A direct, rapid, and easy electrospray ionization mass spectrometry (ESI-MS) method to determine concentrations of sebacic acid (SA) and terephthalic acid (TA) residues in biodegradable copolymers was developed. Copolyester samples were synthesized from 1,4-butanediol and sebacic and terephthalic acids by melt polymerization. Extraction of monomers was performed in methanol. Their concentrations were determined by direct infusion ESI-MS, without chromatographic separation, using 1,12-dodecanedioic acid (DDA) as an internal standard. Calibration curves were obtained by plotting the ratio of the areas of the peaks relative to monomers and DDA standard as a function of their concentration ratio. We validated the method by determining the concentration of TA residue using both the ESI-MS protocol and high-performance liquid chromatography (HPLC) analysis with UV detection. The linearity range and the detection limit of this assay were 0.1-5.0 and 0.01 ppm for SA and 0.1-6.0 and 0.03 ppm for TA. This assay represents a useful alternative to conventional methods currently employed for acid quantification, resulting advantageous for its speed and high sensitivity. 相似文献

Single layers of dielectric spheres are an interesting system to study from the fundamental and applied points of view. In this paper we present a systematic study of the influence of structural disorder on the transmission spectra of arrangements of spheres of different compactness. Glass sphere (ε=7) planes were built and their transmission spectra in the microwave range measured. Transmission behavior of this system is highly tolerant to disorder. Even in completely disordered arrangements, there is a highly rejected band with the dips of the spectrum observable. These results suggest that the collective modes of the sphere planes are formed by weakly coupled Mie modes of the individual spheres, and this coupling is governed by the average distance among the spheres. Disorder tolerance allows simpler fabrication procedures where the position of the spheres does not need to be precisely controlled. 相似文献

The use of polymers naturally occurring in the extracellular matrix (ECM) is a promising strategy in regenerative medicine. If compared to natural ECM proteins, proteins obtained by recombinant DNA technology have intrinsic advantages including reproducible macromolecular composition, sequence and molecular mass, and overcoming the potential pathogens transmission related to polymers of animal origin. Among ECM-mimicking materials, the family of recombinant elastin-like polymers is proposed for drug delivery applications and for the repair of damaged elastic tissues. This work aims to evaluate the potentiality of a recombinant human elastin-like polypeptide (HELP) as a base material of cross-linked matrices for regenerative medicine. The cross-linking of HELP was accomplished by the insertion of cross-linking sites, glutamine and lysine, in the recombinant polymer and generating ε-(γ-glutamyl) lysine links through the enzyme transglutaminase. The cross-linking efficacy was estimated by infrared spectroscopy. Freeze-dried cross-linked matrices showed swelling ratios in deionized water (≈2500%) with good structural stability up to 24 h. Mechanical compression tests, performed at 37°C in wet conditions, in a frequency sweep mode, indicated a storage modulus of 2/3 kPa, with no significant changes when increasing number of cycles or frequency. These results demonstrate the possibility to obtain mechanically resistant hydrogels via enzymatic crosslinking of HELP. Cytotoxicity tests of cross-linked HELP were performed with human umbilical vein endothelial cells, by use of transwell filter chambers for 1-7 days, or with its extracts in the opportune culture medium for 24 h. In both cases no cytotoxic effects were observed in comparison with the control cultures. On the whole, the results suggest the potentiality of this genetically engineered HELP for regenerative medicine applications, particularly for vascular tissue regeneration. 相似文献

Influence of Fe3+ /Fe2+ Ratio on the Crystallization of Iron-Rich Glasses Made with Industrial Wastes

The influence of the Fe3+ /Fe2+ ratio on the crystallization of iron-rich glasses was investigated in this study. The glass batches were made from two hazardous industrial wastes: mud (goethite and jarosite) originating from the zinc hydrometallurgical process and electric arc furnace dust (EAFD). Glass compositions were prepared by adding different percentages of carbon powder. The crystallization process was investigated by a combined thermogravimetry/differential thermal analysis technique, in air or nitrogen atmospheres, using powder and bulk glass samples. The crystalline phases formed, i.e., pyroxene and spinels, and their relative ratio were determined by X-ray diffractometry. The experimental results indicated that melting temperature and crystallization behavior were influenced by the initial Fe3+ /Fe2+ ratio and by the amount of carbon added to the glass batch. For goethite and jarosite glass compositions, decreasing the Fe3+ /Fe2+ ratio increased the crystallization rate by favoring magnetite formation. For EAFD glass compositions, the addition of carbon to the batch inhibited chromite–magnetite spinel formation and favored the attainment of an amorphous glassy phase. 相似文献

The GGP1/GAS1/CWH52 gene of Saccharomyces cerevisiae encodes a major exocellular 115 kDa glycoprotein (gp115) anchored to the plasma membrane through a glycosylphosphatidylinositol (GPI). The function of gp115 is still unknown but the analysis of null mutants suggests a possible role in the control of morphogenesis. PHR1 gene isolated from Candida alibicans is homologous to the GGP1 gene. In this report we have analysed the ability of PHR1 to complement a ggp1Δ mutation in S. cerevisiae. The expression of PHR1 controlled by its natural promoter or by the GGP1 promoter has been studied. In both cases we have observed a complete complementation of the mutant phenotype. Moreover, immunological analysis has revealed that PHR1 in budding yeast gives rise to a 75–80 kDa protein anchored to the membrane through a GPI, indicating that the signal for GPI attachment present in the C. albicans gene product is functional in S. cerevisiae. 相似文献

Two experiments were conducted to evaluate the proximal composition, lipids and cholesterol content of meat from pigs fed diets with peach-palm meal (PPM), with or without addition of synthetic lysine (LYS). In experiment 1, 24 pigs were randomly allotted into six treatments with three levels of PPM (0.16 and 32%) and two levels of LYS (0 and 0.27%). In experiment II, 16 finishing pigs were fed with two levels of PPM (0 and 17.50%) and two levels of LYS (0 and 0.27%). At the end of each experiment (42 and 35 d, respectively), pigs were slaughtered and loin samples were obtained to determine crude protein, dry matter, moisture, ash, total lipids, and cholesterol content. In experiment I, pork loin from 16% PPM had more dry matter (26.45 g/100 g) and less moisture (73.49 g/100g) than pork loin from 32% PPM (25.11 y 75.03 g/100g, respectively). Meat samples from pigs without LYS had higher (p < 0.05) content of lipids (2.11 g/100 g) than meat from pigs that consumed LYS (1.72 g/100 g). In experiment II, the proximal, lipids and cholesterol content were similar among treatments. The PPM addition to pig diets did not affect the proximal composition of pork, while LYS addition indicated a reduction of total lipids, which could result as an alternative to obtain leaner meat. 相似文献

Basal cell carcinoma (BCC) is the most common cancer in individuals with fair skin type (I–II) and steadily increasing in incidence (70% of skin malignancy). It is locally invasive but metastasis is usually very rare, with an estimated incidence of 0.0028%–0.55%. Conventional therapy is surgery, especially for the H region of the face and infiltrative lesions; in case of inoperable tumors, radiotherapy is a valid option. Recently, topical photodynamic therapy (PDT) has become an effective treatment in the management of superficial and small nodular BCC. PDT is a minimally invasive procedure that involves the administration of a photo-sensibilizing agent followed by irradiation at a pre-defined wavelength; this determines the creation of reactive oxygen species that specifically destroy target cells. The only major side effect is pain, reported by some patients during the irradiation. The high cure rate and excellent cosmetic outcome requires considering this possibility for the management of patients with both sporadic and hereditary BCC. In this article, an extensive review of the recent literature was made, in order to clarify the role of PDT as a possible alternative therapeutic option in the treatment of BCC. 相似文献

Thermotropic main-chain liquid crystalline polymers typically have very low melt viscosity with strong temperature dependence compared to other common thermoplastics. While this is beneficial in some processing applications, such as injection molding, it presents challenges for others, such as coextrusion. In this study, the rheological properties of a thermotropic main-chain liquid crystalline polymer (Vectra A950) were enhanced by melt-state reactive processing with triphenyl phosphite (TPP), which can react with up to three polymer chain-ends through their chain-end functionalities. The influence of processing time and TPP content on the shear viscosity and other important material properties were investigated. Optimal conditions, which increased the shear viscosity by nearly a factor of 20 over the neat polymer, were found to be 4 wt% TPP and 30 min of reaction time at 290 °C. Further results from differential scanning calorimetry, wide-angle X-ray diffraction and polarized optical microscopy confirmed that coupling with TPP did not affect the microstructure, melting/crystallization behavior or liquid crystallinity. The stability of TPP-modified samples was also studied at 80 °C in air and following melt reprocessing at 290–300 °C under N2 or air. Samples were stable (as measured by shear viscosity) for more than one month at 80 °C in air or when reprocessed in N2 at 290 °C for up to 10 min. However, when reprocessed at 300 °C in air, the viscosity enhancement was partially reversed due to scission of P–O bonds that were formed during the initial reaction between the polymer chain-ends and TPP. 相似文献

This article presents a new methodology to potentially quantify polycyclic aromatic hydrocarbon (PAH) isomers using high-resolution time of flight aerosol mass spectrometer (HR-AMS). The fragmentation of PAHs within the HR-AMS is such that significant signal remains at the molecular ion. After quantifying the molecular ion signal and taking into account potential interferences, the amount of the parent PAH in the aerosol may be inferred once its fragmentation pattern is also known. The potential of this approach was evaluated using mixed gasoline and diesel engine exhaust sampled under varying conditions. This dataset led to the identification and quantification within the aerosol mass spectra of the molecular ions associated with 53 PAH isomers, including both unsubstituted and functionalized species. An evaluation of anticipated interferences shows that interferences from larger molecular weight PAHs (i.e., PAH/PAH interferences) could be constrained based on the fragmentation behavior of PAHs from existing HR-AMS laboratory spectra. Other signal interferences for this data set are typically less than 5% of the total signal or, for 13C isotopic interferents, are well constrained by measurements of the dominant isotope. The experimental data reveal that the fractional PAH molecular ion signal remained stable despite dramatic temporal variability of the total particulate organic signal. The fractional contributions of the molecular ions for grouped PAH species and even individual compounds were remarkably consistent across experiments. The distribution of PAHs showed no apparent dependence on engine load or exhaust type. Full application of this approach will require a greater number of standard HR-AMS spectra for PAHs, so that the relationship between compounds and their molecular ions may be understood more precisely.Copyright 2015 American Association for Aerosol Research 相似文献
