Gymnosporia arenicola Jordaan (Celastraceae) is a shrub or small tree, which naturally occurs in coastal sand dunes of Southern Mozambique and South Africa. Its dried leaf is often used in traditional medicine for the treatment of infectious and inflammatory diseases. Hereby, we present results of studies carried out according to the pharmacopoeia standards for the identification of herbal drugs, in the whole, fragmented, and powdered plant material. These results were complemented with scanning electron microscopy and histochemical techniques. The leaf microscopic analysis revealed a typical dorsiventral mesophyll with a corresponding spongy parenchyma–palisade parenchyma ratio of 0.60, anomocytic and paracytic stomata, papillate cells with a diameter of 4.00 ± 0.40 µm, multicellular uniseriate nonglandular trichomes with a length of 27.00 ± 4.10 µm and cristalliferous idioblasts containing calcium oxalate cluster crystals with a diameter of 23.04 ± 5.84 µm. The present findings demonstrate that the G. arenicola leaf has both nonglandular trichomes and hypoderm, features not previously described in the corresponding botanical section (Gymnosporia sect. Buxifoliae Jordaan). The establishment of these new botanical markers for the identification of G. arenicola leaf is essential for quality, safety and efficacy reasons. Microsc. Res. The paper deals with a novel method of obtaining nanocarbon-conjugated polymer nanocomposites (NCPC) using nanocarbon colloids (NCC) and polyethylenimine (PEI) for water purification from metal ions. Size of NCC, process of NCPC synthesis, its chemical characteristics, ratio of NCC and PEI in NCPC, speed of coagulation of NCPC, mechanism of interaction of metal ions with NCPC, ability of removing metal ions from water by NCPC against pH have been studied. NCPC has a bonding capacity of 4.0-5.7 mmol/g at pH 6 for most of the divalent metal ions. Percent of sorption of Zn2+, Cd2+, Cu2+, Hg2+, Ni2+, Cr6+ ions is higher than 99%. Lifetime of NCPC before coagulation in the treated water is 1 s-1000 min and depends on the ratio of polymeric molecules and carbon nanoparticle concentrations. This paper describes a new method for precise determination of viral aerosol lethal dose (ALD50) and reports the results obtained for birds exposed by H5N1 Avian influenza A strain under controlled laboratory conditions. The reported method utilizes our recently developed personal bioaerosol sampler capable of monitoring viable virus concentration in the ambient air. Up to six laboratory animals could be located in a specially designed aerosol chamber and exposed in parallel by an airborne strain of interest to generate the amount of data sufficient for representative statistical analysis. A concentration of viable airborne virus was measured by the personal aerosol samplers directly in the breathing zone of each particular bird. The results show a very low inter-sampler variation used for each particular run, which was confirmed by a single factor ANOVA test undertaken for all six personal samplers involved in each experiment. As was shown, the difference in amount of viruses collected by all samplers during each particular run was not statistically significant. It was found that the ALD50 for approximately 400-g birds exposed to H5N1 Avian influenza A strain A/Chicken/Suzdalka/Nov-11/2005 was around 26.5 FFU (focus forming units).  相似文献

The preparation of microfibrillar composites (MFCs) based on oriented blends of polyamide 6 (PA6) and high‐density polyethylene (HDPE) is described. By means of conventional processing techniques, the PA6 phase was transformed in situ into fibrils with diameters in the upper nanometer range embedded in an isotropic HDPE matrix. Three different composite materials were prepared through the variation of the HDPE/PA6 ratio with and without a compatibilizer: MFCs reinforced by long PA6 fibrils arranged as a unidirectional ply; MFCs containing middle‐length, randomly distributed reinforcing PA6 bristles; and a nonoriented PA6‐reinforced material in which the PA6 phase was globular. The evolution of the morphology in the reinforcing phase (e.g., its visible diameter, length, and aspect ratio) was followed during the various processing stages as a function of the blend composition by means of scanning electron microscopy. Synchrotron X‐ray scattering was used to characterize selected unidirectional ply composites. The presence of transcrystalline HDPE was demonstrated in the shell of the reinforcing PA6 fibrils of the final MFCs. The impact of the compatibilizer content on the average diameter and length of the fibrils was assessed. The influence of periodic operation on trickle-bed reactor (TBR) hydrodynamics and gas–liquid mass transfer was investigated. Two-phase pressure drop, dynamic liquid hold-up and gas–liquid mass transfer coefficient (kLa) were determined at various liquid flow rates and for different modes of liquid flow variation (increasing and decreasing liquid flow rate). The results reveal the considerable influence of type of liquid flow rate modulation on kLa values (deviations of up to 80% in kLa). Simulation studies on gas-limited reaction in a periodically operated TBR indicate that an enhancement in conversion of about 14% can be expected from an appropriate selection of the operating mode, thus clearly demonstrating the quantitative process intensification feasible through increased gas–liquid mass transfer.  相似文献

We compared thermally induced denaturation and aggregation of two isoforms of the isolated myosin head (myosin subfragment 1, S1) containing different "essential" (or "alkali") light chains, A1 or A2. We applied differential scanning calorimetry (DSC) to investigate the domain structure of these two S1 isoforms. For this purpose, a special calorimetric approach was developed to analyze the DSC profiles of irreversibly denaturing multidomain proteins. Using this approach, we revealed two calorimetric domains in the S1 molecule, the more thermostable domain denaturing in two steps. Comparing the DSC data with temperature dependences of intrinsic fluorescence parameters and S1 ATPase inactivation, we have identified these two calorimetric domains as motor domain and regulatory domain of the myosin head, the motor domain being more thermostable. Some difference between the two S1 isoforms was only revealed by DSC in thermal denaturation of the regulatory domain. We also applied dynamic light scattering (DLS) to analyze the aggregation of S1 isoforms induced by their thermal denaturation. We have found no appreciable difference between these S1 isoforms in their aggregation properties under ionic strength conditions close to those in the muscle fiber (in the presence of 100 mM KCl). Under these conditions kinetics of this process was independent of protein concentration, and the aggregation rate was limited by irreversible denaturation of the S1 motor domain.  相似文献

Manganese oxides of various stoichiometry were prepared via Mn-oxalate precipitation followed by thermal decomposition in the presence of oxygen. A non-stoichiometric manganese oxide, MnO x (x = 1.61…1.67) was obtained by annealing at 633 K and demonstrated superior CO oxidation activity, i.e. full CO conversion at room temperature and below. The activity gradually decreased with time-on-stream of the reactants but could be easily recovered by heating at 633 K in the presence of oxygen. CO oxidation over MnO x in the absence of oxygen proved to be possible with reduced rates and demonstrated a Mars—van Krevelen—type mechanism to be in operation. A TEM structural analysis showed the MnO x phase to form microrods with large aspect ratio which broke up into nanocrystalline manganese oxide (MnO x ) particles with diameters below 3 nm and a BET specific surface area of 525 m2/g. Annealing at 798 K rather than 633 K produced well crystalline Mn2O3 which showed lower CO oxidation activity, i.e. 100% CO conversion at 335 K. The catalytic performance in CO oxidation of various Mn-oxides either studied in this work or elsewhere was compared on the basis of specific reaction rates.  相似文献

Occurrence and prevalence of different bacterial enteric pathogens as well as their relationships with conventional (total and fecal coliforms) and alternative fecal indicators (host-specific Bacteroides 16S rRNA genetic markers) were investigated for various water samples taken from different sites with different degrees of fecal contamination. The results showed that a wide range of bacterial pathogens could be detected in both municipal wastewater treatment plant samples and in surface water samples. Logistic regression analysis revealed that total and human-specific Bacteroides 16S rRNA genetic markers showed significant predictive values for the presence of Escheriachia coli O-157, Salmonella, heat-labile enterotoxin (LT) of enterotoxigenic E. coli (ETEC), and heat-stable enterotoxin for human (STh) of ETEC. The probability of occurrence of these pathogenic bacteria became significantly high when the concentrations of human-specific and total Bacteroides 16S rRNA genetic markers exceeded 10(3) and 10(4) copies/100 mL. In contrast, Clostridium perfringens was detected at high frequency regardless of sampling sites and levels of Bacteroides 16S rRNA genetic markers. No genes related to Shigella spp., Staphylococcus aureus and Vibrio cholerae were detected in any samples analyzed in this study. Conventional indicator microorganisms had low levels of correlation with the presence of pathogens as compared with the alternative fecal indicators. These results suggested that real-time PCR-based measurement of alternative Bacteroides 16S rRNA genetic markers was a rapid and sensitive tool to identify host-specific fecal pollution and probably associated bacterial pathogens. However, since one fecal indicator might not represent the relative abundance of all pathogenic bacteria, viruses and protozoa, combined application of alternative indicators with conventional ones could provide more comprehensive pictures of fecal contamination, its source and association with pathogenic microorganisms.  相似文献

Consumption of unpasteurized melon and watermelon juices has caused several disease outbreaks by pathogenic microorganisms worldwide. Pulsed electric field (PEF) has been recognized as a technology that may inactivate those bacteria present in fluid food products at low temperatures. Hence, PEF treatment at 35 kV/cm, 4 mus pulse duration in bipolar mode and square shape were applied on Salmonella Enteritidis, E. coli and L. monocytogenes populations inoculated in melon and watermelon juices without exceeding 40 degrees C outlet temperatures. Authenticity and traceability have been two of the most important issues in the food chain. Authenticity in particular, is closely related with both food quality and safety issues. Vegetables stand for a category of foods heavily affected by adulteration either in terms of geographic origin (national or international level) or production methods (organic or conventional production, fertilizers, pesticides, genetically modified vegetables). This review aims at addressing most of the currently applied methods for ensuring quality control of vegetables; a) instrumental: ion chromatography, high pressure liquid chromatography, atomic absorption spectrophotometry, electronic nose and mass spectroscopy and b) sensory analysis. The results of all the above mentioned methods were analyzed by means of multivariate analysis (principal component analysis, discriminant analysis, cluster analysis, canonical analysis, and factor analysis). All ensuing results and conclusions are summarized in eight comprehensive tables.  相似文献
