The effect of cranberry juice (CJ) and cranberry proanthocyanidins (PAC) on the infectivity of human enteric virus surrogates, murine norovirus (MNV-1), feline calicivirus (FCV-F9), MS2(ssRNA) bacteriophage, and phiX-174(ssDNA) bacteriophage was studied. Viruses at high (∼7 log10 PFU/ml) or low (∼5 log10 PFU/ml) titers were mixed with equal volumes of CJ, 0.30, 0.60, and 1.20 mg/ml final PAC concentration, or water and incubated for 1 h at room temperature. Viral infectivity after treatments was evaluated using standardized plaque assays. At low viral titers, FCV-F9 was undetectable after exposure to CJ or the three tested PAC solutions. MNV-1 was reduced by 2.06 log10 PFU/ml with CJ, and 2.63, 2.75, and 2.95 log10 PFU/ml with 0.15, 0.30, and 0.60 mg/ml PAC, respectively. MS2 titers were reduced by 1.14 log10 PFU/ml with CJ, and 0.55, 0.80, and 0.96 log10 PFU/ml with 0.15, 0.30, and 0.60 mg/ml PAC, respectively. ?-X174 titers were reduced by 1.79 log10 PFU/ml with CJ, and 1.95, 3.67, and 4.98 log10 PFU/ml with PAC at 0.15, 0.30, and 0.60 mg/ml, respectively. Experiments using high titers showed similar trends but with decreased effects. CJ and PAC show promise as natural anti-virals that could potentially be exploited for foodborne viral illness treatment and prevention.  相似文献

Nanodiamond comb-shaped lateral field emitter arrays in diode configuration were fabricated and characterized for high current field emission. Nitrogen-incorporated nanocrystalline diamond with grain size of 5–10 nm was micropatterned using RIE to realize interconnected arrays of comb structures equipped with uniformly spaced high aspect ratio lateral emitter fingers. A 9000-fingered nanodiamond lateral comb array diode with an inter-electrode spacing of 8 μm demonstrated a high emission current of  25 mA at an anode voltage of 260 V (electric field  32 V/μm) in 10− 7 Torr vacuum. The lateral emitter configuration shows potential for higher power with no emission current saturation observed. These vacuum micro/nanoelectronic devices comprised of nanodiamond lateral field emission diodes are attractive for low-voltage operating high current electron sources, high-power and high-speed switches, and other extreme demand/extreme environment electronics.  相似文献

Carcasses along slaughter lines were exposed to normal slaughterhouse air or ultraclean air provided from a unit fitted with a HEPA filter. In cattle slaughterhouses, aerobic viable counts were measured by sponging the brisket at the end of the line to determine whether the slaughterhouse air had led to contamination of the carcasses. Furthermore, a replica cattle carcass with settle plates attached was exposed to similar conditions. The greatest contamination of the plates occurred at the hide puller (P < 0.01). The use of ultraclean air reduced the deposition of organisms onto settle plates (P < 0.01). The airborne route contributed to contamination in cattle slaughterhouses, but other vectors were more important. Further study of contamination of the brisket, at the time that it was first exposed, showed that knives transfer contamination from the hide. The use of ultraclean air at this position showed that the airborne route was a contributor to contamination (P < 0.1), but it was not the greatest vector. In lamb slaughterhouses, the highest counts on settle plates were found at the fleece puller (P < 0.05). The highest counts on the lamb carcasses were found on the brisket exposed from the start of the line to just after the fleece puller (P < 0.05). There was no clear relationship between the measured counts and the concentration of organisms in the air, indicating that the airborne route in lamb slaughterhouses contributes less to carcass contamination than do the surface contacts.  相似文献

Studies to determine the appropriateness of the use of populations of indicator bacteria on poultry carcasses for process verification were undertaken in commercial slaughterhouses. Samples were collected from neck skin by excision or from whole carcass rinses and were examined for a range of presumptive process hygiene indicator bacteria. Coefficients of variation were calculated for each bacterial indicator and were significantly lower in excised samples, indicating more reproducible bacterial recovery by this sampling method. Total viable counts of aerobic bacteria, Enterobacteriaceae, and Pseudomonas in samples collected by excision had the lowest coefficients of variation when compared with other indicators and were therefore used for further study. The uncertainties associated with the quantification of each bacterial indicator were calculated and were lowest overall for total viable counts of aerobic bacteria. In general, uncertainty was higher for lower bacterial numbers. Results of microbiological testing on pooled excised neck skin samples were not significantly different from the mean of individually analyzed samples. Bacterial numbers increased by 1 log unit when cultures were stored under chilled conditions typical of those used for transporting samples to external laboratories, but the increases were not significant for Pseudomonas and aerobic bacteria when storage time was less than 17 h. Weak relationships were identified between bacterial indicator numbers and duration of processing, although cleanliness of the processing environment diminished visibly during this time. In the plants visited for this study, there was a poor relationship between presumptive bacterial indicator numbers and process hygiene. Consequently, bacterial analyses for process verification purposes may be of limited value.  相似文献

An assessment of the proposed new International Organization for Standardization quantitative method for Campylobacter was undertaken on poultry carcass samples collected after the chilling phase of processing. Using a critical differences method, we determined the uncertainty associated with log-transformed Campylobacter numbers by dual analyses of 346 samples collected from 22 processing plants located throughout the United Kingdom. Overall, using log-transformed Campylobacter numbers that ranged between -1 and 5 log, we calculated the expanded measurement of uncertainty (EMU) to be 3.889 for the new method. The EMU changed when ranges of bacterial numbers were grouped for analyses. For low numbers of Campylobacter (< 1 log), the EMU was calculated to be 5.622. There was less measurement error with higher bacterial numbers because the EMU was found to be 0.612 for samples containing Campylobacter numbers of 3 log or above. The draft method was used to measure numbers of Campylobacters on poultry carcasses collected from 18 United Kingdom processing plants in summer and winter. Numbers were significantly lower in winter. We conclude that, although the new method is adequate at quantifying high numbers of Campylobacter on poultry carcasses, further development is required to improve the measurement of small numbers of this causative agent of foodborne illness.  相似文献

Prediction of risks and therapeutic outcome in nuclear medicine largely rely on calculation of the absorbed dose. Absorbed dose specification is complex due to the wide variety of radiations emitted, non-uniform activity distribution, biokinetics, etc. Conventional organ absorbed dose estimates assumed that radioactivity is distributed uniformly throughout the organ. However, there have been dramatic improvements in dosimetry models that reflect the substructure of organs as well as tissue elements within them. These models rely on improved nuclear medicine imaging capabilities that facilitate determination of activity within voxels that represent tissue elements of approximately 0.2-1 cm(3). However, even these improved approaches assume that all cells within the tissue element receive the same dose. The tissue element may be comprised of a variety of cells having different radiosensitivities and different incorporated radioactivity. Furthermore, the extent to which non-uniform distributions of radioactivity within a small tissue element impact the absorbed dose distribution is strongly dependent on the number, type, and energy of the radiations emitted by the radionuclide. It is also necessary to know whether the dose to a given cell arises from radioactive decays within itself (self-dose) or decays in surrounding cells (cross-dose). Cellular response to self-dose can be considerably different than its response to cross-dose from the same radiopharmaceutical. Bystander effects can also play a role in the response. Evidence shows that even under conditions of 'uniform' distribution of radioactivity, a combination of organ dosimetry, voxel dosimetry and dosimetry at the cellular and multicellular levels can be required to predict response.  相似文献

Research on the radiation-induced bystander effect has been carried out mainly in 2-D tissue culture systems. This study uses a 3-D model, wherein apparently normal human diploid fibroblasts (AG1522) are grown in a carbon scaffold, to investigate the induction of a G(1) checkpoint in bystander cells present alongside radiolabelled cells. Cultures were simultaneously pulse-labelled with (3)H-deoxycytidine ((3)HdC) to selectively irradiate a minor fraction of cells, and bromodeoxyuridine (BrdU) to identify the radiolabelled cells. After thorough washing of cultures, iododeoxyuridine (IdU) was administered to detect proliferating bystander cells. The cultures were harvested at various times thereafter, and cells were reacted with two monoclonal antibodies specific to IdU/BrdU or BrdU, respectively, stained with propidium iodide, and subjected to multi-parameter flow cytometry. Cell-cycle progression was followed in radiolabelled cells (BrdU(+)) that were chronically irradiated by low energy beta particles emitted by DNA-incorporated (3)H, and in unlabelled bystander cells (BrdU(-)) by a flow cytometry based cumulative labelling index assay. As expected, radiolabelled cells were delayed, in a dose-dependent manner, in G(2) and subsequently G(1). No delay occurred in progression of bystander cells through G(1), when the labelled cells were irradiated at dose rates up to 0.32 Gy h(-1).  相似文献

The austenitic stainless steel UNS S63019 was evaluated regarding its potential as bipolar plate material in a polymer electrolyte membrane fuel cell (PEMFC) environment. Segregated grains of niobium carbide (NbCx) were identified in polished cross-sections of the alloy, offering a possible pathway for enhanced electrical conductivity through the passive surface oxide. Additionally, the alloy was tested for corrosion resistance in a simulated PEMFC environment. It was considered that perhaps the elevated nitrogen concentration in the alloy would provide some benefit for corrosion resistance.Results for interfacial contact resistance (ICR) testing of the air-formed surface film on UNS S63019 showed decreased electrical conductivity as compared to UNS S30400. Niobium carbide particles did not improve film conductivity due to a non-conductive niobium oxide layer that formed on the surface. Corrosion resistance of the alloy was also poor as compared with UNS S30400, demonstrating that elevated nitrogen concentration in the alloy was not adequate in itself to enhance corrosion resistance. Poor corrosion resistance was attributed primarily to high carbon content in the alloy which combined with a significant amount of chromium to form carbides.  相似文献

The coarsening of -Al2O3 crystals to a 'critical size' is often interpreted as the first step in the shear nucleation of -Al2O3. The existence of this so-called critical size has also been used to explain the observation that -Al2O3 nuclei are generally twice as large as the crystals in the -Al2O3 matrix. This paper discusses the important issues in the nucleation of -Al2O3 from -Al2O3. A few key experiments are also presented to clarify the nucleation process. It is concluded that a critical -Al2O3 crystal size is not a prerequisite for -Al2O3 nucleation, but is primarily a result of the incubation time required to produce -Al2O3 nuclei by diffusional nucleation. It is proposed that the large observed -Al2O3 crystal size also does not result from a shear nucleation event in a 'critical size' -Al2O3 crystal, but is due to the intrinsically low -Al2O3 nucleation density, together with rapid growth of -Al2O3 after nucleation.w  相似文献

This paper reports a Monte Carlo simulation where a single atom scattering model is adopted. The element taking part in each electron-atom interaction is selected on the basis of its contribution eitherto the total elastic cross section or to the electron's mean free path. Both Rutherford and Mott scattering are considered, with the continuous slowing down process of Bethe used to calculate the energy loss to the system. The backscattered electron coefficients show good agreement with experimental results from a large group of low atomic number materials when using a model which selects the scattering atom by its contribution to the whole compound calculated from its atomic fraction of the total elastic cross-section.  相似文献
