Drug-resistance markers for yeast transformation are useful because they can be applied to strains without auxotrophic mutations. However, they are susceptible to technical difficulties, namely lower transformation efficiency and the appearance of drug-resistant mutants without the marker. To avoid these problems, we have constructed a phosphoglycerate kinase (PGK) promoter-driven YAP1 expression cassette, called PGKp-YAP1. Yeast cells containing PGKp-YAP1 were resistant to cycloheximide, a protein synthesis inhibitor, and also to cerulenin, a fatty acid synthesis inhibitor, but not to other drugs tested. The transformation efficiency of PGKp-YAP1 using cerulenin selection was comparable to that using a URA3 auxotrophic marker when low concentrations of cerulenin were used. Non-transformed drug-resistant colonies did appear on the low-concentration cerulenin plates. However, these non-transformed colonies could easily be identified, based on their cycloheximide sensitivity and/or their resistance to aureobasidin A to which the transformants were sensitive. Therefore, the dual drug resistance of PGKp-YAP1 could be used as an effective selection for PGKp-YAP1 recipient cells. The PGKp-YAP1 marker was used to disrupt the LYS2 gene and to transform an industrial yeast strain, indicating that this marker can be used for efficient and reliable gene manipulations in any Saccharomyces cerevisiae strain.  相似文献

Heat capacities of a submonolayer 3He solid film adsorbed on a graphite surface are measured in magnetic fields up to 400 Oe. The measured heat capacity shifts to higher temperatures with an increasing magnetic field. The amplitudes of the shifts are twenty times larger than the magnitude of the Zeeman energy, which is anomalously large. With regard to the origin of these large shifts, the reduction of frustrations and the weakening of the competition between multiple spin exchange interactions are discussed.  相似文献

Expansion and stress relaxation of expressed cake are discussed.

The expansion process of expressed cake after release of load is very similar to consolidation of solidndashliquid mixtures, although a liquid flow in the expansion process is opposite in direction to that in consolidalion. This process can be analyzed well by use of the Terzaghi Voigt combined model. It is shown that the rate of primary deformation in expansion is much smaller than that in consolidation under the same change in compressive pressure. It is also found that the ratio of secondary deformation to total deformation in expansion is much larger than that in consolidation.

Stress relaxation of expressed cake, which is observed when expression is stopped before attaining an equilibrium compression state of a material, can be explained by use of a consolidation and expansion theory. In the relaxation process, local expansion appears near the drainage surface while local consolndash idation appears near the center of the cake. The equilibrium cake stress is uniquely determined by the average consolidation ratio when the expression is interrupted.  相似文献

This paper describes a new write/erase method for flash memory to improve the read disturb characteristics by means of drastically reducing the stress leakage current in the tunnel oxide. This new write/erase operation method is based on the newly discovered three decay characteristics of the stress leakage current. The features of the proposed write/erase method are as follows: 1) the polarity of the additional pulse after applying write/erase pulse is the same as that of the control gate voltage in the read operation; 2) the voltage of the additional pulse is higher than that of a control gate in a read operation, and lower than that of a control gate in a write operation; and 3) an additional pulse is applied to the control gate just after a completion of the write/erase operation. With the proposed write/erase method, the degradation of the read disturb life time after 106 write/erase cycles can be drastically reduced by 50% in comparison with the conventional bipolarity write/erase method used for NAND type flash memory. Furthermore, the degradation can he drastically reduced by 90% in comparison with the conventional unipolarity write/erase method fur NOR-, AND-, and DINOR-type flash memory. We present a patient with continuous melena, diagnosed as rectal varices bleeding. She had a history of esophageal varices, which was treated by endoscopic ligation therapy. Eight years after the treatment of esophageal varices, the continuous melena began. Colonoscopic examination showed that the melena was caused by rectal varices, which were so severe that they could not be treated by either endoscopic sclerotherapy or surgical devascularization. Taking into considering the overall risk of treating rectal varices, we chose the approach of double balloon-occluded embolotherapy (DBOE) with 5% ethanolamine oleate with iopamodol as a liquid embolic material. DBOE is one of the interventional radiology techniques (Morita et al., Acta Hepatol Jpn 1994;35:109-120), but in this case was a completely new and novel clinical procedure for rectal varices. After the DBOE therapy, the condition of rectal varices was markedly improved. Thus, DBOE might be a new tool for treating inoperable rectal varices.  相似文献

Anodic oxidation effects on the structure of the basal and edge surfaces of pyrolytic graphite in alkaline electrolytes have been studied. Laser Raman spectroscopy, a gas-phase chemical modification method, coupled with X-ray photoelectron spectroscopy and secondary ion-mass spectroscopy techniques, were used. Anodic oxidation of the surfaces of pyrolytic graphite in alkaline electrolytes does not cause destruction of their surface structure, even at a higher level of treatment, unlike oxidation of acid electrolytes. In alkaline electrolytes, the number of hydroxyl groups added on the edge surface gradually increases with the increase in treatment level, whereas the number of carboxyl groups does not increase. It was found that anodic oxidation in alkaline electrolytes has a wider permitted range of treatment, in which hydroxyl groups can be added without destroying the edge surface structure, than that found in acid electrolytes. On the other hand, the number of hydroxyl groups added by treating with alkaline electrolytes is smaller than that with acid electrolytes. At a higher treatment level with acid electrolytes, oxidation occurs, even to a depth of 40 nm from the edge surface, whereas with alkaline electrolytes, oxidation occurs only at the surface. On the basis of these results, the effects of electrolytes on the adhesion between carbon fibres and epoxy resin matrix are discussed.  相似文献

The expectation of LET (unrestricted linear energy transfer) for a tritium beta particle around its parent nucleus in water has been calculated by the use of the energy-range relation of electrons. The amount of energy deposited by a beta particle with a certain initial energy over the interval of one micron at a definite position is obtained from the relation by the calculus of difference, and the probability that the event takes place per decay of tritium is estimated from the energy spectrum of tritium beta particles. The calculated values of expectations of LET are higher at positions close to the parent nucleus.  相似文献

Water-silicone oil emulsion with an additive, (C2H5O)3SiC3H6NH2, was examined as a heat storage material. A spiral tube used as a heat exchanger was immersed in a low temperature bath and the emulsion was circulated in the tube to make ice continuously. Ice was separated from the ice–liquid suspension in an outlet tank. The amount of formed ice, the temperatures of the inlet and the outlet of the heat exchanger, and the temperatures in the tube wall were measured and the overall heat transfer coefficient and the heat flux through the tube were calculated. Experiments were carried out, varying the flow rate, the temperature of cooling brine, and the thickness of tube wall. The condition under which slurry ice was formed continuously without adhesion of ice to the cooling wall was clarified. Though decrease in the thermal resistance of the tube increased the rate of ice formation or raised the brine temperature, it narrowed the range of the flow rate and of the brine temperature in which slurry ice was formed continuously.  相似文献
