Contamination of ready-to-eat meat products such as beef franks with Listeria monocytogenes has become a major concern for the meat processing industry and an important food safety issue. The objective of this study was to determine the effectiveness of combinations of antimicrobials as aqueous dipping solutions to control L. monocytogenes on vacuum-packaged beef franks stored at 4 degrees C for 3 weeks. Commercial beef franks were dipped for 5 min in three antimicrobial solutions: pediocin (6,000 AU), 3% sodium diacetate and 6% sodium lactate combined, and a combination of the three antimicrobials. Samples were then inoculated with 10(7) CFU/g of either four L. monocytogenes strains individually or a cocktail of the four strains, vacuum packaged, and stored at 4 degrees C for 3 weeks. Sampling was carried out at day 0 and after 2 and 3 weeks of storage. Individual strains, as well as the cocktail, exhibited different responses to the antimicrobial treatments. After 2 and 3 weeks of storage at 4 degrees C, pediocin-treated beef franks showed a less than 1-log reduction for all bacterial strains. Samples treated with the sodium diacetate-sodium lactate combination showed about a 1-log reduction after 2 weeks of storage for all strains and between a 1- and 2-log reduction after 3 weeks of storage, depending on the bacterial strain. When the three antimicrobials were combined, reductions ranged between 1 and 1.5 log units and 1.5 to 2.5 log units after 2 and 3 weeks of storage, respectively, at 4 degrees C. These results indicate that the use of combined antimicrobial solutions for dipping treatments is more effective at inhibiting L. monocytogenes than treatments using antimicrobials such as pediocin separately.  相似文献

In the present work the existence of mass transfer limitations in slurry, photocatalytic reactors is studied. Experimental validation is made in a flat plate reactor that is part of a recycling system. The reactor is described with a mathematical model previously developed [Ballari et al., 2008a. Chemical Engineering Journal 136, 50], considering a transient, two-dimensional mass balance (TDM). The complete reactor model was developed to show the existence of these effects, which result from the occurrence of concentration gradients in reaction space. They develop when these reactors are operated under some operating conditions whose effects should be always analyzed before assuming the validity of existence of perfect mixing in reaction space. Dichloroacetic acid (DCA) was the adopted model compound. To solve TDM, a kinetic expression for DCA acid was determined before under well mixed conditions [Ballari et al., 2009. Industrial and Engineering Chemistry Research 48(4), 1847]. The studied variables are flow rate, catalyst loading, and irradiation rates. The experimental data agree quite well when they are interpreted in terms of the two-dimensional model (TDM) regardless of the operating mode. The perfect mixing model (PMM), normally employed to describe this and other types of slurry photoreactors, does not have the same level of universal application; i.e. it is restricted to perfect mixing, but in many cases far simpler to use. However, it can be concluded that when the photocatalytic reaction is not fast, employing catalyst loadings below 1 g L–1, irradiation rates at the reactor wall below 1×10?6 Einstein cm?2 s?1 and good mixing operation (Re>1700) it will be always safe to assume that mass transport limitations in the bulk of the fluid are nonexistent. In a typical batch reactor the above flow conditions are equivalent to very intense mixing. If the catalyst concentration is increased, the mixing conditions should be improved in the same proportion. Within limits, higher solid loadings can be compensated with lower irradiation rates [Ballari et al., 2008a. Chemical Engineering Journal 136, 50]. In addition, with the validated model, additional simulations are shown, operating the reactor under different virtual reactor thicknesses to widen amplitude of the reached conclusions. These findings will be useful in kinetic studies to prevent incursion in certain ranges of experimental conditions that could lead to erroneous interpretation of the obtained kinetic data.  相似文献

It is well known that the content of water does affect materials behaviour and may cause important damages to buildings, including structural deterioration. Thermography is a non-intrusive imaging technique that allows the inspection of different materials based on their surface temperatures. Within an appropriate thermal analysis, moisture and other structural damages can be detected. The present research work deals with multitemporal thermal analysis to detect moisture on buildings, as well as other alterations. Two different approaches are tested to determine the effectiveness on the moisture surveys, one simple based on image differences, and another more sophisticated based on principal components analysis (PCA). The results are tested at the front door of the Arenberg Castle (Leuven, Belgium). Some moisture maps are derived and used to determine the reliability of either the computation of image differences or the PCA to carry out semi-automatically moisture surveys on architectural façades. The latter method is proven to be superior when using thermal imagery acquired at different times.  相似文献

Native kainate receptors expressed by cultured hippocampal cells were studied in the whole-cell configuration of the patch-clamp technique by using a fast perfusion system. About 80% of the neurons expressed kainate receptors independently of the time in culture (0-4 days), which coincided with the number of cells immunoreactive for a monoclonal antibody against the GluR5/6/7 subunits. Three types of cells were considered: neurons in which the rapid application of kainate induced a rapidly desensitizing current, cells in which kainate induced a more slowly rising, non-desensitizing, response and those in which a mixture of both responses was apparent. Steady responses induced by 300 microM kainate were inhibited by 6-cyano-7-nitroquinoxaline-2,3-dione (CNQX) in a dose-dependent manner (IC50 = 0.92 microM). CNQX was less potent in blocking transient kainate-induced responses (IC50 = 6.1 microM). Responses to kainate, whether steady or transient, were also inhibited by NS102, showing poor selectivity for the transient response (IC50 = 4.1 and 2.2 microM respectively). The new alpha-amino-3-hydroxy-5-methyl-4-isoxazole (AMPA) receptor antagonist NS394 was very potent in inhibiting steady kainate-induced currents (IC50 = 0.45 microM), but was even more effective in preventing peak responses (IC50 = 0.13 microM). In contrast, cyclothiazide did not affect transient kainate-induced responses but did potentiate current induced by activation of AMPA receptors by AMPA or kainate. These results demonstrate the lack of complete selectivity amongst some available competitive antagonists for AMPA and kainate receptors, and indicate that kainate receptors expressed by hippocampal cells lack the cyclothiazide modulatory site present at AMPA receptors. In addition, the present data support the idea that low-affinity kainate binding sites in the brain correspond to receptor channels selectively activated by kainate.  相似文献

The scope of this work is to evaluate some properties of the oils and derived fatty acid ethyl esters (FAEE) from two different Jatropha Curcas species planted in Cuba. The properties that were determined include the acid value, peroxide value, p-anisidine value and fatty acid ethyl esters composition. In order to study the influence of the genus species and geographic conditions on the fuel properties, the oils from Jatropha Curcas planted in two regions of Cuba and their derived FAEE were analyzed and compared. The two plantations were in San José (SJ) and Guantanamo (Gt) representing respectively the western and eastern part of the island.The analyses indicated that the FAEE obtained from Guantanamo has a higher acid value and peroxide value compared with the FAEE from San José. The p-anisidine values did not show a clear trend and the results of gas chromatography-mass spectrometry indicated a similar FAEE composition. The results obtained by gas chromatography are in good agreements with previous reports.  相似文献

The photochemical behaviour of three relevant metabolites of the analgesic and antipyretic drug dipyrone, 4-methylaminoantipyrine (4-MAA), 4-formylaminoantipyrine (4-FAA) and 4-acetylaminoantipyrine (4-AAA), was evaluated under simulated solar irradiation (Suntest system). For 4-MAA, different aqueous solutions (synthetic seawater, freshwater and Milli-Q water) as well as different operational conditions were compared. According to the experimental results, 4-MAA resulted as being an easily degraded molecule by direct photolysis, with half-life times (t1/2) ranging from 0.12 to 0.58 h, depending on the irradiation conditions. Faster degradation was observed in synthetic waters, suggesting that the photolysis was influenced by the salt composition of the waters. However, no effect on the degradation rate was observed by the presence of natural photosensitizers (dissolved organic matter, nitrate ions). 4-FAA and 4-AAA showed slower photodegradation kinetics, with t1/2 of 24 and 28 h, respectively. A study of photoproduct identification was carried out by gas chromatography-mass spectrometry (GC-MS) and liquid chromatography-time-of-flight mass spectrometry (LC-TOF-MS) (ESI positive mode), which allowed us to propose a tentative photodegradation pathway for 4-MAA and the identification of persistent by-products in all the cases. Finally, the application of an acute toxicity test (Daphnia magna) showed an increase in toxicity during the photolytic process, a consequence of the formation of toxic photoproducts.  相似文献
