Clinical guidelines recommend concurrent treatment of anemia in end‐stage renal disease with erythropoiesis‐stimulating agents (ESAs) and iron. However, there are mixed data about optimal iron supplementation. To help address this gap, the relationship between iron markers and hemoglobin (Hb) response to ESA (Epoetin alfa) dose was examined. Electronic medical records of 1902 US chronic hemodialysis patients were analyzed over a 12‐month period between June 2009 and June 2010. The analysis included patients who had at least one Hb value during each 4‐week interval for four consecutive intervals (k ? 2, k ? 1, k, and k + 1; k is the index interval), received at least one ESA dose during intervals k ? 1 or k, had at least one transferrin saturation (TSAT) value at interval k, and at least one ferritin value during intervals k ? 2, k ? 1, or k. Effect modification by TSAT and ferritin on Hb response was evaluated using the generalized estimating equations approach. Patients had a mean (standard deviation) age of 62 (15) years; 41% were Caucasian, 34% African American, 65% had hypertension, and 39% diabetes. Transferrin saturation, but not ferritin, had a statistically significant (P < 0.05) modifying effect on Hb response. Maximum Hb response was achieved when TSAT was 34%, with minimal incremental effect beyond these levels. Of the two standard clinical iron markers, TSAT should be used as the primary marker of the modifying effect of iron on Hb response to ESA. Long‐term safety of iron use to improve Hb response to ESA warrants further study. 相似文献

PURPOSE: Rhabdomyosarcomas (RMS) are heterogeneous in their clinical presentation, histology, and cytogenetics. The growth of some RMS cells has been found to be regulated by the tyrosine kinase insulin-like growth factor (IGF) type I receptor. However, RMS cells exhibit variable sensitivity to inhibitors of tyrosine kinases and IGF receptors. Collectively, these heterogeneous features suggest that differences exist in the growth regulatory pathways of RMS. The objective of this study is to identify active tyrosine kinase signal transduction pathways in embryonal and alveolar RMS cells. METHODS: RMS tumor samples and cell lines representing both embryonal and alveolar histologic subtypes have been analyzed by immunoprecipitation and immunoblotting techniques to characterize phosphotyrosyl protein patterns and to identify tyrosine phosphorylated proteins. RESULTS: RMS cells can be characterized based on the patterns of phosphotyrosyl proteins, including the phosphorylation status of the catenin-like protein Cas1 and the signal adapter protein SHC, and the activation of IGF type I receptor signaling cascades including the formation of SHC-GRB2 signal protein complexes and MAP kinase activation. CONCLUSIONS: Rhabdomyosarcomas, especially the embryonal histologic subtype, are heterogeneous at the level of tyrosine kinase signal transduction. It will be important to characterize the growth regulatory pathways active in individual RMS tumors before targeting molecular therapies to this malignancy. 相似文献

Tungsten heavy alloys (WHAs) are metal–metal composites consisting of nearly pure spherical tungsten particles embedded in a Ni–Fe–W or Ni–Co–W or Ni–Cu–W ductile matrix. In this dual phase alloy, there are several complicated relations between the ductile matrix and hard tungsten particles. The aim of this research was to examine the effect of varying tungsten content on the microstructure and mechanical properties of tungsten heavy alloys. The microstructural parameters (grain size, connectivity, contiguity and solid volume fraction) were measured and were found to have a significant effect on the mechanical properties of tungsten-based heavy alloys. The result shows that the binding strength between the W and the matrix phase has a major influence on the ductility of tungsten-based alloys. Synthesis of zirconia nanoparticles by flame spray pyrolysis (FSP) at high production rates is investigated. Product powder is collected continuously in a baghouse filter unit that is cleaned periodically by air-pressure shocks. Nitrogen adsorption (BET), X-ray diffractometry (XRD), transmission electron microscopy (TEM), and thermogravimetric analysis (TGA) are used to characterize the product powder. The effect of powder production rate (up to 600 g/h), dispersion gas flow rate, and precursor concentration on product particle size, crystallinity, morphology, and purity is investigated. The primary particle size of zirconia is controlled from 6 to 35 nm, while the crystal structure consists of mostly tetragonal phase (80–95 wt%), with the balance monoclinic phase at all process conditions. The tetragonal crystal size is close to the primary particle size, which indicates weak agglomeration of single crystals. 相似文献

Banyan tree saw dust powder (BSD) filled Polypropylene (PP) green composites have been fabricated with varying amounts viz., 0%, 20%, and 40% of BSD particulate filler by using a co‐rotating twin screw extruder followed by injection molding. The mechanical properties such as surface hardness, tensile behavior, and impact strength of the fabricated PP/BSD green composites have been studied in order to standardize the composites. Abrasive water jet (AWJ) machining has been reported mainly for ceramics, concrete, and glass but not much literature is available on AWJ machining of polymer composites. This research is aimed at examining the AWJ machining of green polymer composites. The effect of BSD loading on the AWJ machining behavior of the PP/BSD green composites has been investigated. Furthermore, the effect of addition of 4% maleic anhydride grafted PP (coupling agent) and 4% talc (mineral filler) on the machining behavior of PP/BSD composites has also been evaluated. Surface roughness and optical micrographs of the AWJ cut composite specimens were examined to assess the effect of BSD content, AWJ traverse speed and pressure on the machining behavior of the composites. In order to probe the mechanism of AWJ machining behavior of PP/BSD composites, the kerf width and taper have been measured and results are correlated. POLYM. MoB and SiC particulate reinforced MoSi2 matrix composites were synthesized in situ from Mo, Si, and B4C powder mixtures by self‐propagating high‐temperature synthesis (SHS). The SHS MoSi2–MoB–SiC products were vacuum hot‐pressed (HPed) at 1400°C for 90 min to fabricate high‐density (> 97.5% relative density) bulk composites. Microstructure refinement and improvements in the Vickers hardness and fracture toughness of the HPed composites were observed with increasing B4C content in the reaction mixture. The HPed composite of composition MoSi2–0.4MoB–0.1SiC exhibited grain size of 1–5 μm, Vickers hardness of 12.5 GPa, bending strength of 537 MPa, and fracture toughness of 3.8 MPa.m1/2. These excellent mechanical properties indicate that MoB and SiC particulate reinforced MoSi2 composites could be promising candidates for structural applications. 相似文献

In this work, we assessed the in-vitro effects of eicosapentaenoic acid (EPA; C20:5n-3) and docosahexaenoic acid (DHA; C22:6n-3) (final concentration, 15 microM) on T cell blastogenesis, interleukin-2 and -4 (IL-2, IL-4) secretion, fatty acid composition and intracellular oxidative status in type I diabetic patients with or without complications. Con A stimulated lymphocyte proliferation, glucose uptake, intracellular reduced glutathione levels and catalase activity were lower in diabetics as compared to controls, regardless to the presence of complications. EPA and DHA diminished T-lymphocyte proliferation and IL-2 production but enhanced IL-4 secretion in both diabetic and control groups. No changes in the levels of reduced glutathione and in the activities of catalase and SOD were observed in control T cells cultured in the presence of EPA and DHA. However, in diabetic patients, addition of n-3 PUFA to culture induced an increase in T cell levels of reduced glutathione and hydroperoxide, and in activities of catalase and SOD. Low levels of arachidonic acid (C20:4n-6) were found in plasma membrane phospholipids of lymphocytes from diabetic patients compared to controls. Incubation of lymphocytes with EPA and DHA was associated with an incorporation of these fatty acids in membrane phospholipids. In conclusion, the beneficial effects of n-3 PUFA on T cell functions in type I diabetes could be attributed to their suppressive action and modulation of cytokine secretion, and to the improvement of intracellular oxidative status. 相似文献

Electron beam welding (EBW) technique is becoming popular in nuclear, chemical and aerospace industries due to its high penetration depth and fast cooling rate. Samples of Hastelloy C-276 have been welded by electron beam (EB). A scanning electron microscope (SEM) having the attachment of an energy dispersive system (EDS) has been employed to study the resulting microstructure and micro-eutectic phases. The microstructure of the molten zone (MZ) is found to be of fine lamellar type. The hardness of the MZ is found to be 35% higher compared to as-received alloy. The micro-eutectoids are rich in Mo and W. The X-ray diffraction patterns of the MZ show shifting of peaks towards higher angle compared to the diffraction pattern of the as-received alloy. Broadening of the peaks is also observed in the diffraction pattern of the MZ. Formation of the μ-phase was observed in the MZ after a tempering treatment at 950 °C. 相似文献
