(1) Background: We previously demonstrated that disruption of IP6K1 improves metabolism, protecting mice from high-fat diet-induced obesity, insulin resistance, and non-alcoholic fatty liver disease and steatohepatitis. Age-induced metabolic dysfunction is a major risk factor for metabolic diseases. The involvement of IP6K1 in this process is unknown. (2) Methods: Here, we compared body and fat mass, insulin sensitivity, energy expenditure and serum-, adipose tissue- and liver-metabolic parameters of chow-fed, aged, wild type (aWT) and whole body Ip6k1 knockout (aKO) mice. (3) Results: IP6K1 was upregulated in the adipose tissue and liver of aWT mice compared to young WT mice. Moreover, Ip6k1 deletion blocked age-induced increase in body- and fat-weight and insulin resistance in mice. aKO mice oxidized carbohydrates more efficiently. The knockouts displayed reduced levels of serum insulin, triglycerides, and non-esterified fatty acids. Ip6k1 deletion partly protected age-induced decline of the thermogenic uncoupling protein UCP1 in inguinal white adipose tissue. Targets inhibited by IP6K1 activity such as the insulin sensitivity- and energy expenditure-inducing protein kinases, protein kinase B (PKB/Akt) and AMP-activated protein kinase (AMPK), were activated in the adipose tissue and liver of aKO mice. (4) Conclusions: Ip6k1 deletion maintains healthy metabolism in aging and thus, targeting this kinase may delay the development of age-induced metabolic dysfunction. 相似文献

Typically developing children understand and predict others' behavior by extracting and processing relevant information such as the logic of their actions within the situational constraints and the intentions conveyed by their gaze direction and emotional expressions. Children with autism have difficulties understanding and predicting others' actions. With the use of eye tracking and behavioral measures, we investigated action understanding mechanisms used by 18 children with autism and a well-matched group of 18 typically developing children. Results showed that children with autism (a) consider situational constraints in order to understand the logic of an agent's action and (b) show typical usage of the agent's emotional expressions to infer his or her intentions. We found (c) subtle atypicalities in the way children with autism respond to an agent's direct gaze and (d) marked impairments in their ability to attend to and interpret referential cues such as a head turn for understanding an agent's intentions. We report that cation-selective transmembrane pores formed by synthetic p-octiphenyl beta barrels with internal aspartate residues can be transformed into anion-permeable metallopores with internal Mg(2+)-aspartate complexes. These metallopores are shown to be useful for fluorimetric sensing of a broad variety of organic anions of biological relevance such as phytate, heparin, thiamine phosphates, and adenosine triphosphate. The negligible flippase activity measurable for Mg(2+)-free pores indicates that transmembrane p-octiphenyl beta barrels do not disturb the lipid bilayer suprastructure, in other words, they form barrel-stave rather than toroidal pores. 相似文献

Electrophoretic karyotype studies have shown that clinical isolates of Candida albicans have extensive chromosome length polymorphisms. Chromosome translocation is one of the causes of karyotypic variation. Chromosome translocation events have been shown to occur very frequently at or near the major repeat sequence (MRS) on chromosomes. The MRS consists of the repeated sequences RB2, RPS and HOK, and the repeated sequences are considered to be the template for recombination. To investigate which element of the MRS is important for chromosome translocation, we constructed three cassettes, each containing a URA blaster and sequences homologous to one of the repeats, for insertion into the MRS region on the chromosomes. The ura3 strain STN22u2, which shows a stable, standard karyotype, was transformed with each construct. Insertion events with each cassette occurred at almost all chromosomes. Insertion into the RB2 repeat, but not into the RPS repeat, was accompanied by chromosome translocation in some transformants: chromosome translocations between chromosomes R and 7 and chromosomes 1 and 7 were found, as well as deletions of 7A and 7C from chromosome 7. The buffy-headed marmoset (Callithrix flaviceps) is apparently the only predispersal herbivore of the seeds of Siparuna guianensis at the Caratinga Biological Station in southeastern Brazil. Both the fruit receptacles and the frutioles (seeds) of S. guianensis are relatively rich in nutrients such as carbohydrates, proteins, and lipids, but the receptacles contain high concentrations of benzylisoquinoline alkaloids. The latter presumably act as a qualitative chemical defense, impeding the access of potential predators to the alkaloid-poor frutioles. However, on ripening, the receptacle splits open, exposing the frutioles, which enables C. flaviceps to avoid the plant's chemical defenses effectively. Taking care to avoid contact with the receptacle, the marmosets pluck out the frutioles and ingest the seeds. Qualitative and quantitative changes (in particular a significant reduction in daily ranging) in the marmosets' behavior during the period when S. guianensis frutioles were accessible indicate that this was a preferred plant resource. This is the first record of the consumption of S. guianensis seeds by callitrichine monkeys (which are not known to be systematic seed eaters), despite the fact that both are widely distributed in the Neotropics. It is thus possible that the behavioral strategy observed here is a unique phenomenon resulting from a specific combination of factors, including the abundance of S. guianensis within the study area. The lack of other records may nevertheless be a result of insufficient sampling effort, in which case, the feeding strategies of callitrichines may have an important influence on the reproductive biology of S. guianensis in many areas. 相似文献

A network of biological databases is reviewed, supplying a framework for studies of human genes and the association of their genomic variations with human phenotypes. The network is composed of GeneCards, the human gene compendium, which provides comprehensive information on all known and predicted human genes, along with its suite members GeneDecks and GeneLoc. Two databases are shown that address genes and variations focusing on olfactory reception (HORDE) and transduction (GOSdb). In the realm of disease scrutiny, we portray MalaCards, a novel comprehensive database of human diseases and their annotations. Also shown is GeneKid, a tool aimed at generating novel kidney disease biomarkers using systems biology, as well as Xome, a database for whole-exome next-generation DNA sequences for human diseases in the Israeli population. Finally, we show LifeMap Discovery, a database of embryonic development, stem cell research and regenerative medicine, which links to both GeneCards and MalaCards. 相似文献

Taking John Willinsky's observation that in Canadian identity politics, the question 'who am I?' often necessitates the additional question 'where is here?' (Willinsky, 1998), this article examines excerpts from interviews with a group of teacher education students in order to explore how they used family (hi)stories about 'country of origin' in the construction of their identities. Making 'country of origin' problematic, the article also examines the limits and possibilities of being and becoming Canadian. 'Where are you from?' complicates and challenges the entitlement to belong. Drawing on critiques by Canadian educators, the article challenges the widely held significations given to Canadian immigration and to official policies of multiculturalism. By juxtaposing data from eight interviews, the issue of racial identity is explored, in terms of how it appears or does not appear in the stories the students tell. These juxtapositions highlight the unofficial intersections between Canadian identity, race and ethnicity. 相似文献
