Necrostatin-1 Supplementation to Islet Tissue Culture Enhances the In-Vitro Development and Graft Function of Young Porcine Islets

Pre-weaned porcine islets (PPIs) represent an unlimited source for islet transplantation but are functionally immature. We previously showed that necrostatin-1 (Nec-1) immediately after islet isolation enhanced the in vitro development of PPIs. Here, we examined the impact of Nec-1 on the in vivo function of PPIs after transplantation in diabetic mice. PPIs were isolated from pancreata of 8–15-day-old, pre-weaned pigs and cultured in media alone, or supplemented with Nec-1 (100 µM) on day 0 or on day 3 of culture (n = 5 for each group). On day 7, islet recovery, viability, oxygen consumption rate, insulin content, cellular composition, insulin secretion capacity, and transplant outcomes were evaluated. While islet viability and oxygen consumption rate remained high throughout 7-day tissue culture, Nec-1 supplementation on day 3 significantly improved islet recovery, insulin content, endocrine composition, GLUT2 expression, differentiation potential, proliferation capacity of endocrine cells, and insulin secretion. Adding Nec-1 on day 3 of tissue culture enhanced the islet recovery, proportion of delta cells, beta-cell differentiation and proliferation, and stimulation index. In vivo, this leads to shorter times to normoglycemia, better glycemic control, and higher circulating insulin. Our findings identify the novel time-dependent effects of Nec-1 supplementation on porcine islet quantity and quality prior to transplantation.     

Comparison of pH-dependent sonodisruption of re-assembled casein micelles by 35 and 130 kHz ultrasounds

Sonodisruption behavior of re-assembled casein micelles was compared at two ultrasound frequencies (35 and 130 kHz) by turbidity measurement and laser-diffraction based particle size analysis. Sonochemical ultrasound (130 kHz) was more effective than power ultrasound (35 kHz) in micelle disruption. This was attributed to the higher strain rates generated upon implosion of cavities, as well as the liberation of more free radicals to the surrounding medium. The higher the pH of solution, the more effective was the ultrasonic disruption due to a looser expanded assembly of particles at higher pH values. Sonochemical ultrasound decreased the consistency coefficient of casein solutions and increased their flow index except at a pH value of 6.35, while power ultrasound did not affect the flow behavior of solutions across the whole pH range.     

Controlled-pH tissue cleanup protocol for signal enhancement of small molecule drugs analyzed by MALDI-MS imaging

The limit of detection of low-molecular weight compounds in tissue sections, analyzed by matrix assisted laser desorption ionization mass spectrometry imaging (MALDI-MSI), was significantly improved by employing sample washing using a pH-controlled buffer solution. The pH of the washing solutions were set at values whereby the target analytes would have low solubility. Washing the tissue sections in the buffered solution resulted in removal of endogenous soluble ionization-suppressing compounds and salts, while the target compound remained in situ with minor or no delocalization during the buffered washing procedure. Two pharmaceutical compounds (cimetidine and imipramine) and one new protease inhibitor compound were successfully used to evaluate the feasibility of the pH-controlled tissue washing protocol for MALDI-MSI. Enhancement in signal-to-noise ratio was achieved by a factor of up to 10.     

Synthesis and Characterization of the Antibacterial Activity of Zinc Oxide Nanoparticles against Salmonella typhi

Metal oxides can be used as a series of new and effective anti-bacterial agents.In this study,four concentrations of ZnO nanoparticles(0.2,0.5,0.7 and 1.0 mol/L) were synthesized using a low-temperature sol-gel method annealed at400 and 550 °C.The products were characterized by X-ray diffraction(XRD),transmission electron microscopy(TEM)and Fourier transform infrared spectroscopy(FTER).XRD results show the hexagonal wurtzite structure of the nanoparticles with the grain size in the range of 38-43 nm.TEM micrographs exhibit a polyhedral form of the synthesized nanoparticles.The antimicrobial activity of different concentrations of nanoparticles against Salmonella typhi PTCC 1609 was determined by disk diffusion and agar dilution method at five concentrations of 10,5,2.5,1.25 and 0.625 mg/mL.Analysis shows that the prepared ZnO nanoparticles have a very effective antimicrobial activity against Salmonella typhi.This activity increases by reducing the size of nanoparticles and increasing their content in the bacterial growth medium.     

Contribution of City Prosperity to Decisions on Healthy Building Design:A cases tudy of Tehran

In this paper,the city prosperity manifest is analyzed through a logical process and a framework is then proposed for designing healthy buildings in Tehran. The current status of urbanization in terms of the number of people living together and the changes in old behavior and perspectives have resulted in deficiencies to the health and hygiene of buildings apart from their surrounding environment. Consequently,these problems have affected people's well-being.This study mainly aims to determine policies and strategies for the architecturaldesign of healthy buildings according to health and safety conditions that influence the quality of internal spaces and external environment of cities.The study is conducted based on logical reasoning and uses focus group and in-depth interviews to assess the final result. The result is aframework that suggests a number of policies that can promote the mental and physical health as well as hygiene of residents through healthy buildings.     

Ethylene glycol biodegradation in microbial fuel cell

Ethylene glycol is an environmental pollutant, which exists in airport runoff and industrial waste. In this article, biodegradation of ethylene glycol in a two-chamber, batch-mode microbial fuel cell was investigated. Glucose and ethylene glycol at different concentrations were used as carbon and energy sources. Chemical oxygen demand removal in the range of 92–98% indicated that microbial fuel cell can be used for biodegradation of ethylene glycol. Ethylene glycol also improved power generation and the maximum power density was 5.72 mW/m² (137.32 mW/m³), with respect to the same glucose and ethylene glycol concentrations (500 ppm).     

Development of an integrated model for process planning and parameter selection for machining processes

The concept of ‘do it right the first time’ in the machining industry not only expects the best quality products but also at the best possible cost. The cost of machining depends on intelligent process planning and selection of machining parameters such as speed, feed, and depth of cut. The problem of machining parameter selection has received great attention by researchers and many techniques have been developed. A review of these techniques reveals that the selection of the machine and cutting tool is done before the process of cutting parameter selection and process sequencing, and often the selection is based on experience. The current research is an attempt to develop an integrated model (ExIMPro: Expert system based Integrated model for Machining Processes) which finds the sequence of operations with set of machines, tools, and other process parameters to minimise the cost of machining for a cylindrical part. This system consists of existing expert system Machining Parameter SELection (MPSEL) for machine and tool selection and a Microsoft Excel® and Visual Basic® based parameter selection model. The present model focuses on turning and cylindrical grinding operations but other processes can be incorporated with little modification to the software.
