Polyvinyl catheters were inserted into the right and left utero-ovarian veins (UOV) and saphenous vein (SV) and artery (SA) of six non-pregnant (O) and five pregnant (P) gilts on day 11 after onset estrus. Beginning on day 12, UOV blood samples were collected at 15-min intervals from 0800 to 1100 hr and 2000 to 2300 hr, and single samples were taken at 1200 and 2400 hrs. Peripheral blood (SA or SV) was sampled at 0800, 1200, 2000 and 2400 hr until gilts returned to estrus (X = 20.6 days) or day 24 of pregnancy. UOV plasma PGF concentrations (ng/ml; n = 1929) were measured by RIA. Status (P vs O) by day interactions were detected (P less than .01) but variances among treatments were heterogenous (P less than .01). Curvilinear day trends were detected for PGF in 0 gilts (P less than .01) but not P gilts. PGF peaks, defined as concentrations greater than two SD above the mean concentration for each gilt, occurred with greater frequency (chi2 = 16.4; P less than .01) in O than P gilts; and mean peak levels (X +/- SE) were 5.04 +/- .27 and 3.84 +/- .13 ng/ml, respectively. Progesterone concentrations were maintained in pregnant pigs and were indicative of luteal maintenance. Systematic differences in day trends of utero-ovarian venous plasma estradiol were detected between O and P pigs. These differences may be of paramount physiological importance and are discussed.  相似文献

The recent discovery of forepaw preferences (handedness) in toad species has provided some insight into the evolution of brain lateralization. We tested the prediction that, as in higher vertebrates, visual lateralization and other motor preferences (footedness) also exists in toad species. During feeding periods, South American cane toads, Bufo marinus, showed a population bias to strike with the tongue at other toads occupying their left visual field. This is the first demonstration of lateralized visual behaviour in an amphibian species. Tongue striking at an individual's eyes or head may sometimes delay its approach to prey already seen by the attacker, or may dislodge prey from its mouth. In addition, we report hindlimb preferences (footedness) for contact righting in three species of toad (B. marinus, the European green toad, B. viridis, and the European common toad, B. bufo). After being fully overturned on to their back on a horizontal surface, toads initiated and completed righting using the hindlimbs and with only perfunctory use of the forepaws. Together, the findings of visual lateralization and footedness demonstrate that in toads, as in higher vertebrates, behavioural lateralization is not restricted to handedness. The hypothesis that lateralized brain functions in birds and mammals might have arisen from a common lateralized ancestor is therefore supported. Mental health practitioners are often called upon to provide services to children, adolescents, and families in the aftermath of traumatic experiences such as child neglect, sexual or physical abuse, family/domestic violence, sexual assault, interpersonal violence, school and community violence, serious accidental injury, catastrophic medical illness, traumatic bereavement, or mass casualty events, including natural and man-made disasters. The National Child Traumatic Stress Network (NCTSN) was established in 2001 to raise the standard of care and improve access to services for traumatized children, their families, and communities throughout the United States. This article describes the development of the NCTSN, its structure, programs, and many of the products and resources--including online lectures, training programs and videos, and searchable databases of child trauma resources--available through the NCTSN Web site (www.nctsn.org) to assist professionals in providing state-of-the-art assessment, treatment, and services to these children and their families. Electrospray ionization transfers thermally labile biomolecules, such as proteins, from solution into the gas phase, where they can be studied by mass spectrometry. Covalent bonds are generally preserved during and after the phase transition, but it is less clear to what extent noncovalent interactions are affected by the new gaseous environment. Here, we present atomic-level computational data on the structural rearrangement of native cytochrome c immediately after solvent removal. The first structural changes after desolvation occur surprisingly early, on a timescale of picoseconds. For the time segment of up to 4.2 ns investigated here, we observed no significant breaking of native noncovalent bonds; instead, we found formation of new noncovalent bonds. This generally involves charged residues on the protein surface, resulting in transiently stabilized intermediate structures with a global fold that is essentially the same as that in solution. Comparison with data from native electron capture dissociation experiments corroborates both its mechanistic postulations and our computational predictions, and suggests that global structural changes take place on a millisecond timescale not covered by our simulations.  相似文献

The effect of nitrate on perchlorate and chlorate reduction by perchlorate-respiring bacteria (PRB), and on chlorate reduction by chlorate-respiring bacteria (CRB), is not well understood, particularly with respect to the induction of pathways used to degrade these different chemicals. Based on kinetic data obtained in a series of batch tests, we determined that perchlorate respiratory enzymes were inducible (by chlorate or perchlorate) and separate from those used for denitrification by PRB strain Dechlorosoma sp. KJ. Aerobically grown cultures of KJ had lag times of greater than 0.3-2 days when transferred to a medium containing only perchlorate, chlorate, or nitrate as an electron acceptor. There were no lag times for transfers between identical media. Washed cells reduced very little nitrate (<10%) when grown only on chlorate or perchlorate. When grown on nitrate, they degraded little chlorate or perchlorate. The same lack of activity with these electron acceptors was also observed using cell extracts and methyl viologen as an electron carrier, indicating a lack of reactivity was not due to failure of the chemical to diffuse into the cell. Taken together, these results indicated that enzymes for perchlorate and nitrate reduction are separately expressed in strain KJ. The presence of small amounts of nitrate in contaminated groundwater may actually help to increase rates of perchlorate reduction once the nitrate is completely removed. When strain KJ was pre-grown on nitrate and perchlorate, perchlorate degradation (in the absence of nitrate) was more rapid compared to cells grown only on perchlorate. Pseudomonas sp. PDA was unable to degrade perchlorate or grow using nitrate, and the induction of enzymes necessary for chlorate respiration differed for strains KJ and PDA. While chlorate reductase and chlorite dismutase activity were induced in KJ by chlorate or perchlorate under anaerobic conditions, these two enzymes were constitutively expressed by PDA under anaerobic and aerobic conditions independent of the presence of chlorate. To our knowledge, this is the first report of constitutive expression of both chlorate reductase and chlorite dismutase in a bacterium.  相似文献

The U.S.-Canadian power system has evolved into a highly integrated grid. Cross-border transmission and coordination of system operations create an interconnected power system with combined imports and exports of electricity of greater than 77?TWh per year. Currently, more than 5 GW of new international transmission lines are in various stages of permitting and development. These transmission lines may enable even greater integration and coordination of the U.S. and Canadian systems, which can in turn increase the reliability and flexibility of North America’s electricity grid and help address challenges associated with integrating high levels of variable renewables. Using a version of the National Renewable Energy Laboratory’s Regional Energy Deployment System (ReEDS) model that incorporates Canada, this analysis quantifies the differences in the evolution of the power system under scenarios in which cross-border transmission capacity is restricted to today’s levels, and scenarios in which new transmission is less restricted. These impacts are analyzed under a “business-as-usual” reference scenario and a scenario in which deep cuts in power sector carbon dioxide emissions levels are achieved. A set of key impact metrics is analyzed, including 1) the composition of generating capacity by technology, 2) system costs, 3) wholesale electricity prices, 4) international electricity exports and imports, 5) transmission capacity, and 6) carbon dioxide emission levels. When new cross-border transmission is not allowed, the United States needs additional capacity (primarily natural gas and renewable energy) to meet domestic needs, while total Canadian capacity is lower because less capacity is needed to export to the United States. This effect is amplified under the carbon cap scenario. Impacts vary on a regional basis, largely due to the different relative sizes of the generation portfolio between countries and regions and the relative impact from cross-border electricity trade. The total impact from restricting cross-border trade on carbon emissions and average wholesale electricity prices is limited, due to the relative size of the domestic power systems and the cross-border trade volume. Cross-border transmission capacity is projected to more than double under the unrestricted transmission capacity scenarios, which exceeds the rate of projected domestic transmission capacity additions in each country.  相似文献

Limit-cycle thermo-acoustic velocity coupling mechanisms are studied in a perfectly-premixed swirl-stabilized combustor using data from 10 kHz repetition-rate stereoscopic particle image velocimetry (S-PIV) and OH planar laser induced fluorescence (PLIF). Five cases over a range of thermal powers and equivalence ratios were investigated, each of which underwent different amplitude limit-cycle oscillations. Proper orthogonal decomposition (POD) of the velocity data showed that each case contained a dynamic helical vortex core (HVC) that rotated around the combustor and greatly affected the flame behavior. Flow and flame statistics were compiled as a function of both the phase in the thermo-acoustic cycle and a phase representing the azimuthal position of the HVC relative to the measurement plane. These data were used to determine the thermo-acoustic energy transfer field at each HVC azimuthal angle, as described by the Rayleigh integral. It was found that periodic deformations of the HVC caused large-scale flame motions, resulting in regions of positive and negative energy transfer. The deformation of the HVC was linked to a swirl number wave that propagated from the burner nozzle. While the mechanism of thermo-acoustic coupling was the same for all cases, the phase between heat release and pressure oscillations varied significantly. This phase relationship was determined by the interaction of the pressure field, swirl wave, HVC deformation, and flame response. It was shown that these can be described by the combination of a Helmholtz resonator and a convective disturbance.  相似文献
