The effects of total ionising radiation dose upon commercial off-the-shelf semiconductors fitted to satellites operating in low Earth orbit (LEO) conditions was evaluated. The evaluation was performed for the Korea Institute of Technology SATellite-1, (KITSAT-1) which was equipped with commercial solid state components. Two approximate calculation models for space radiation shielding were developed. Verification was performed by comparing the results with detailed three-dimensional calculations using the Monte-Carlo method and measured data from KITSAT-1. It was confirmed that the developed approximate models were reliable for satellite shielding calculations. It was also found that commercial semiconductor devices, which were not radiation hardened, could be damaged within their lifetime due to the total ionising dose they are subject to in the LEO environment. The biodegradability of water samples is usually estimated with bioassays under oxic conditions. In order to overcome some of the drawbacks linked to the incubation of the samples in aerobic batches, a new protocol is proposed and tested, which is based on an organic carbon (OC) balance after a 45 days incubation under anoxic conditions with excess nitrate. The biodegradable fractions of organic matter obtained with the anoxic protocol are slightly lower than those obtained under oxic conditions. Several possible reasons for a systematic underestimation of the biodegradable organic matter under anoxic conditions are evaluated and discussed: a reduced microbial metabolic potential, significantly reduced degradation rates for the slowly biodegradable organic matter, an additional production of refractory organic compounds during the incubation, or the inhibition of the recycling of the organic matter stored in bacterial biomass. Nevertheless, the 7% difference observed on the biodegradable total OC estimations keeps low enough so that the anoxic protocol can be proposed as a convenient alternative to the oxic one. 相似文献

Ambient urban particulate matter (PM) contains various transition metals. When the PM is inhaled into the lung, not all but some part of metals from the particles might be mobilized to participate in a reaction that can damage various biomolecules, such as DNA and proteins. The dust particle size as well as organic acids may influence the metal mobilization. Thus, the mobilization of the metal from two standard reference materials (SRM; NIST, USA) and urban PM (PM2.5 and PM10) collected in the Seoul area was measured in the presence of artificial or biological chelator with or without reductant. The degree of the mobilization was higher with the artificial or biological chelator than the control with saline. In some cases, a reductant increased the mobilization as much as about 5 times the control without the reductant. Especially, the mobilization of Fe was greatly influenced by the presence of reductants. In general, the degree of the mobilization of the transition metal was higher with PM2.5 than with PM10. Therefore, it is expected that, considering the previously known toxicities of the transition metals, PM2.5 is more damaging to various biomolecules than PM10. The results also suggest that not the total amount but the mobilizable fraction of the metal in the ambient PM should be considered with regard to the toxicity of the urban particulate matter. 相似文献

Growth and metabolism of five probiotic strains with well-documented health effects were studied in ultra-high temperature (UHT) treated milk, supplemented with 0.5% (w/v) tryptone or 0.75% (w/v) fructose. The probiotic strains were Lactobacillus acidophilus La5, Lb. acidophilus 1748, Lactobacillus rhamnosus GG, Lactobacillus reuteri SD 2112 and Bifidobacterium animalis BB12. Fermentation was followed for 72 h at 37 degrees C and the samples were analysed for pH, log cfu ml(-1), volatile compounds, organic acids and carbon dioxide. The strains reduced pH from 6.7 to between 3.9 and 4.4 after 24 h of incubation. All strains attained viable cell counts above 8.7-9.18 log cfu ml(-1) after 6-16 h of incubation. The two Lb. acidophilus strains showed a stable level of viable cells during 12-72 h of incubation but the three other strains showed a reduction of 0.4-1.1 log cfu ml(-1) from 24 to 72 h of incubation. However, all strains showed cell levels between 7.8 and 8.7 log cfu ml(-1) after 72 h of incubation. After 48 h of incubation, the amount of lactic acid produced varied according to strain from 6949 to 14,000 mg kg(-1) and acetic acid produced varied from 0 to 6901 mg kg(-1). Three of the strains metabolised citrate but only low amounts of diacetyl and acetoin were detected within strains, 0.2-0.8 and 6.5-10 mg kg(-1), respectively. Carbon dioxide produced varied from 221 to 3942 mg kg(-1) and was connected to the citrate-fermenting ability of the strain used and their carbohydrate fermentation pathway. Three of the strains produced detectable levels of acetaldehyde and the concentration varied from 9.4 to 12.6 mg kg(-1) after 24 h of incubation. All five probiotic strains showed very different profiles of metabolites during fermentation; however, the two Lb. acidophilus strains were the most alike. The objective of this study was to examine the prevalence of Salmonella enterica in cull sows at various stages from the farm to the abattoir. Cull sows (n=181) were sampled over 10 weeks. Fecal samples (10 g each) were collected on the farm ca. 24 h before loading and at the live-hog market ca. 3 h before loading. Samples (ileocecal lymph nodes, cecal contents, feces from the transverse colon, ventral thoracic lymph nodes, subiliac lymph nodes, sponge swabs of the left and right carcass sections, and chopped meat) were collected at the abattoir. The percentages of positive fecal samples on the farm and at the live-hog market were 3% (5 of 181 samples) and 2% (3 of 181 samples), respectively. After transport from the live-hog market (10 h) and holding at the abattoir (6 h), 41% (74 of 180) of cull sows yielded S. enterica in one or more sampled tissues. The isolation rate for total cecal contents (33%; 60 of 180 samples) was significantly (P<0.05) higher than those for ileocecal lymph nodes (7%; 12 of 181 samples), feces (11%; 20 of 181 samples), and ventral thoracic and subiliac lymph nodes (2%; 4 of 181 samples). Before a 2% lactic acid carcass wash (lasting 8 to 9 s), 14% (25 of 180) of carcasses were positive, compared with 7% (12 of 179) after the wash (P<0.05). Two S. enterica serotypes, Derby and Infantis, were found on the farm and at the live-hog market. At the abattoir, 12 serotypes that had not previously been found on the farm or at the live-hog market were recovered. The results of this study demonstrate that transport and holding practices may contribute to an increase in S. enterica infection prior to slaughter to levels much higher than those found on the farm. 相似文献

The influence of inoculum populations and environmental factors on attachment of Salmonella Montevideo to the surface of tomatoes and tomatillos was evaluated. To study the effect of inoculum size, red, ripe tomatoes were spot-inoculated with bacterial suspensions (10(5) and 10(8) CFU/fruit) and stored at 22 degrees C under 100% relative humidity. The effects of temperature (12, 22, and 30 degrees C) and relative humidity (75, 85, and 97%) on attachment of the pathogen (10(7) CFU/fruit) to tomatoes (red and green) and ripe tomatillos were also evaluated. Inoculated fruits were stored for 90 min at all combinations of temperature and relative humidity, and after rinsing with water, the number of cells attached to the surface was determined. Salmonella Montevideo attached to the surface of tomatoes within 90 min. A direct correlation between the number of attached cells and the population in the inoculum was observed. The percentage of cells that attached immediately after inoculation was approximately 0.3% for the three test products. After storage for 90 min at various temperature and relative humidity conditions, the number of adhering cells ranged from 4.0 to 5.4 log CFU/fruit (1.2% of inoculum). Both the type of product and the temperature/relative humidity combination had a significant (P < 0.05) effect on attachment of Salmonella Montevideo to the surfaces of tomatoes and tomatillos. Scanning electron micrographs of the cuticles of inoculated washed tomatoes and tomatillos revealed typical skin cell patterns, and only a few randomly dispersed Salmonella Montevideo were observed. Deposition of Salmonella Montevideo on the surface of tomatoes and tomatillos could result in attachment and subsequent colonization under suitable conditions. 相似文献

Previous research showed that toxic and total alkaloid pools in tall larkspur (Delphinium barbeyi) increased during early growth, then declined precipitously during the late flower and pod stage of growth. The objective of this study was to measure the concentration and pools of toxic and total alkaloids in tall larkspur plant parts, including roots, and to evaluate the changes in these pools over the growing season as an estimate of diterpenoid alkaloid kinetics in tall larkspur. Twenty entire plants were harvested at each phenological stage: beginning of growth in the spring, early flower, early pod, late pod, and senescence. The plants were separated into their respective parts, freeze-dried, extracted, and analyzed for toxic and total alkaloid concentration, and alkaloid pools were calculated. Concentration of toxic and total alkaloids in leaves and stems declined as the plants matured, while concentration in flowers and pods increased (P < 0.004). Concentration of alkaloids in the root declined in the early growth, then increased at the end of the season (P = 0.002). Alkaloid pools in the root decreased during early growth, with a corresponding increase of pools in foliar parts. In the late flower and pod stage, alkaloid pools in the leaves and stems declined rapidly, while the pool in the crown and roots tended to increase. 相似文献

We have investigated a method for solving the inverse problem of determining the optical properties of a two-layer turbid model. The method is based on deducing the optical properties (OPs) of the top layer from the absolute spatially resolved reflectance that results from photon migration within only the top layer by use of a multivariate calibration model. Then the OPs of the bottom layer are deduced from relative frequency-domain (FD) reflectance measurements by use of the two-layer FD diffusion model. The method was validated with Monte Carlo FD reflectance profiles and experimental measurements of two-layer phantoms. The results showed that the method is useful for two-layer models with interface depths of >5 mm; the OPs were estimated, within a relatively short time (<1 min), with a mean error of <10% for the Monte Carlo reflectance profiles and with errors of <25% for the phantom measurements. 相似文献

Theoretical analysis was applied to analyze the refractive-index distribution (RID) and bandwidth (BW) of gradient-index polymer optical fibers (GI POFs) prepared by a centrifugal field process. The RID of the prepared GI POF could be represented by the equation of n(r) = n1[1 - 2delta(r/alpha)g](1/2). The studied material systems were poly(hexafluoroisopropyl 2-fluoroacrylate) (PHFIP 2-FA)/dibutyl phthalate (DBP) and poly(methyl methacrylate) (PMMA)/benzyl benzoate (BEN). The RID and the BW were significantly affected by an essential parameter k, which was related to thematerial properties (density difference and molecular weight) and processing properties (rotating speed, temperature, and radius). As k increased, the characteristic constant of RID, g, decreased to a minimum and then increased sharply, owing to the separation of the polymer and the dopant. On the other hand, the relative refractive-index difference of RID, delta, increased to a steady value after k increased to a certain value. The variation of RID with k resulted in a local minimum of intermodal dispersion, and thus a maximum bandwidth was obtained. The maximum BW of the PHFIP 2-FA/DBP and PMMA/BEN systems at 1550 nm (100-m fiber length and 2-nm spectral width) for the case of k not equal to 0 were 6.7 and 3.2 Gb/s, respectively. The wavelength of light source affects the BW significantly only at k around zero because of the importance of the intramodal dispersion in this case. 相似文献
