Methods of measuring susceptibility of anaerobic bacteria to trovafloxacin, including quality control parameters

Three methods approved by the National Committee for Clinical Laboratory Standards for testing the susceptibility of anaerobic bacteria were used to evaluate the fluoroquinolone, trovafloxacin. The methods gave essentially comparable results with 126 anaerobes and with three quality control strains. A collaborative study defined the quality control range for trovafloxacin MICs. Trovafloxacin had good in vitro activity against the more common anaerobes (MIC 90 < = or 2.0 micrograms/ml).     

Spatial modeling for groundwater arsenic levels in North Carolina

To examine environmental and geologic determinants of arsenic in groundwater, detailed geologic data were integrated with well water arsenic concentration data and well construction data for 471 private wells in Orange County, NC, via a geographic information system. For the statistical analysis, the geologic units were simplified into four generalized categories based on rock type and interpreted mode of deposition/emplacement. The geologic transitions from rocks of a primary pyroclastic origin to rocks of volcaniclastic sedimentary origin were designated as polylines. The data were fitted to a left-censored regression model to identify key determinants of arsenic levels in groundwater. A Bayesian spatial random effects model was then developed to capture any spatial patterns in groundwater arsenic residuals into model estimation. Statistical model results indicate (1) wells close to a transition zone or fault are more likely to contain detectible arsenic; (2) welded tuffs and hydrothermal quartz bodies are associated with relatively higher groundwater arsenic concentrations and even higher for those proximal to a pluton; and (3) wells of greater depth are more likely to contain elevated arsenic. This modeling effort informs policy intervention by creating three-dimensional maps of predicted arsenic levels in groundwater for any location and depth in the area.     

Analysis and quantification of the secretory products of the subcommissural organ by use of monoclonal antibodies

Bovine Reissner's fiber (RF) glycoproteins were used as antigen for the production of polyclonal and monoclonal antibodies (Mabs). We also produced Mabs against intracellular secretory glycoproteins of the bovine subcommissural organ (SCO). These Mabs were used for immunodetection of secretory proteins in situ (structural and ultrastructural immunocytochemistry), in blots, and in solutions. Three different antigen-mediated ELISA were designed to evaluate the affinity of the Mabs, to study the nature of the epitopes, and for competition test among Mabs. Two double antibody sandwich ELISA were designed to detect and quantify soluble secretory materials in different samples, to study coexistence of epitopes, and to elucidate whether epitopes for Mabs are repeated or not in the RF-glycoproteins. Twenty-three Mabs recognizing the bovine RF- and SCO-glycoproteins in solutions (ELISA) as well as in tissue sections, were obtained. Nineteen of these Mabs also recognized the pig SCO, 11 the rabbit SCO, 6 the dog SCO, and 5 the rat SCO. None of the Mabs recognized the SCO of non-mammalian species. The different types of ELISA demonstrated that: (1) the epitopes reside in the proteinaceous moiety of the secretion, (2) they coexist in the same molecular forms and, with few exceptions, they did not overlap, (3) they were not repeated in the secretory molecule(s). Three Mabs were used for immunoblotting of RF; one of them revealed the same band pattern as that shown by an anti-RF serum. It is concluded that all Mabs raised in our laboratory are directed against non-repeated sequences of RF-glycoproteins that have not been conserved in vertebrate phylogeny.     

How far can one compress digital fault records? Analysis of a matching pursuit-based algorithm

In this paper, we verify how far electric disturbance signals can be compressed without compromising the analysis of encoded fault records. A recently proposed compression algorithm, referred to as Damped Sinusoidal Matching Pursuit (DSMP) has the remarkable feature of obtaining both compact and physically interpretable representations. However, for fault analysis applications, one is primarily interested in how accurate can be the analysis performed on compressed signals, instead of evaluating mean-squared error figures. Unlike previous works in digital fault records compression, the performance of the DSMP compression method is evaluated using a protocol based on fault analysis procedures commonly performed by expert engineers. This protocol is applied for comparing the results obtained in the analysis of both uncompressed records and their compressed versions at different compression ratios. The results show that the DSMP is a reliable compression system since it achieves high compression ratios (6.4:1) without causing fault analysis misinterpretation.     

Thioredoxin fusion/HIV-1 protease coexpression system for production of soluble human IL6 in E. coli cytoplasm

In this paper, thioredoxin (TRX) fusion expression system has been modified to produce soluble human IL6 (hIL6) without TRX moiety in E. coli cytoplasm. A novel TRX gene fusion vector was developed that contained at the 3'-end of TRX gene a short DNA sequences encoding a linker peptide '-GSGSGVSQNYPIVQHHHHHH-', serving not only as a specific HIV-1 protease site but also providing six contiguous histidine (His) residues to foreign proteins. The cDNA for hIL6 was cloned into this vector resulting in plasmid pTRX@HISIL6. The cDNA for the HIV-1 protease has been cloned into another compatible plasmid pHMM2, resulting in plasmid pHMM2-PR. Both plasmids were transformed into E. coli strain GI724, and when induced for expression of both proteins, the correct processing of TRX@HISIL6 was obtained, producing hIL6 with His6-tag at the N terminus named HISIL6. A fraction of HISIL6 was found in soluble form and could be purified to homogeneity by Ni-NTA Superflow and ion-exchange chromatography. The biological activity of purified HISIL6 was measured by MTT method in an IL-6-dependent cell line 7TD1 to be 2.1 x 10(8) unit/mg.     

Percutaneous balloon pericardiotomy for patients with recurrent pericardial effusion: using a novel double-balloon technique with one long and one short balloon

Percutaneous balloon pericardiotomy is effective and less invasive for the treatment of recurrent pericardial effusion. This study suggests that the double-balloon method with 1 longer and 1 shorter balloon is the procedure of choice for percutaneous balloon pericardiotomy.     

Phylogenetic and physiological diversity of sulphate-reducing bacteria isolated from a salt marsh sediment

Xenopus blastula cells activate different mesodermal genes as a concentration-dependent response to activin, which behaves like a morphogen. To understand how cells recognize morphogen concentration, we have bound naturally labeled activin to cells and related this to choice of gene activation. We find that the increasing occupancy of a single receptor type can cause cells to switch gene expression. Cells sense ligand concentration by the absolute number of occupied receptors per cell (100 and 300 molecules of bound activin induce Xbra and Xgsc, respectively, i.e., 2% and 6% of the total receptors) and not by a ratio of occupied to unoccupied receptors. The long duration of occupancy explains a previously described ratchet effect. Our results suggest a new concept of morphogen gradient formation and interpretation that is particularly well suited to the needs of early development.
