T cells express CD28 and CD27 which transduce co-stimulatory signals after interaction with their ligands on antigen-presenting cells (APC). These ligands, CD80, CD86 and CD70, are also expressed to some extent on activated T cells. Here, we show that in human immunodeficiency virus (HIV)-infected individuals, CD28 and CD27 expression is decreased on CD8+ T cells. On the other hand, T cell stimulation in vitro induced high CD80, CD86 and CD70 expression on T cells from HIV-infected individuals. It appeared that an inverted CD4:CD8 T cell ratio could explain this enhanced expression of co-stimulatory ligands. Indeed, high expression levels of CD80, CD86 and CD70 were found on activated CD8+ T cells from HIV- individuals cultured in the absence of CD4+ T cells. Addition of CD4+ T cells prevented this up-regulation. However, in HIV-infected individuals, addition of excess autologous or healthy control CD4+ T cells did not completely counteract up-regulation of co-stimulatory ligand expression on CD8+ T cells. Thus, to some extent, CD8+ T cells in HIV-infected individuals appeared to be refractory to CD4+ T cell-mediated regulation of ligand expression in vitro. Activated T cells from HIV-infected individuals and activated CD8+ T cells from healthy controls were able to act as accessory cells in CD3-induced T cell proliferation, which was dependent on cell-cell contact. Thus, we showed that T cells from HIV-infected individuals express enhanced levels of co-stimulatory ligands upon activation, which provides them with accessory cell properties. Enhanced stimulatory potential of these nonprofessional APC may contribute to persistently high levels of immune activation in HIV infection related to disease progression.  相似文献

Sustained attention was assessed in 20 patients with severe closed head injury (CHI) and 20 normal matched controls. Participants were presented with a visual continuous performance task (CPT) with 3 levels of complexity. Performance was assessed by examining response latencies and error rates. Across all levels of complexity, the CHI patients demonstrated a vigilance decrement, whereas the performance of the matched controls was stable across time. The vigilance decrement was not differentially affected by the manipulation of task complexity in the CHI patients. However, findings suggest that the overall vigilance performance of CHI patients was differentially affected by increasing the complexity of the task. This review focuses on some selected aspects of the endocrine heart and natriuretic peptides. The endocrine heart is composed of specific myoendocrine cells of the cardiac atria. The myoendocrine cells synthesize and secrete the natriuretic peptide hormones which exhibit natriuretic, diuretic, and vasorelaxant properties. Immunohistochemical analyses show that natriuretic peptides of the A-type and B-type are localized not only in the specific granules of these myoendocrine cells but also in many other organs including the brain, adrenal medulla, and kidney. Also, their receptors are detected in many organs showing the multiple functions of these regulatory peptides. Of the members of the natriuretic peptide family, ANP (ANP for atrial natriuretic peptide; also denominated cardiodilatin, CDD), brain natriuretic peptide (BNP), C-type natriuretic peptide (CNP), and the A-type, including its renal form, urodilatin, are emphasized in this review. Urodilatin is localized in the kidney, differentially processed, and secreted into the urine. The intrarenal synthesis and secretion is the basis for a paracrine system regulating water and sodium reabsorption at the level of the collecting duct. CDD/ANP-1-126, cleaved from a precursor of 126 amino acids in the heart to a 28-amino acid-containing circulating molecular form (CDD/ANP-99-126), and urodilatin (CDD/ANP-95-126) share similar biochemical features and biological functions, but urodilatin may be more involved in the regulation of body fluid volume and water-electrolyte excretion, while circulating CDD/ANP-99-126 is responsible for blood pressure regulation. The physiological and pharmacological properties of these peptides have great clinical impact, and as a consequence urodilatin is involved in drug development for the treatment of acute renal failure, cardiomyopathia, and acute asthma.  相似文献

Oncostatin M (OSM), a member of the interleukin-6 family, functions as a major mediator of cardiomyocyte remodeling under pathological conditions. Its involvement in a variety of human cardiac diseases such as aortic stenosis, myocardial infarction, myocarditis, cardiac sarcoidosis, and various cardiomyopathies make the OSM receptor (OSMR) signaling cascades a promising therapeutic target. However, the development of pharmacological treatment strategies is highly challenging for many reasons. In mouse models of heart disease, OSM elicits opposing effects via activation of the type II receptor complex (OSMR/gp130). Short-term activation of OSMR/gp130 protects the heart after acute injury, whereas chronic activation promotes the development of heart failure. Furthermore, OSM has the ability to integrate signals from unrelated receptors that enhance fetal remodeling (dedifferentiation) of adult cardiomyocytes. Because OSM strongly stimulates the production and secretion of extracellular proteins, it is likely to exert systemic effects, which in turn, could influence cardiac remodeling. Compared with the mouse, the complexity of OSM signaling is even greater in humans because this cytokine also activates the type I leukemia inhibitory factor receptor complex (LIFR/gp130). In this article, we provide an overview of OSM-induced cardiomyocyte remodeling and discuss the consequences of OSMR/gp130 and LIFR/gp130 activation under acute and chronic conditions.  相似文献

The adsorption of nitrogen oxides on Cu-ZSM-5 was studied by infrared spectroscopy to elucidate the species associated with the band at 2133 cm–1. The band was found for both NO and NO2 adsorption. Labeling experiments with15NO revealed that the associated surface species contained nitrogen and, most likely, an N-O bond. Co-adsorption experiments of NO and oxygen produced adsorbed nitronium, NO 2 + , as the principal, associated species. Adsorption of nitrogen oxides on dispersed CuO and the HZSM-5 support demonstrated that the 2133 cm–1 band was not necessarily associated with copper ions. A relatively strong correlation between the bands at 2133 and 3615 cm–1 indicates that the primary adsorption sites of NO 2 + are the strongly protic, bridging Si(OH)Al framework hydroxyls. Once these were filled, other, weaker acid sites began to adsorb NO 2 O .  相似文献

In rapeseed meals (RSM) of a conventional (c) or a newly bred (n) variety 117 or 44 mmol glucosinolates and glucosinolate degradation products per kg dry matter were detected. Soaking meals in aqueous myrosinase or copper sulphate solution and subsequent drying reduced the content of antinutrients by more than 90%. Broiler chickens were fed (i) a glucosinolate-free diet, or this diet in which half the soybean meal was replaced by RSM of various origins and treatment; (ii) untreated cRSM; (iii) cRSM treated with myrosinase: (iv) cRSM treated with Cu (v) untreated nRSM; or (vi) nRSM treated with myrosinase. These diets were administered with or without supplementary iodine. Chickens receiving the iodine-deficient diets with myrosinase-treated RSM showed growth depression, incomplete feathering, leg injuries and severe goitre. In the serum T4 could not be detected. Giving myrosinase-treated RSM plus iodine, or giving other RSM diets irrespective of iodine administration, no growth depression was observed. RSM diets without iodine dosage markedly increased thyroid weight: there were no differences between the RSM variants. In contrast to the treatment with myrosinase, in the sera of the chickens fed on untreated RSM or RSM treated with copper T4 could be detected, suggesting that in iodine-deficient conditions differences in serum T4 concentration between RSM groups indicate a differing anti-thyroid activity. With iodine supplementation the RSM had a significant effect on thyroid weight. The largest thyroids (five-fold heavier) were in the animals with myrosinase-treated cRSM. The untreated cRSM and the nRSM trebled or doubled the thyroid weight, and the myrosinase-treated nRSM trebled it. The thyroid weight of chicks fed cRSM treated with copper did not differ significantly from the glucosinolate-free control. There was evidence that heating the myrosinase-treated RMS produced anti-thyroid compounds; these should be identified in further investigations.  相似文献

The solubility of methane in 3 kmol/m3 solutions of monoethanolamine, diethanolamine, and triethanolamine was measured from 25° to 125°C and pressures up to about 13 MPa. Measurements were also made for the solubility of methane in water at 25° to 125°C and pressures up to 18 MPa in order to confirm the accuracy of the experimental technique. It is demonstrated that methane is more soluble (in terms of mole fraction) in the amine solution than in pure water. Furthermore, the solubility is an increasing function of the size of the alkanolamine. The solubility data were modeled using a Henry's-law approach and the results summarized in terms of salting-in coefficients.  相似文献

A detailed analysis has been carried out on the deasphaltened Cold Lake bitumen using two different series of chromatographic separations followed by i.r., u.v., n.m.r. and computerized GC/MS studies of the separated fractions: saturates (21.3), monoaromatics (8.3), diaromatics (3.6), polyaromatics and non-specific polar compounds (24.35), acids (15.2), bases (6.38), and neutral nitrogen compounds (1.15% of bitumen). The acyclic paraffin content of the maltene is low but decidedly higher than in the related Athabasca bitumen, indicating that the Cold Lake bitumen suffered a less complete biodegradation. Straight-chain paraffins and the isoprenoids phytane and pristane are present. The mono- and diaromatic fractions were analysed by high-voltage, low-resolution mass spectrometry. The monoaromatic fraction contains alkylbenzenes, naphthenebenzenes and dinaphthenebenzenes in a ratio of approximately 1:1:1. The ratio for naphthalenes, acenaphthenes + dibenzofurans and fluorenes in the diaromatic fraction is approximately 1:0.9:0.6. The composition of Cold Lake bitumen closely resembles that of the Athabasca bitumen except that its asphaltene content is somewhat lower and its acidic and saturate content is slightly higher.  相似文献

Modern water-borne paints are widely used in different areas of applications ranging from high-gloss lacquers to flat, scrub-resistant interior paints. From this point of view, the pigment volume concentration (PVC) is one key-parameter adjusting the desired application properties. In high-gloss paints, for example, a low PVC is required to accommodate the proper surface roughness to achieve a high gloss. Consequently, a high concentration of TiO2 is needed to obtain a good hiding power at the same time. Flat paints nonetheless are highly filled due to cost reasons preferentially by CaCO3 and the pigment binding capacity of the binder is crucial. In this work, paint formulations differing in PVC, and the type of binder or dispersing agent were investigated by various techniques concerning the distribution and aggregation of pigment particles, e.g. TiO2. To get a detailed insight into the structure of the liquid paints and the corresponding dried paint films, suitable analytical tools were applied for characterization. The structure of the liquid paints was analyzed by remission light spectroscopy (RLS), disc centrifugation, cryogenic-replication transmission electron microscopy (Cryo-replica TEM) and cryogenic-scanning electron microscopy (Cryo-SEM). The pigment distribution in the corresponding dried paint films was examined by means of atomic force microscopy (AFM), TEM and RLS. The tendency of the TiO2-pigments to form aggregates was found to depend on both: first on the type of binder used in the formulation and second on the employed dispersing agent. It is shown that only by adjusting the properties of the binder in combination with common dispersants, it is possible to get well-distributed TiO2 particles within the paint. Correlation of application properties, e.g. gloss and blocking to the microscopic structure is presented.  相似文献
