Analysis of the stabilization of hen lysozyme by helix macrodipole and charged side chain interaction

In the N-terminal region of the alpha-helix of the c-type lysozymes, two Asx residues exist at the 18th and 27th positions. Hen lysozyme has Asp18/Asn27 (18D/27N), and we prepared three mutant lysozymes, Asn18/Asn27 (18N/27N), Asn18/Asp27 (18N/27D), and Asp18/Asp27 (18D/27D). The stability of the wild-type (18D/27N) lysozyme supported the existence of a hydrogen bond between the side chain of Asp18 and the amide group at the N1 position in the alpha-helix, while the stability of the 18N/27D lysozyme supported the presence of the capping box between the Ser24 (N-cap) and Asp27 residues. Although electrostatic repulsion was observed between Asp18 and Asp27 residues in 18D/27D lysozyme, the dissociation of each residue contributed to stabilizing the B-helix in 18D/27D lysozyme through hydrogen bonding and charge-helix macrodipole interaction. This is the first evidence that two neighboring negative charges at the N-terminus of the helix both increased the stability of the protein.

文中以精制盐加工工艺理论为指导，进行了精制盐代原盐，取消原盐焙烧制造煤矿许用铵梯炸药的尝试。精制盐替代原盐具有降低材料成本，概治环境污染，简化工艺过程，提高炸药组分初检合格率等优点。     

有源扩散方程的一种解和“双滤纸法”测氢的换算公式

文章从流速场中级联衰变下气载核素的扩散规律出发,提出了强吸附性壁圆管内氡子体扩散方程的完整定解问题,求出了等速分布廓线时的方程的精密解,弥补了C.W.Tan这部分工作的不足。引进了新的物理量m(μ),导出了等速分布条件下m(μ)与F(μ)的互换关系式。从测量氡子体的α计数出发,通过放射性平衡比m(μ)给出了“双滤膜法”测氡的换算公式。由J·W·Thomas的精密实验数据,取镭-A扩散系数D=0.07cm~2·s~(-1)时,理论与实验十分吻合。     

Activin A: an autocrine inhibitor of initiation of DNA synthesis in rat hepatocytes

The present study was conducted to examine the effect of activin A on growth of rat hepatocytes. EGF induced a 10-fold increase in DNA synthesis as assessed by [3H]thymidine incorporation in cultured hepatocytes. When activin A was added together with EGF, DNA synthesis induced by EGF was markedly inhibited. Inhibition was detected at a concentration of 10(-10) M, and 5 x 10(-9) M activin A almost completely blocked EGF-mediated DNA synthesis. Similarly, activin A completely blocked DNA synthesis induced by hepatocyte growth factor/scatter factor. Activin A was capable of inhibiting EGF-mediated DNA synthesis, even when added 36 h after the addition of EGF. With the same time interval, TGF-beta also blocked EGF-induced DNA synthesis. Although both activin A and TGF-beta inhibited growth of hepatocytes in a similar manner, either activin A or TGF-beta did not compete with each other in their binding when assessed by competitive binding using an iodinated ligand. When hepatocytes were incubated with EGF, release of bioactivity of activin A into culture medium was detected after 48 h or later. Activity of activin A was released from parenchymal cells but not from nonparenchymal cells. mRNA for beta A subunit of activin was detected only slightly in unstimulated hepatocytes, but markedly increased at 48 h after the addition of EGF. To determine whether endogenously produced activin A affects DNA synthesis, we examined the effect of follistatin, an activin-binding protein that blocks the action of activin A. An addition of follistatin significantly enhanced EGF-induced DNA synthesis. Finally, in partial hepatectomized rat, expression of mRNA for beta A subunit in liver was markedly increased 24 h after the partial hepatectomy. These results indicate that activin A inhibits initiation of DNA synthesis in hepatocytes by acting on its own receptor and that activin A acts as an autocrine inhibitor of DNA synthesis in rat hepatocytes.

Hydrogen transport in solution-treated and pre-strained austenitic stainless steels and its role in hydrogen-enhanced fatigue crack growth

Hydrogen solubility and diffusion in Type 304, 316L and 310S austenitic stainless steels exposed to high-pressure hydrogen gas has been investigated. The effects of absorbed hydrogen and strain-induced martensite on fatigue crack growth behaviour of the former two steels have also been measured. In the pressure range 10–84 MPa, the hydrogen permeation of the stainless steels could be successfully quantified using Sieverts' law modified by using hydrogen fugacity and Fick's law. For the austenitic stainless steels, hydrogen diffusivity was enhanced with an increase in strain-induced martensite. The introduction of dislocation and other lattice defects by pre-straining increased the hydrogen concentration of the austenite, without affecting diffusivity. It has been shown that the coupled effect of strain-induced martensite and exposure to hydrogen increased the growth rate of fatigue cracks.
