs of increased Wnt activity in the co-cultured melanoma cells compared to reference cells. These data suggest that interaction between poorly aggressive melanoma cells with normal cells of the skin may regulate the transition from localized, poorly aggressive melanoma to invasive, metastatic disease via Nodal and/or Wnt induced EMT.  相似文献

Quaternary ammonium compounds (QACs) are a vital class of antiseptics. Recent investigations into their construction are uncovering novel and potent multicationic variants. Based on a trisQAC precedent, we have implemented a scaffold‐hopping approach to develop alternative QAC architectures that display 1–3 long alkyl chains in specific projections from cyclic and branched core structures bearing 3–4 nitrogen atoms. The preparation of 30 QAC structures allowed for correlation of scaffold structure with antimicrobial activity. We identified QACs with limited conformational flexibility that have improved bioactivity against planktonic bacteria as compared to their linear counterparts. We also confirmed that resistance, as evidenced by an increased minimum inhibitory concentration (MIC) for methicillin‐resistant Staphylococcus aureus (MRSA) compared to methicillin‐susceptible Staphylococcus aureus (MSSA), can reduce efficacy up to 64‐fold for monocationic QACs. Differentiation of antimicrobial and anti‐biofilm activity, however, was not observed, suggesting that these compounds utilize a non‐specific mode of eradication.  相似文献

Hollandite materials, as a class of crystalline nuclear waste forms, are promising candidates for the immobilization of radioactive elements, such as Cs, Ba, as well as a variety of lanthanide and transition-metal fission products. In this study, three Ga-doped titanate hollandite-type phases, Ba1.33Ga2.67Ti5.33O16, Ba0.667Cs0.667Ga2Ti6O16, and Cs1.33Ga1.33Ti6.67O16, were synthesized using a solid-state reaction route. All synthesized phases adopted a single phase tetragonal structure, as determined by powder X-ray diffraction (XRD), and elemental analysis confirmed the measured stoichiometries were close to targeted compositions. Extended X-ray absorption fine structure spectroscopy (EXAFS) was used to determine the local structural features for the framework of octahedrally coordinated cations. EXAFS data indicated that Cs1.33Ga1.33Ti6.67O16 possessed the most disordered local structure centered around the Ga dopant. The enthalpies of formation of all three hollandite phases measured using high-temperature oxide melt solution calorimetry were found to be negative, indicating enthalpies of formation of these hollandites from oxides are thermodynamically stable with respect to their constituent oxides. Furthermore, the formation enthalpies were more negative and hence more favorable with increased Cs content. Finally, aqueous leaching tests revealed that high Cs content hollandite phases exhibited greater Cs retention as compared to low Cs content hollandite. While preliminary in nature, this work draws attention to the links between the capacity for radionuclide retention, atomistic level structural features and bulk thermodynamic properties of materials.  相似文献

Describes 2 common shortcomings of assessment procedures in agencies that deal with client problems and adjustment: the concentration on negative aspects of functioning, with insufficient concern for positive aspects, and the focus on personal attributes and behavior, with insufficient attention to environmental aspects. These deficiencies distort the realistic assessment of client problems and interfere with optimal treatment plans. It is suggested that client assessments take into account environmental circumstances and that as much attention be devoted to clients' strengths as to their weaknesses. Fluoride is a potentially toxic ion that occurs in aquifers both naturally and as a result of anthropogenic activity. Sometimes remediation of the aquifer is required. One potential aquifer decontamination strategy is an “interception-sorption trench”—one of a number of “reactive wall” technologies. This remediation strategy relies upon natural hydraulic gradients to transport the fluoride through the aquifer to the interception-sorption trench where it partitions onto a strong sorbent—alumina. In this paper, the focus is on the development and calibration of an equilibrium-based geochemical model that will be employed in the development of a quantitative reactive transport model, which in turn will be used for the design of an interception-sorption trench. The geochemical model described here takes into account a variety of ions likely to be present in a sandy aquifer, chemical activities, and the surface charge on the alumina. The model is calibrated over a wide pH range and for high initial fluoride concentrations using experimental results obtained from batch tests. It is found that pH dependent equilibrium constants are needed to capture the behavior of the experimentally observed fluoride sorption. The presence of sodium sulfate in solution is investigated, and it is found that sodium significantly interferes with the sorption of fluoride onto alumina under alkaline conditions. The geochemical model indicates that under acidic conditions, the alumina may release potentially large and unacceptable concentrations of aluminum into the aquifer. As a way of managing this potential problem, it is proposed that aluminum concentrations in the pore fluid may be mitigated by the inclusion of tree bark within the interception-sorption trench.  相似文献

Third grade children (N = 404) and their mothers completed questionnaires and participated in interviews designed to identify children's friendships across multiple contexts, determine levels of social network closure for these friendships, and assess child well-being. Cluster analyses revealed distinct patterns in the contexts in which children's friendships were maintained. Closure was highest for children whose friendship clusters heavily represented relatives as friends and lowest when friends were from schools and the broader community. Intermediate levels of closure were observed for the clusters of neighborhood friends and friends from church and school. Wrought magnesium alloys have been extensively used in the aerospace, electronics and automotive industries, where component weight is of concern and ambient temperatures remain below 100 °C. Undesirable creep relaxation of the wrought alloys above this temperature has been generally attributed to grain boundary sliding and plastic deformation leading to intergranular failure. The objective of this study was to investigate the compressive creep performance and microstructure of two wrought magnesium alloys (AE42 and ZE10) developed for high temperature applications. The total deformation of the AE42 and ZE10 alloys was 2.4 and 0.2 %, respectively, after 24 h creep test at 175 °C and 50 MPa. The poor creep performance of the AE42 alloy was explained via neutron diffraction studies which revealed that the elastic compressive response of $ (10\bar{1}0),\;(10\bar{1}1)\;{\text{and}}\;(2\bar{1}\bar{1}0) $ planes was significantly more anisotropic in the AE42 than in the ZE10 alloy. Further, microstructural analysis revealed ~10 % increase in grain size due to creep, with additional $ (10\bar{1}2) $ and $ (11\bar{2}1) $ twinning in the AE42 alloy. Precipitation of β-Mg17Al12 phase in the AE42 alloy possibly contributed to grain boundary sliding and high plastic strain during creep testing.  相似文献

At present a large number of patients with atherosclerotic disease are not receiving aggressive cholesterol-lowering therapy. Consequently they are being deprived of a cost-effective, risk-reducing treatment. Every physician who treats patients with clinical atherosclerotic disease should become fully informed about the results of cholesterol-lowering trials in patients at high risk. All physicians who care for high-risk patients should take responsibility for cholesterol management, including primary care physicians and cardiovascular specialists. Highly effective and generally safe drugs for cholesterol lowering are available. The benefits of therapy for reducing recurrent CHD and prolonging life are considerable. There is no justification for unduly delaying institution of therapy for the majority of patients. The many advantages of nonpharmaceutical therapy call for its use in almost all patients, but drug treatment should not be postponed if the target for LDL cholesterol lowering (< or = 100 mg/dL) is unlikely to be achieved in the near term by a nonpharmaceutical approach alone. The view that patients with CHD or other forms of atherosclerotic disease do not receive substantial clinical benefits from aggressive cholesterol-lowering therapy is no longer warranted. Intensive cholesterol reduction, initiated immediately, has the potential to significantly reduce both morbidity and mortality. Cholesterol-lowering therapy thus should become a routine part of clinical management to reduce risk of future coronary events and to prolong life in patients with CHD or other forms of atherosclerotic disease.  相似文献
