By adding a gold core to silica nanoparticles (BrightSilica), silica‐like nanoparticles are generated that, unlike unmodified silica nanoparticles, provide three types of complementary information to investigate the silica nano‐biointeraction inside eukaryotic cells in situ. Firstly, organic molecules in proximity of and penetrating into the silica shell in live cells are monitored by surface‐enhanced Raman scattering (SERS). The SERS data show interaction of the hybrid silica particles with tyrosine, cysteine and phenylalanine side chains of adsorbed proteins. Composition of the biomolecular corona of BrightSilica nanoparticles differs in fibroblast and macrophage cells. Secondly, quantification of the BrightSilica nanoparticles using laser ablation inductively coupled plasma mass spectrometry (LA‐ICP‐MS) micromapping indicates a different interaction of silica nanoparticles compared to gold nanoparticles under the same experimental conditions. Thirdly, the metal cores allow the investigation of particle distribution and interaction in the cellular ultrastructure by cryo nanoscale X‐ray tomography (cryo‐XT). In 3D reconstructions the assumption is confirmed that BrightSilica nanoparticles enter cells by an endocytotic mechanism. The high SERS intensities are explained by the beneficial plasmonic properties due to agglomeration of BrightSilica. The results have implications for the development of multi‐modal qualitative and quantitative characterization in comparative nanotoxicology and bionanotechnology.  相似文献

Electron beam induced deposition (EBID) has recently been developed into a method to directly write optically active three-dimensional nanostructures. For this purpose a metal-organic precursor gas (here dimethyl-gold(III)-acetylacetonate) is introduced into the vacuum chamber of a scanning electron microscope where it is cracked by the focused electron beam. Upon cracking the aforementioned precursor gas, 3D deposits are realized, consisting of gold nanocrystals embedded in a carbonaceous matrix. The carbon content in the deposits hinders direct plasmonic applications. However, it is possible to activate the deposited nanostructures for plasmonics by coating the EBID structures with a continuous silver layer of a few nanometers thickness. Within this silver layer collective motions of the free electron gas can be excited. In this way, EBID structures with their intriguing precision at the nanoscale have been arranged in arrays of free-standing dimer antenna structures with nanometer sized gaps between the antennas that face each other with an angle of 90°. These dimer antenna ensembles can constitute a reproducibly manufacturable substrate for exploiting the surface enhanced Raman effect (SERS). The achieved SERS enhancement factors are of the order of 10? for the incident laser light polarized along the dimer axes. To prove the signal enhancement in a Raman experiment we used the dye methyl violet as a robust test molecule. In future applications the thickness of such a silver layer on the dimer antennas can easily be varied for tuning the plasmonic resonances of the SERS substrate to match the resonance structure of the analytes to be detected.  相似文献

The crystallographic textures and the resulting plastic anisotropy of five interstitial-free (IF) steels with different carbon equivalents and Nb and Ti microalloying content have been investigated. The steels were industrially hot rolled, cold rolled, annealed, and finally hot-dip galvanized. An alternative heat treatment of the cold rolled samples was conducted in laboratory scale using parameters close to those in industry-scale continuous annealing lines. The anisotropy parameters were both measured and predicted on the basis of the measured texture data. The calculated values were corrected by using functions that were fitted to the experimental data. It was found that for a given hot and cold rolling state even minor changes in the annealing conditions can improve the anisotropy parameter by up to 13 pct. Increase in recrystallization texture and improvement of the resulting anisotropy parameters are discussed in terms of partial transformation of ferrite to austenite in the intercritical regime.  相似文献

This paper considers a singularly perturbed reaction diffusion problem. It is investigated whether adaptive approaches are successful to design robust solution procedures. A key ingredient is the a posteriori error estimator. Since robust and mathematically analysed error estimation is possible in the energy norm, the focus is on this choice of norm and its implications. The numerical performance for several model problems confirms that the proposed adaptive algorithm (in conjunction with an energy norm error estimator) produces optimal results. Hence the energy norm is suitable for the purpose considered here. The investigations also provide valuable justification for forthcoming research. Generating a Condensed Representation for Association Rules   总被引:1,自引:0,他引:1  
Association rule extraction from operational datasets often produces several tens of thousands, and even millions, of association rules. Moreover, many of these rules are redundant and thus useless. Using a semantic based on the closure of the Galois connection, we define a condensed representation for association rules. This representation is characterized by frequent closed itemsets and their generators. It contains the non-redundant association rules having minimal antecedent and maximal consequent, called min-max association rules. We think that these rules are the most relevant since they are the most general non-redundant association rules. Furthermore, this representation is a basis, i.e., a generating set for all association rules, their supports and their confidences, and all of them can be retrieved needless accessing the data. We introduce algorithms for extracting this basis and for reconstructing all association rules. Results of experiments carried out on real datasets show the usefulness of this approach. In order to generate this basis when an algorithm for extracting frequent itemsets—such as Apriori for instance—is used, we also present an algorithm for deriving frequent closed itemsets and their generators from frequent itemsets without using the dataset.  相似文献

As part of the European Mercury Emissions from Chlor Alkali Plants (EMECAP) project, we tested the hypothesis that contamination of ambient air with mercury around chlor alkali plants using mercury cells would increase the internal dose of mercury in people living close to the plants. Mercury in urine (U-Hg) was determined in 225 individuals living near a Swedish or an Italian chlor alkali plant, and in 256 age- and sex-matched individuals from two reference areas. Other factors possibly affecting mercury exposure were examined. Emissions and concentrations of total gaseous mercury (TGM) around the plants were measured and modeled. No increase in U-Hg could be demonstrated in the populations living close to the plants. This was the case also when the comparison was restricted to subjects with no dental amalgam and low fish consumption. The emissions of mercury to air doubled the background level, but contributed only about 2 ng/m(3) to long-term averages in the residential areas. The median U-Hg levels in subjects with dental amalgam were 1.2 microg/g creatinine (micro/gC) in Italy and 0.6 microg/gC in Sweden. In individuals without dental amalgam, the medians were 0.9 microg/gC and 0.2 microg/gC, respectively. The number of amalgam fillings, as well as chewing, fish consumption, and female sex were associated with higher U-Hg. The difference between the countries is probably due to higher fish consumption in Italy, demethylated methyl mercury (MeHg) being partly excreted in urine. Post hoc power calculations showed that if the background mercury exposure is low it may be possible to demonstrate an increase in U-Hg of as little as about 10 ng/m(3) as a contribution to ambient mercury from a point source.  相似文献

Social resource sharing systems are central elements of the Web 2.0 and use the same kind of lightweight knowledge representation, called folksonomy. Their large user communities and ever-growing networks of user-generated content have made them an attractive object of investigation for researchers from different disciplines like Social Network Analysis, Data Mining, Information Retrieval or Knowledge Discovery. In this paper, we summarize and extend our work on different aspects of this branch of Web 2.0 research, demonstrated and evaluated within our own social bookmark and publication sharing system BibSonomy, which is currently among the three most popular systems of its kind. We structure this presentation along the different interaction phases of a user with our system, coupling the relevant research questions of each phase with the corresponding implementation issues. This approach reveals in a systematic fashion important aspects and results of the broad bandwidth of folksonomy research like capturing of emergent semantics, spam detection, ranking algorithms, analogies to search engine log data, personalized tag recommendations and information extraction techniques. We conclude that when integrating a real-life application like BibSonomy into research, certain constraints have to be considered; but in general, the tight interplay between our scientific work and the running system has made BibSonomy a valuable platform for demonstrating and evaluating Web 2.0 research.  相似文献
