The copolymers poly(propylene-co-ethylene) (PP/E) and poly(ethylene-co-vinyl acetate) (EVA) and blends of these were modified to develop an artificial matrix which promotes the growth of endothelial cells. Covalent immobilization of amino acids or sequences of adhesion glycoproteins should trigger the formation of an endothelial cell monolayer onto the polymeric surface. Reactive functional groups were generated by saponifying the ester groups of the EVA component. Esterification with oxalylic or malonic dichlorides in the gas phase yielded the required monoesters and gave the best results for further immobilization of amino acids, while reaction with α,ω-dicarboxylic acid dichlorides in solution led to diester formation. Subsequently, various protected amino acids were immobilized via the carbodiimide method. Surface analytical methods like infrared spectroscopy using at tenuated total reflection (IR-ATR), X-ray photoelectron spectroscopy (XPS), and secondary ion mass spectrometry (SIMS) were used to prove the modification steps. The analytical results confirmed covalent side-chain generation in the upper surface region. © 1995 John Wiley & Sons, Inc.  相似文献

To discover genes contributing to mental retardation in 3p- syndrome patients we have used in silico searches for neural genes in NCBI databases (dbEST and Uni-Gene). An EST with strong homology to the rat CAM L1 gene subsequently mapped to 3p26 was used to isolate a full-length cDNA. Molecular analysis of this cDNA, referred to as CALL (cell adhesion L1-like), showed that it is encoded by a chromosome 3p26 locus and is a novel member of the L1 gene family of neural cell adhesion molecules. Multiple lines of evidence suggest CALL is likely the human ortholog of the murine gene CHL1: it is 84% identical on the protein level, has the same domain structure, same membrane topology, and a similar expression pattern. The orthology of CALL and CHL1 was confirmed by phylogenetic analysis. By in situ hybridization, CALL is shown to be expressed regionally in a timely fashion in the central nervous system, spinal cord, and peripheral nervous system during rat development. Northern analysis and EST representation reveal that it is expressed in the brain and also outside the nervous system in some adult human tissues and tumor cell lines. The cytoplasmic domain of CALL is conserved among other members of the L1 subfamily and features sequence motifs that may involve CALL in signal transduction pathways.  相似文献

Regeneration of descending spinal cord tracts occur spontaneously in adult goldfish. Very little information is available regarding the fate of ascending fibers. Using Dextran amines as a tracer, we studied the normal and regenerated ascending axonal projection patterns in adult goldfish brain nuclei. Present study includes spinal projections to torus semicircularis, hypothalamus, thalamus and the telencephalon. Regenerated fibers had finer caliber axons and the terminal axonal arbors covered a larger area than the corresponding normal ones.  相似文献

This study suggests a macroheterogeneity in prevalence of platelet glycoprotein PL(A1/A2) polymorphism in different ethnic populations. In patients undergoing percutaneous transluminal coronary angioplasty, this polymorphism does not represent an independent risk factor but seems to be implicated in restenosis after percutaneous transluminal coronary angioplasty.  相似文献

The effects of physiologic alterations in endogenous angiotensin II activity on basal renal sympathetic nerve activity and its cardiac baroreflex regulation were studied. The effect of angiotensin II type 1 receptor blockade with intracerebroventricular losartan was examined in conscious rats consuming a low, normal, or high sodium diet that were instrumented for the simultaneous measurement of right atrial pressure and renal sympathetic nerve activity. The gain of cardiac baroreflex regulation of renal sympathetic nerve activity (% delta renal sympathetic nerve activity/mmHg mean right atrial pressure) was measured during isotonic saline volume loading. Intracerebroventricular losartan did not decrease arterial pressure but significantly decreased renal sympathetic nerve activity in low (-36+/-6%) and normal (-24+/-5%), but not in high (-2+/-3%) sodium diet rats. Compared with vehicle treatment, losartan treatment significantly increased cardiac baroreflex gain in low (-3.45+/-0.20 versus -2.89+/-0.17) and normal (-2.89+/-0.18 versus -2.54+/-0.14), but not in high (-2.27+/-0.15 versus -2.22+/-0.14) sodium diet rats. These results indicate that physiologic alterations in endogenous angiotensin II activity tonically influence basal levels of renal sympathetic nerve activity and its cardiac baroreflex regulation.  相似文献

DRESS/DiHS is a complex and potentially fatal drug reaction. Little is known about risk factors and elements that can help to identify patients with a severe reaction early. The aim of the study was to investigate those factors favoring the disease and its severity by analyzing the clinical conditions and therapies preceding the reaction. We conducted a retrospective analysis on patients admitted to our center between 2010 and 2020 who were discharged with a diagnosis of DRESS. We used the RegiSCAR diagnostic criteria. We defined the severity of DRESS using the criteria of Mizukawa et al. We included 25 patients (15 females) with a median age of 66 years. Skin involvement, eosinophilia, and liver injury were the most important aspects. Allopurinol was found to be the most involved drug. Reaction severity was significantly associated with the number of daily medications (p = 0.0067) and an age of at least 68 years (p = 0.013). In addition, 75% of severe cases had at least three comorbidities in history, and most of the severe cases were female. In our study the advanced age, the high number of comorbidities and home therapies, and the inflammatory state were found to be predisposing elements to the development of the disease and its severity.  相似文献

Electrospinning is a versatile technique to generate tissue engineering matrices possessing structural features similar to the extracellular matrix. Biodegradable polylactides are well suited for processing by this technique, but their innate hydrophobicity impairs initial protein adsorption and cell adhesion. In this work, therefore, electrospun poly(L ‐lactide‐co‐D,L ‐lactide) (70/30) non‐wovens are modified with an ultrathin plasma‐polymerized allylamine (PPAAm) coating. Using scanning electron microsocopy (SEM), it is shown that the fiber structure of the non‐woven is not affected by the plasma treatment. X‐ray photoelectron spectroscopy (XPS) and contact angle measurements of PPAAm‐coated non‐wovens confirm the presence of nitrogen and oxygen‐functional groups in the coating and a hydrophilic nature of the coated non‐woven surface. Cell experiments in vitro demonstrate that the PPAAm‐coated surface promotes occupancy of the non‐woven by human MG‐63 osteoblasts accompanied by improved initial cell spreading and filopodia formation along and between the electrospun polylactide fibers. Overall, plasma‐assisted incorporation of amino groups into electrospun polylactone non‐wovens represents a promising approach to tissue engineering scaffolds with improved cell–material interfaces.  相似文献

We report a nanoparticulate system capable of targeting the heart after myocardial infarction (MI). Targeting is based on overexpression of angiotensin II type 1 (AT1) receptor in the infarcted heart. Liposomes 142 nm in diameter were conjugated with a ligand specific to AT1. The nanoparticles were able to specifically target cardiac cells in vitro, and in the infarcted heart after intravenous injection in vivo. This system may be useful for delivering therapeutic agents specifically to the infarcted heart.  相似文献

A dynamical model to calculate the performance of an absorber roof-assisted heat pump is developed. The model is applied to a specific example, a normal residence building in Berlin. The heat pump is layed out to meet the heat demand at 0°C outside temperature. The auxiliary heat is provided by a conventional backup system. Conditions of insolation and outdoor temperature are derived from real weather data and simulated in their dynamical behaviour. The analysis investigates how the variation of the storage tank volume and the lower limiting COP is reflected in the annual cost savings.  相似文献
