BACKGROUND: Detoxification is an essential process in all living organisms. Humans accumulate heavy metals primarily as a result of lifestyle and environmental contamination. However, not all humans experience the estimated individual exposure. This suggests the presence of genetic regulatory mechanisms. OBJECTIVE: In order to identify genetic factors underlying the inter-individual variance in detoxification capacity for the heavy metal mercury, 192 students were investigated. We focused on the relationship between polymorphisms in glutathione-S-transferase (GST) genes and mercury concentrations in blood, urine, and hair. The correlation between blood mercury levels, GSTT1 and GSTM1 polymorphism, and gene expression of certain metallothionein subgroups (MT1, MT3) was evaluated in a further group of students (N=30). METHODS: Mercury levels in acid digested samples were measured by cold vapor AAS. Genotyping of the GSTT1 and GSTM1-gene deletion polymorphism was performed by means of PCR. Gene expression of several MT genes was analyzed in lymphocytes from fresh peripheral blood by semiquantitative RT-PCR. RESULTS: The following was noted: a) hair mercury concentrations are significantly increased in persons with the double deleted genotype (GSTT1-/- and GSTM1-/-) as compared to persons with the intact genotype, and b) MT1X expression is higher in persons with the intact genotype (GSTT1+/+ and GSTM1+/+). CONCLUSIONS: We conclude that the epistatic effect of the GSTT1 and the GSTM1 deletion polymorphism is a risk factor for increased susceptibility to mercury exposure. The relationship between MT gene expression and GST gene polymorphisms needs further investigation. If MT expression depends on GST polymorphisms it would have important implications on the overall metal detoxification capability of the human organism. 相似文献

Gas chromatography and the determination of natural isotope ratios are powerful analytical methods which can be used to check the authenticity of alcoholic beverages and to detect any adulteration. To check the origin and the authenticity of commercial fruit spirits, whiskies, etc., 197 samples were analysed by gas chromatography, 2H-NMR and 13C isotope mass spectrometry. The discrimination between different varieties was demonstrated by bivariate and multivariate discriminant analysis using different concentrations of volatile compounds such as methanol, butan-1-ol, 2- and 3-methyl-butanol, benzaldehyde and hexanol as well as isotopic data like (D/H)I, (D/H)II and 13C/12C isotopomers of ethanol. The results show that by using multivariate discriminant analysis it is possible to distinguish not only between different groups of spirits, e.g. those made of stone-fruit, malaceous fruit, grain and corn, but also between individual varieties, such as cherry, plum, mirabelle and apple. If the detection of highly rectified ethyl alcohol of agricultural origin and the identification of its raw materials are required, then natural isotope ratios are the only discriminant analytical parameters available. 相似文献

BACKGROUND: Optimization of nitrogen (N) and sulfur (S) nutrition of wheat influences the bread‐making quality of grain. This study was conducted to determine whether the application of N and S in winter wheat at anthesis stage during two field trials could influence the absorption and distribution of N and S in grain and flour bread‐making quality RESULTS: The incorporation of 15N and 34S tracers in various plant parts (leaves, stems, spikes) was determined by isotopic ratio mass spectrometry coupled with an elemental analyser (EA‐IRMS). The effects of foliar fertilization on percentage of 15N and 34S recovery in various plant parts, grain yield, flour protein content, and dough properties were investigated after a combination of treatments: N (urea), S (micronized elemental S) and NS (both urea and micronized elemental S). Nitrogen (70–80%) and S (35–55%) were quickly assimilated during the days following application. Sixty days after application, 42–68% N and 6–12% S were recovered in the grain. When N and S fertilizers were applied simultaneously, N and S recoveries (68% and 12%), N/S ratios (11–14), flour protein content (115 g kg?1) and dough strength (310 × 10?4 J), swelling (21.8 mmH2O) and extensibility (96.4 mm) were increased. CONCLUSION: A synergistic effect between the foliar applied N and S fertilizers appears to increase N and S assimilation in grain and may improve bread‐making qualities. Axial-grooved gas-lubricated journal bearings have been widely applied to precision instrument due to their high accuracy, low friction, low noise and high stability. The rotor system with axial-grooved gas-lubricated journal bearing support is a typical nonlinear dynamic system. The nonlinear analysis measures have to be adopted to analyze the behaviors of the axial-grooved gas-lubricated journal bearing-rotor nonlinear system as the linear analysis measures fail. The bifurcation and chaos of nonlinear rotor system with three axial-grooved gas-lubricated journal bearing support are investigated by nonlinear dynamics theory. A time-dependent mathematical model is established to describe the pressure distribution in the axial-grooved compressible gas-lubricated journal bearing. The time-dependent compressible gas-lubricated Reynolds equation is solved by the differential transformation method. The gyroscopic effect of the rotor supported by gas-lubricated journal bearing with three axial grooves is taken into consideration in the model of the system, and the dynamic equation of motion is calculated by the modified Wilson-0-based method. To analyze the unbalanced responses of the rotor system supported by finite length gas-lubricated journal bearings, such as bifurcation and chaos, the bifurcation diagram, the orbit diagram, the Poincar6 map, the time series and the frequency spectrum are employed. The numerical results reveal that the nonlinear gas film forces have a significant influence on the stability of rotor system and there are the rich nonlinear phenomena, such as the periodic, period-doubling, quasi-periodic, period-4 and chaotic motion, and so on. The proposed models and numerical results can provide a theoretical direction to the design of axial-grooved gas-lubricated journal bearing-rotor system. 相似文献

Abstract The understanding of the flow processes in microchannels and micromixers is essential for the design of microfluidic devices like microreactors or analytical equipment. We have performed a systematic numerical CFD-study of mixing and mass transfer in sharp 90° bends and heat transfer in T-joints to obtain a detailed insight into the flow patterns and corresponding transfer processes in a wide range of Reynolds numbers. With increasing flow velocity, the straight laminar flow starts to form symmetrical vortices in the bend, at the entrance of the mixing channel, and in T-joints. The vortices enhance the transport processes like heat and mass transfer in the channels significantly. The influence of the geometry and the flow conditions is shown by an analytical estimation of the relevant forces. The appearance of convective transport processes is used for the definition of microflows, which are controlled by viscous forces and diffusive transfer processes. 相似文献

There are many knowledge-based data mining frameworks and it is common to think that new ones cannot come up with anything new. This article refutes such claims. We propose a sophisticated unification mechanism and two-tier machine cache system aimed at saving time and memory. No machine is run twice. Instead, machines are reused wherever they are repeatedly requested (regardless of request context). We also present an exceptional task spooler. Its unique design facilitates efficient automated management of large numbers of tasks with natural adjustment to available computational resources. Dedicated task scheduler cooperates with machine unification mechanism to save time and space. The solutions are possible thanks to very general and universal design of machine, configuration, machine context, unique machine life cycle, machine information exchange, configuration templates and other necessary concepts. Results gained by machines are stored in a uniform way, facilitating easy results exploration and collection by means of a special query system and versatile analysis with series transformations. No knowledge about internals of particular machines is necessary to extensively explore the results. The ideas presented here, have been implemented and verified inside Intemi framework for data mining and meta-learning tasks. They are general engine-level mechanisms that may be fruitful in all aspects of data analysis, all applications of knowledge-based data mining, computational intelligence, machine learning or neural networks methods. 相似文献
