Photosynthetic reaction centers are integral plant membrane protein complexes and molecular photovoltaic structures. We report here that addition of Photosystem I (PSI)-proteoliposomes to retinoblastoma cells imparts photosensitivity to these mammalian cells, as demonstrated by light-induced movement of calcium ions. Control experiments with liposomes lacking PSI demonstrated no photosensitivity. The data demonstrate that PSI, a nanoscale molecular photovoltaic structure extracted from plants, can impart a photoresponse to mammalian cells in vitro.  相似文献

In recent years we have witnessed the emergence and establishment of research in sensor network security. The majority of the literature has focused on discovering numerous vulnerabilities and attacks against sensor networks, along with suggestions for corresponding countermeasures. However, there has been little guidance for understanding the holistic nature of sensor network security for practical deployments. In this paper, we discuss these concerns and propose a taxonomy composed of the security properties of the sensor network, the threat model, and the security design space. In particular, we try to understand the application-layer goals of a sensor network, and provide a guide to research challenges that need to be addressed in order to prioritize our defenses against threats to application-layer goals.  相似文献

Abstract: This study was conducted to evaluate the effect of pulsed electric field (PEF) treatment on anthocyanin extraction from red cabbage using water as a solvent. Mashed cabbage was placed in a batch treatment chamber and subjected to PEF (2.5 kV/cm electric field strength; 15 μs pulse width and 50 pulses, specific energy 15.63 J/g). Extracted anthocyanin concentrations (16 to 889 μg/mL) were determined using HPLC. Heat and light stabilities of the control and PEF-treated samples, having approximately the same initial concentrations, were studied. PEF treatments enhanced total anthocyanin extraction in water from red cabbage by 2.15 times with a higher proportion of nonacylated forms than the control (P < 0.05). The heat and light stabilities of the PEF-treated samples and control samples were not significantly different (P > 0.05). Practical Application: An innovative pretreatment technology, pulsed electric field processing, enhanced total anthocyanin extraction in water from red cabbage by 2.15 times. Manufacturers of natural colors can use this technology to extract anthocyanins from red cabbage efficiently.  相似文献

Iron is essential for the pathogenicity and virulence of Mycobacterium tuberculosis, which synthesises salicyl-capped siderophores (mycobactins) to acquire this element from the host. MbtA is the adenylating enzyme that catalyses the initial reaction of mycobactin biosynthesis and is solely expressed by mycobacteria. A 3200-member library comprised of lead-like, structurally diverse compounds was screened against M. tuberculosis for whole-cell inhibitory activity. A set of 846 compounds that inhibited the tubercle bacilli growth were then tested for their ability to bind to MbtA using a fluorescence-based thermal shift assay and NMR-based Water-LOGSY and saturation transfer difference (STD) experiments. We identified an attractive hit molecule, 5-hydroxyindol-3-ethylamino-(2-nitro-4-trifluoromethyl)benzene ( 5 ), that bound with high affinity to MbtA and produced a MIC90 value of 13 μm . The ligand was docked into the MbtA crystal structure and displayed an excellent fit within the MbtA active pocket, adopting a binding mode different from that of the established MbtA inhibitor Sal-AMS.  相似文献

Acute respiratory distress syndrome (ARDS) and sepsis are risk factors contributing to mortality in patients with pneumonia. In ARDS, also termed acute lung injury (ALI), pulmonary immune responses lead to excessive pro-inflammatory cytokine release and aberrant alveolar neutrophil infiltration. Systemic spread of cytokines is associated with systemic complications including sepsis, multi-organ failure, and death. Thus, dampening pro-inflammatory cytokine release is a viable strategy to improve outcome. Activation of cannabinoid type II receptor (CB2) has been shown to reduce cytokine release in various in vivo and in vitro studies. Herein, we investigated the effect of HU-308, a specific CB2 agonist, on systemic and pulmonary inflammation in a model of pneumonia-induced ALI. C57Bl/6 mice received intranasal endotoxin or saline, followed by intravenous HU-308, dexamethasone, or vehicle. ALI was scored by histology and plasma levels of select inflammatory mediators were assessed by Luminex assay. Intravital microscopy (IVM) was performed to assess leukocyte adhesion and capillary perfusion in intestinal and pulmonary microcirculation. HU-308 and dexamethasone attenuated LPS-induced cytokine release and intestinal microcirculatory impairment. HU-308 modestly reduced ALI score, while dexamethasone abolished it. These results suggest administration of HU-308 can reduce systemic inflammation without suppressing pulmonary immune response in pneumonia-induced ALI and systemic inflammation.  相似文献

One of the major challenges in the treatment of cancer are differential responses of patients to existing standard of care anti-cancer drugs. These differential responses may, in part, be due to a diverse range of genomic, epigenomic, proteomic, and metabolic alterations among individuals suffering from the same type of cancer. Precision medicine is an emerging approach in cancer therapeutics that takes into account specific molecular alterations, environmental factors as well as lifestyle of individual patients. This approach allows clinicians and researchers to select or predict treatments that would most likely benefit the patient based on their individual tumor characteristics. One class of precision medicine tools are predictive, in vitro drug-response assays designed to test the sensitivity of patient tumor cells to existing or novel therapies. These assays have the potential to rapidly identify the most effective treatments for cancer patients and thus hold great promise in the field of precision medicine. In this review, we have highlighted several drug-response assays developed in ovarian cancer and discussed the current challenges and future prospects of these assays in the clinical management of this disease.  相似文献

In this work, melt blending of fumed nanosilica with cyclic olefin copolymer (COC) was carried out to prepare high strength transparent composites. The effects of various loadings (1, 2, 3 and 5 wt%) of nanosilica on the physical, mechanical, dynamic mechanical, thermal, tribological and optical properties of the COC composites were investigated in detail. The tensile test results showed that the nanocomposite with 3 wt% nanosilica content provides the highest tensile strength (55.6 MPa) compared with the nanocomposite with 5 wt% nanosilica content (54.6 MPa), which is believed to be significantly dependent on better dispersion. Moreover, the glass transition temperature (from tan δ) increased from 184 °C for pure COC to 194.3 °C for the COC composite with 3 wt% nanosilica. The scratch test and nano‐indentation results showed that addition of nanosilica increased the stiffness and hardness of the composite, providing higher scratch resistance and lower frictional coefficient. UV?visible spectroscopy measurements showed that the nanocomposites have excellent optical transparency which is similar to that of the pure COC film. © 2013 Society of Chemical Industry  相似文献
