Trypanothione reductase (TR) is a flavoenzyme unique to trypanosomatid parasites and a target for lead discovery programs. Various inhibitor scaffolds have emerged in the past, exhibiting moderate affinity for the parasite enzyme. Herein we show that the combination of two structural motifs of known TR inhibitors — diaryl sulfides and mepacrine — enables the simultaneous addressing of two hydrophobic patches in the active site. The binding efficacy of these conjugates is enhanced over that of the respective parent inhibitors. They show Kic values for the parasite enzyme down to 0.9±0.1 μm and exhibit high selectivity for TR over human glutathione reductase (GR). Despite their considerable molecular mass and in some cases permanent positive charges, in vitro studies revealed IC50 values in the low micromolar to sub‐micromolar range against Trypanosoma brucei rhodesiense and Trypanosoma cruzi, as well as the malaria parasite Plasmodium falciparum, which lack trypanothione metabolism. The inhibitors exhibit strong fluorescence due to their aminoacridine moiety. This feature allows visualization of the drugs in the parasite where high accumulation was observed by fluorescence microscopy even after short exposure times. 相似文献

Geological Process Models (GPMs) have been used in the past to simulate the distinctive stratigraphies formed in carbonate sediments, and to explore the interaction of controls that produce heterogeneity. Previous GPMs have only indirectly included the supersaturation of calcium carbonate in seawater, a key physicochemical control on carbonate production in reef and lagoon environments, by modifying production rates based on the distance from open marine sources. We here use the residence time of water in the lagoon and reef areas as a proxy for the supersaturation state of carbonate in a new process model, Carbonate GPM. Residence times in the model are calculated using a particle-tracking algorithm. Carbonate production is also controlled by water depth and wave power dissipation. Once deposited, sediment can be eroded, transported and re-deposited via both advective and diffusive processes. We show that using residence time as a control on production might explain the formation of non-ordered, three-dimensional carbonate stratigraphies by lateral shifts in the locus of carbonate deposition on timescales comparable to so-called 5th-order sea-level oscillations. We also show that representing supersaturation as a function of distance from open marine sources, as in previous models, cannot correctly predict the supersaturation distribution over a lagoon due to the intricacies of the flow regime. 相似文献

The tropical diseases human African trypanosomiasis, Chagas disease, and the various forms of leishmaniasis are caused by parasites of the family of trypanosomatids. These protozoa possess a unique redox metabolism based on trypanothione and trypanothione reductase (TR), making TR a promising drug target. We report the optimization of properties and potency of cyclohexylpyrrolidine inhibitors of TR by structure‐based design. The best inhibitors were freely soluble and showed competitive inhibition constants (Ki) against Trypanosoma (T.) brucei TR and T. cruzi TR and in vitro activities (half‐maximal inhibitory concentration, IC50) against these parasites in the low micromolar range, with high selectivity against human glutathione reductase. X‐ray co‐crystal structures confirmed the binding of the ligands to the hydrophobic wall of the “mepacrine binding site” with the new, solubility‐providing vectors oriented toward the surface of the large active site. 相似文献

This paper is concerned with the design and performance evaluation of a direct-sequence spread-spectrum transmission and signal-detection technique for application to code-division multiple-access mobile radio networks. Unlike the conventional differential encoding/detection of data symbols, such a technique envisages differential encoding/detection of the spreading code chips to counteract the fast time-selective fading encountered in mobile radio transmissions. This paper is concerned with the estimation of the rate-of-change of the instantaneous frequency of an unmodulated carrier. Such an issue arises, for instance, in satellite communication systems based on a low-Earth-orbit constellation, wherein the speed of each satellite relative to earth is rapidly time-varying, and consequently, the received carrier is affected by nonnegligible Doppler shift and Doppler rate. First, we present simple yet efficient estimators of the Doppler rate derived from maximum-likelihood estimation theory. BACKGROUND: Partial harvesting of the left internal mammary artery (LIMA) is a widespread technique used during minimally invasive coronary operations performed through a left anterior small thoracotomy. The influence of persisting LIMA branches was investigated to evaluate their effect on the blood flow of the left anterior descending artery. METHODS: Thirty patients, 15 with totally (group A) and 15 with partially (group B) harvested LIMAs, were evaluated. All the patients underwent postoperative angiography, during which a flow map of the LIMA was performed. The average peak velocity and the diastolic-to-systolic peak velocity ratio were recorded. The LIMA graft flow pattern was recorded in the proximal and distal thirds of the artery. Intramammary adenosine (12 to 14 microg) was injected and the average peak velocities before and after injection were calculated. RESULTS: The average peak velocity was similar in both groups in the proximal and distal thirds of the LIMA (25 +/- 7 and 26 +/- 5 cm/sec, respectively, in group A versus 27 +/- 5 and 25 +/- 5 cm/sec, respectively in group B; p = NS). The diastolic-to-systolic peak velocity ratio was similar proximally (0.78 +/- 0.3 in group A versus 0.69 +/- 0.3 cm/s in group B; p = NS), but not distally (1.72 +/- 0.1 in group A versus 0.97 +/- 0.3 in group B; p < 0.0005). The LIMA graft flow reserve was similar both proximally and distally (2.6 +/- 0.6 and 2.5 +/- 0.3 cm/s, respectively, in group A versus 2.6 +/- 0.5 and 2.6 +/- 0.3 cm/s, respectively, in group B; p = NS). CONCLUSIONS: The persistence of LIMA branches does not influence the blood flow of the left anterior descending artery after acute adenosine-induced myocardial hyperemia. If a left anterior small thoracotomy is used in left anterior descending artery direct revascularization, complete LIMA harvesting is not mandatory and depends on the personal preference of the surgeon. 相似文献
