Changes in proteasome activity during postmortem aging of bovine muscle   总被引:1,自引:0,他引:1  
The effects of adding commercial-grade and eggshell calcium lactate on the microbiological and physicochemical properties of Nhams (Thai-style fermented pork sausage) were studied. The Nham calcium levels were 150, 300 and 450 mg/100 g. Compared to controls (no added calcium), calcium fortification did not affect the number of lactic acid bacteria or the colour value. The shear force of Nhams fortified with eggshell calcium lactate decreased (P<0.05) from 32.2 N in the controls to 19.5-22.8 N in Nhams fortified with eggshell calcium lactate. However, Nhams fortified with commercial calcium lactate had the same shear force as the controls. Sensory scores of sour taste, flavour and overall acceptance were not different between the control and calcium-fortified Nhams at a calcium level of 150 mg/100 g.  相似文献

Immobilised DNA-oligo layers are scientifically and technologically appealing for a wide range of sensor applications such as DNA chips. Using microcantilever-based sensors with integrated readout, we demonstrate in situ quantitative studies of surface-stress formation during self-assembly of a 25-mer thiol-modified DNA-oligo layer. The self-assembly induces a surface-stress change, which closely follows Langmuir adsorption model. The adsorption results in compressive surface-stress formation, which might be due to intermolecular repulsive forces in the oligo layer. The rate constant of the adsorption depends on the concentration of the oligo solution. Based on the calculated rate constants a surface free energy of the thiol-modified DNA-oligo adsorption on gold is found to be -32.4 kJ mol(-1). The adsorption experiments also indicate that first a single layer of DNA-oligos is assembled on the gold surface after which a significant unspecific adsorption takes place on top of the first DNA-oligo layer. The cantilever-based sensor principle has a wide range of applications in real-time local monitoring of chemical and biological interactions as well as in the detection of specific DNA sequences, proteins and particles.  相似文献

In this study, the authors examined the correlates of psychological aggression victimization and perpetration among a community sample of 145 heterosexual couples. For both women and men, psychological aggression victimization was associated with greater psychological distress, anxiety, and physical health symptoms beyond the effects of physical aggression. Psychological aggression victimization was also uniquely associated with higher levels of depression for women. Trait anger and poor relationship adjustment were the strongest correlates of psychological aggression perpetration across genders. Childhood father-to-child and father-to-mother aggressions were associated with psychological aggression perpetration for men only, suggesting possible distinct etiologies across genders. Polypropylene fibers produced in a compact-spinning process were studied by differential scanning calorimetry (DSC). With unrestrained fibers, the onset of melting increases with decreasing draw ratio, increasing Mw/Mn, decreasing extrusion temperature, increasing annealing ratio, and increasing draw-down ratio. These trends are discussed in terms of restraints and reorganization. The onset of melting is shifted to lower temperatures as the heating rate increases for all combinations of material and processing parameters, indicating suppressed reorganization. At low draw ratios, the height and width of the endotherm are affected by the spinline stress, and a secondary peak or shoulder is observed on the high temperature side of the main peak. The magnitude of the secondary peak increases with decreasing Mw/Mn, increasing draw ratio, decreasing draw-down ratio, and decreasing heating rate, but its position mainly depends on the heating rate. This indicates that the secondary peak may be due to the melting of structures that have been reorganized during the heating scan. As the draw ratio increases, the melting regime broadens, especially towards lower temperatures, and several maxima emerge on the DSC curve. The isolation of anti-hepatitis C virus (HCV) in hemodialyzed (HD) patients has been repeatedly advocated to prevent nosocomial HCV transmission. We evaluated the incidence of seroconversion for HCV in Belgian HD patients, and demonstrate the complete prevention of HCV transmission by adherence to the universal precautions advocated by the Centers for Disease Control (Atlanta, GA, USA). All (N = 963) HD patients from 15 units, none of which isolates anti-HCV positive patients, were tested by a second or third generation enzyme-linked immunosorbent assay (with confirmation by a second- or third-generation recombinant immunoblot assay or the polymerase chain reaction) every 18 months from May 1991 to November 1995. Follow-up was available in 488 patients (drop-outs resulting from death or transplantation mainly). The yearly incidence of seroconversion for HCV over the initial 18 months was 1.41%, with evidence suggestive of nosocomial HCV transmission. Universal precautions were therefore reinforced. The incidence of seroconversion subsequently fell to 0.56% and 0%, respectively (P = 0.014), despite the facts that the average transfusion load and the proportion of patients with dialyzer reuse or with monitors disinfected after each session did not change significantly. We conclude that the strict enforcement of universal precautions fully prevents HCV transmission to HD patients. The isolation of anti-HCV positive patients is not warranted.  相似文献

The surface tension of the CMSX-4®reg; superalloy has been determined by the drop-weight and oscillating-drop methods which are well adapted to reactive materials. The recommended values are 1.59 J·m–2 for the surface tension at the liquidus temperature and –0.14× 10–3 J·m–2· K–1 for the temperature coefficient. A conclusion of the present work is that the interpretation of surface tension measurements performed on a complex alloy generally requires additional work to be performed on simpler associated binary or ternary systems, as well as some support from solidification experiments.  相似文献

The cytoskeleton is an intracellular highway system, teaming with signalling ions that zip from site to site along filaments. These tiny particles alternately embrace and slip free of protein receptors with wide-ranging affinities, as they propagate in a blur of motion along cytoskeletal corridors at transport rates far exceeding ordinary diffusive motion. Recent experimental breakthroughs have enabled optical tracking of these single ion-binding events in the physiological and diseased states. However, traditional continuum modelling methods have proven ineffective for modelling migration of biometals such as copper and zinc, whose cytosolic concentrations are putatively vanishingly small, or very tightly controlled. Rather, the key modelling problem that must be solved for biometals is determination of the optimal placement of biosensors that bind and detect the metal ions within the heterogeneous environment of the cell. We discuss herein how percolation concepts, in combination with atomistic simulation and sensor delivery models, have been used to gain insights on this problem, and a roadmap for future breakthroughs.  相似文献

In this letter, we show a direct relation between spectral embedding methods and kernel principal components analysis and how both are special cases of a more general learning problem: learning the principal eigenfunctions of an operator defined from a kernel and the unknown data-generating density. Whereas spectral embedding methods provided only coordinates for the training points, the analysis justifies a simple extension to out-of-sample examples (the Nystr?m formula) for multidimensional scaling (MDS), spectral clustering, Laplacian eigenmaps, locally linear embedding (LLE), and Isomap. The analysis provides, for all such spectral embedding methods, the definition of a loss function, whose empirical average is minimized by the traditional algorithms. The asymptotic expected value of that loss defines a generalization performance and clarifies what these algorithms are trying to learn. Experiments with LLE, Isomap, spectral clustering, and MDS show that this out-of-sample embedding formula generalizes well, with a level of error comparable to the effect of small perturbations of the training set on the embedding.  相似文献
