The quantitative measurement of fat in minced meat by video techniques has, to date, been impractical due to the nature of the raw material and camera limitations in particle resolution. The latter have been enhanced by improving the optical system and by illuminating the meat with a limited wavelength ultraviolet light source. Problems with fat smear and drip stain, causing over- and under-estimation of fat, respectively, have been overcome by better control of mincing conditions. Tempering of the meat to between -2°C and -5°C before mincing gave the best results. The level of fat detection was unchanged for up to 20 min after mincing, but, by 2 h, the initial value had fallen by 50%. Magnifying the field of view, thus reducing the sample size measured by 50%, did not adversely affect results with 4mm-10mm diameter mince. The system could not accurately resolve mince below 4 mm. Estimation of total lipid from video data using prediction equations was in good agreement with chemical analysis (r=0·99 for 10mm mince, r=0·95 for 4 mm mince). Video image analysis (VIA) can now be effectively extended to areas of meat processing using particulate meats.  相似文献

PM28A is a major intrinsic protein of the spinach leaf plasma membrane and the major phosphoprotein. Phosphorylation of PM28A is dependent in vivo on the apoplastic water potential and in vitro on submicromolar concentrations of Ca2+. Here, we demonstrate that PM28A is an aquaporin and that its water channel activity is regulated by phosphorylation. Wild-type and mutant forms of PM28A, in which putative phosphorylation sites had been knocked out, were expressed in Xenopus oocytes, and the resulting increase in osmotic water permeability was measured in the presence or absence of an inhibitor of protein kinases (K252a) or of an inhibitor of protein phosphatases (okadaic acid). The results indicate that the water channel activity of PM28A is regulated by phosphorylation of two serine residues, Ser-115 in the first cytoplasmic loop and Ser-274 in the C-terminal region. Labeling of spinach leaves with 32P-orthophosphate and subsequent sequencing of PM28A-derived peptides demonstrated that Ser-274 is phosphorylated in vivo, whereas phosphorylation of Ser-115, a residue conserved among all plant plasma membrane aquaporins, could not be demonstrated. This identifies Ser-274 of PM28A as the amino acid residue being phosphorylated in vivo in response to increasing apoplastic water potential and dephosphorylated in response to decreasing water potential. Taken together, our results suggest an active role for PM28A in maintaining cellular water balance.  相似文献

Two miners were fatally injured when a pillar bump occurred during retreat mining in a southern West Virginia coal mine. The mine was operating in the Eagle seam with overmining in the No. 2 Gas and Powellton seams. A coal bump is defined as a sudden and violent failure of coal caused by the release of stored strain energy in the pillar. While significant strides have been made by academia, industry,and regulatory agencies to better understand bump conditions and mitigation techniques, coal bumps represent a long standing, highly site-specific engineering problem in which the exact failure mechanism is not clearly understood. In this case history, a cut-by-cut analysis of retreat mining operations was conducted on the 4 East Main leading up to the pillar bump event. Numerical input parameters were derived from site-specific geologic information and mine geometry for the analysis of pillar stress conditions and energy release using LaModel. An overview of stress conditions in the panel was presented including a precursor event that occurred two crosscuts inby the bump site. The methodology presented in the paper for the evaluation of the fatal bump event can be used for the identification of bump prone conditions prior to development and retreat of a mining area.  相似文献

A study of the electrochemistry of trace metals in heavy metal fluoride melts at 550 °C has been conducted at a stationary electrode with a view to establishing electroanalytical methods for these melts. Reproducible differential pulse voltammetry traces were obtained, the trace metal oxidation peaks behaving as predicted by the theory of Osteryoung et al. The peak current for Fe2+ reduction in a fluoride melt was found to be linear in pulse amplitude up to 70 mV and linear in (pulse duration)–1/2. The peak potential was also found to be a linear function of pulse amplitude. Scan rates around 3 mV s–1 and sampling times around 8 ms were found to produce the optimum analytical response.  相似文献

Reconsideration of data from stress corrosion cracking tests conducted at extreme strain rates on homogeneous noble-metal alloys leads to the conclusion that the data are more consistent with the film-induced ‘cleavage’ (in this case intergranular fracture) model than with the surface mobility model. At the highest strain rates, the data for several alloy systems are close to what would be expected from a single brittle event followed by ductile rupture. The opportunity is also taken to comment on some misconceptions regarding the predictions of the film-induced cleavage model, and more generally the correlation between dealloying and SCC in homogeneous noble-metal alloys.  相似文献
