To extract cell surface-associated proteins from living fungal cells, reducing agents such as beta-mercaptoethanol and dithiothreitol are often used. We show here that both compounds are moderately lipophilic and may perturb the plasma membrane, thus causing the release of cytosolic proteins, especially at high extraction temperatures. To avoid artifacts, we recommend using (a) a low concentration of the reducing agent for only a short period of time, and (b) an extraction temperature of 4 degrees C to protect the integrity of the plasma membrane. Similarly, biotinylation of cell surface proteins should be carried out at low temperatures in the absence of dimethylsulphoxide.  相似文献

Polyphenolics (PP) from bearberry (Arctostaphylos uva-ursi L. Sprengel) leaves were extracted with 70% (v/v) acetone. The lyophilised crude PP extract was then separated on a Sephadex LH-20 column using first 95% (v/v) ethanol as a mobile phase to elute fraction I low in tannins, and then 50% (v/v) acetone to elute fraction II rich in hydrolysable and condensed tannins. Bovine serum albumin (BSA) was effectively precipitated by PP in the bearberry-leaf crude extract and fraction II at pH values between 3.8 and 4.5. A statistically significant (P = 0.0001) linear relationship exists between the amount of PP-protein complex formed and the quantity of PP added to the reaction mixture. The slope values of these lines indicate that fraction II from the crude extract was a more effective precipitant of proteins than other examined preparations. Based on the quantity of gelatine, fetuin, and BSA required to inhibit 50% of dye-labelled PP–BSA complex precipitation, gelatine was 3–6 times more effective as a precipitation inhibitor than both unlabelled BSA and fetuin.  相似文献

Several lines of evidence suggest that the image statistics of the environment shape visual abilities. To date, the image statistics of natural scenes and faces have been well characterized using Fourier analysis. We employed Fourier analysis to characterize images of signs in American Sign Language (ASL). These images are highly relevant to signers who rely on ASL for communication, and thus the image statistics of ASL might influence signers' visual abilities. Fourier analysis was conducted on 105 static images of signs, and these images were compared with analyses of 100 natural scene images and 100 face images. We obtained two metrics from our Fourier analysis: mean amplitude and entropy of the amplitude across the image set (which is a measure from information theory) as a function of spatial frequency and orientation. The results of our analyses revealed interesting differences in image statistics across the three different image sets, setting up the possibility that ASL experience may alter visual perception in predictable ways. In addition, for all image sets, the mean amplitude results were markedly different from the entropy results, which raises the interesting question of which aspect of an image set (mean amplitude or entropy of the amplitude) is better able to account for known visual abilities.  相似文献

With the use of the copper(I)-catalyzed (3 + 2) azide-alkyne cycloaddition, an element of "click chemistry," stationary phases carrying long alkyl chains or soybean trypsin inhibitor have been prepared for use in HPLC separations in the reversed-phase and affinity modes, respectively. The ligands were attached via a triazole ring to size monodisperse porous beads containing either alkyne or azide pendant functionalities. Alkyne-containing beads prepared by direct copolymerization of propargyl acrylate with ethylene dimethacrylate were allowed to react with azidooctadecane to give a reversed-phase sorbent. Azide-functionalized beads were prepared by chemical modification of glycidyl methacrylate particles. Subsequent reaction with a terminal aliphatic alkyne produced a reversed-phase sorbent similar to that obtained from the alkyne beads. Soybean trypsin inhibitor was functionalized with N-(4-pentynoyloxy)succinimide to carry alkyne groups and then allowed to react with the azide-containing beads to produce an affinity sorbent for trypsin. The performance of these stationary phases was demonstrated with the HPLC separations of a variety of peptides and proteins.  相似文献

Gingival overgrowth is a common health problem caused by genetic and environmental risk factors. Animal models for quantitative histological studies are needed to uncover genetic predisposition and dose-response data that might put individuals at increased risk for gingival disease. Gingival height, thickness, inflammation, and the degree of encroachment of gingiva over the tooth, are clinical measures of overgrowth; most of these parameters can be measured histologically, but in order to quantify gingival coverage of the tooth, the image of the crown must be present. Tooth and bone typically require decalcification for histology; thus, the tooth crown, a critical landmark, is lost. We describe a method for imaging the crown histologically, using impression materials applied to dissected mouse mandibles. Four dental alginates, three polyvinyl siloxanes, and one polyether and gelatin were used. The impression-material/mandibular tissue blocks were processed routinely. Polyvinyl siloxanes were incompatible with embedding resin; alginates, polyether and gelatin could be fixed, decalcified, embedded, and sectioned. Alginates and gelatin could be stained. Success in imaging the tooth crown varied with the preparation, but the alginates, polyether, and gelatin permitted a useful degree of measurement of exposed crown and enamel thickness, along with other morphometric parameters such as thickness of the dentin, lateral mandibular ramus, rete pegs, height of the gingiva, and volume density of vessels and inflammatory cells in the lamina propria. In conclusion, this new application for impression materials allows gingival coverage of tooth crown, as well as numerous other parameters to be measured for comparison with clinical data.  相似文献

Ferrite‐based, hard‐soft magnetic nanocomposites with the composition (100%?x)SrFe12O19xCoFe2O4, where x = 5, 10, and 15 wt%, were prepared by mixing the constituent powders, followed by spark plasma sintering. In order to control the particle size of the constituent materials, the SrFe12O19 and CoFe2O4 powders were synthesized using the hydrothermal method, mixed and then consolidated with spark plasma sintering. The conditions during the spark plasma sintering process (sintering temperature, time, and applied pressure) were varied in order to prepare composites with a high density and exchange‐coupled hard and soft magnetic phases, leading to an increase in the maximum energy product, when compared with pure SrFe12O19. The microstructural analysis revealed that the relative density of the sintered composite exceeded 90% of the theoretical value and that the CoFe2O4 was uniformly distributed in the SrFe12O19 matrix. Magnetic measurements of the sintered composites showed a single‐phase magnetic behavior. When compared with the single‐phase SrFe12O19 used in this study, the SPS composites exhibited a 22% increase in the maximum energy product (26.1 kJ/m3).  相似文献

Heat treatment and drying processes of wood are always accompanied by material changes. These changes are partly caused by oxidative reactions. This study aims to compare high temperature drying in inert gas atmosphere with conventional kiln drying of spruce wood. The mechanical properties as well as colour changes were measured after drying. In general, higher compression strength was observed for samples dried at high temperatures in nitrogen atmosphere. However, tensile and bending strength proved no significant difference. Corresponding to the strength values increased stiffness values were also observed for the compression samples, whereas for tension and bending samples no significant effect was visible. Up to a temperature of approx. 125 °C discolouration is prevented by drying samples in nitrogen atmosphere.  相似文献

Soil moisture mapping and AMSR-E validation using the PSR in SMEX02   总被引:5,自引:0,他引:5  
Field experiments (SMEX02) were conducted to evaluate the effects of dense agricultural crop conditions on soil moisture retrieval using passive microwave remote sensing. Aircraft observations were collected using a new version of the Polarimetric Scanning Radiometer (PSR) that provided four C band and four X band frequencies. Observations were also available from the Aqua satellite Advanced Microwave Scanning Radiometer (AMSR-E) at these same frequencies. SMEX02 was conducted over a three-week period during the summer near Ames, Iowa. Corn and soybeans dominate the region. During the study period the corn was approaching its peak water content state and the soybeans were at the mid point of the growth cycle. Aircraft observations are compared to ground observations. Subsequently models are developed to describe the effects of corn and soybeans on soil moisture retrieval. Multiple altitude aircraft brightness temperatures were compared to AMSR-E observations to understand brightness temperature scaling and provide validation. The X-band observations from the two sensors were in reasonable agreement. The AMSR-E C-band observations were contaminated with anthropogenic RFI, which made comparison to the PSR invalid. Aircraft data along with ancillary data were used in a retrieval algorithm to map soil moisture. The PSR estimated soil moisture retrievals on a field-by-field comparison had a standard error of estimate (SEE) of 5.5%. The error reduced when high altitude soil moisture estimates were aggregated to 25 km resolution (same as AMSR-E EASE grid product resolution) (SEE ∼ 2.85%). These soil moisture products provide a validation of the AMSR retrievals. PSR/CX soil moisture images show spatial and temporal patterns consistent with meteorological and soil conditions. The dynamic range of the PSR/CX observations indicates that reasonable soil moisture estimates can be obtained from AMSR, even in areas of high vegetation biomass content (∼ 4-8 kg/m2).  相似文献

The stability of water-based pigment dispersions is a key factor in determining their utility in ink-jet applications, and the appropriate choice of dispersant plays a special role. Among the pigment formulations tested to date, literature data on magenta pigments are very limited. Thus, the goal of this work was to study the influence of the type and loading of dispersants on the quality and stability of quinacridone magenta pigment dispersions. Three different commercially available dispersants were tested: (1) a cationic styrene-maleic anhydride copolymer, (2) an anionic polyacrylate, and (3) a nonionic alkyl ethoxylate. Pigment and dispersing agents were analysed using the Fourier Transform–infrared method. The stability of pigment dispersions was determined by comparative studies of changes in the pigment particle size, viscosity, pH, filtration time, optical density and gloss during the accelerated ageing test. Moreover, Turbiscan AGS and LUMiFuge analysers were used for evaluations of the instability of the dispersions. The properties of each dispersing agent affected the performances of the different dispersants. The highest stability for the pigment formulations was provided using the nonionic dispersant (small particle size, < 60 nm; the lowest viscosity < 60 mPa·s at the optimal dispersant content; favourable results of the filtration test; the highest pigment dispersability, proven by optical density and gloss). The Turbiscan AGS and LUMiFuge results indicated that the stability of all tested quinacridone magenta pigment dispersions was excellent (Turbiscan Stability Index values below three, and no significant differences in transmission profiles, respectively). However, it was found that the measurement results were influenced by the viscosity of the formulations.  相似文献
