Membrane mechanisms of conditioning of the defensive reflex in the snails Helix pomatia and H. lucorum were investigated. Tapping on the shell was used as a conditioned stimulus, which under normal conditions produces no defensive reaction. A light blow of air into the pneumostome, called the defensive closure reaction, was used as an unconditioned stimulus. When a combination of conditioned and unconditioned stimuli were presented with 2-4 min interval, the reflex developed over a period of 3 days. The separate conditioned and unconditioned stimuli presented randomly were used as an active control. The electrical characteristics of identified interneurons involved in this defensive behavior were then measured in an isolated preparation. There was shown to be a decrease in the threshold of action potential generation from 20.5 to 16.3 mV and depolarizing shift of membrane potential from -62.1 to -57.0 mV. The electrical characteristics of withdrawal interneurons of active control snails did not differ from those in intact animals. All results show an increase in excitability of withdrawal interneurons after associative learning. 相似文献

Combinations of medetomidine with either propofol or ketamine were compared for the sedation and induction of anaesthesia in dogs undergoing a variety of surgical (60 per cent) and non-surgical (40 per cent) procedures. Eighty-four dogs were used at four sites. Medetomidine was administered intramuscularly at a dose of 1000 micrograms/m2 body surface area 10 to 15 minutes before the induction of anaesthesia by the administration of propofol (n = 44) or ketamine (n = 40) dosed to effect. The dogs became sedated by medetomidine after a mean (sd) time of 6.7 (5.4) minutes, and their heart rates and respiration rates decreased. Sixteen of the dogs suffered an adverse effect, 13 of them vomited. Anaesthesia was induced by the intravenous administration of propofol (2.1 [0.7] mg/kg) or ketamine (3.7 [1.9] mg/kg), and further doses of the anaesthetic were given, depending on the length of the operation, once in 17 per cent, twice in 11 per cent and three or more times in 24 per cent of the cases. The heart rate of the dogs anaesthetised with ketamine was significantly higher than that of the dogs anaesthetised with propofol, but there were no other significant physiological differences. There were 11 adverse side-effects in the ketamine group compared with five in the propofol group and they were generally more severe. The quality of the recovery from anaesthesia was considered to be smooth in 89 per cent of the propofol group but in only 63 per cent of the ketamine group. 相似文献

OBJECTIVE: The expression of contractile isoforms changes during various pathological conditions but little is known about the consequences of these changes for the mechanical properties in human ventricular muscle. We investigated the feasibility of simultaneous determination of protein composition and isometric force development in single cardiac myocytes from human ventricular muscle tissue obtained from small biopsies taken during open heart surgery. METHODS: Small biopsies of about 3 mg wet weight were taken during open heart surgery from patients with aortic valve stenosis. These biopsies were divided in two parts. One part (approximately 2 mg) was used for mechanical isolation of single myocytes and subsequent force measurement while the remaining part was used, in aliquots of 1 microgram dry weight, for protein analysis by polyacrylamide gel electrophoresis. The myocytes were attached with silicon glue to a sensitive force transducer and a piezoelectric motor, mounted on an inverted microscope and permeabilized by means of Triton X-100. Force development was studied at various free calcium concentrations. RESULTS: From all biopsies, myocytes could be obtained and the composition of contractile proteins could be determined. The average isometric force (+/- s.e.m.) at saturating calcium concentration obtained on 20 myocytes from 5 patients amounted to 51 +/- 8 kN/m2. Force was half maximal at a calcium concentration of 2.47 +/- 0.10 microM. CONCLUSION: These measurements indicate that it is possible to study the correlation between mechanical properties and protein composition in small biopsies from human ventricular muscle. 相似文献

1. Eleven kittens were deprived of vision in one eye until the age of between 5 and 14 weeks. Their eyes were then reverse-sutured, they were allowed to survive for a further 3-63 days, and their brains were then examined histologically. 2. Measurement of the cross-sectional area of cells in the lateral geniculate nucleus (LGN) showed that when the reversal of lid suture was performed at the age of 8 or 14 weeks, the mean cell size was smaller in laminae connected to the initially closed right eye than it was in other laminae. 3. When the reversal of lid suture took place at 5 or 6 weeks of age there was a reversal of interlaminar size differences: the initially deprived eye was then connected to laminae containing larger cells. Even within 3 days after the reversal of lid suture, most of the morphological effects of the initial suture had been abolished, and they were fully reversed within 12 days. 4. After removing extracellular Ca2+ with [ethylene bis(oxyethylenenitrilo)]tetraacetic acid, we found that the guinea pig vas deferens (VD) was mechanically responsive to electrical stimulation for a significantly greater length of time than was guinea pig taenia coli (TC). An obvious explanation for these findings is that the VD has more intracellular calcium available for contraction than does the TC. To determine if this explanation is plausible, the volume of internal storage structures within the two muscles was compared. It was found that the volumes of potential sequestering structures in the VD and TC are not significantly different. Next, the affinities of the storage structures for calcium were compared. The VD was found to accumulate approximately twice as much 45Ca as did the TC, as determined by 45Ca autoradiography. Calcium-45 was present to a greater extent in association with surface vesicles, sarcoplasmic reticulum (SR), and mitochondria than in the unassociated state within the cytoplasmic matrix. Based on the results of these experiments, we suggest that the VD and the TC of the guinea pig differ in the affinity of their storage sites for calcium. 相似文献
