This paper describes the development of a simulation method for WEDM (Wire Electrical Discharge Machining) which can accurately reproduce the discharge phenomena of WEDM on computer. The simulation process consists of searching for discharge locations, removing the workpiece and analyzing wire vibration. Since unclarified parameters such as ignition delay time, explosive force, damping coefficient, and permittivity of dielectric in simulation are difficult to measure from experiments, they were obtained by solving the reverse problem using parametric programming. With the parametric programming method, optimum parameters for simulation are searched simultaneously to minimize differences between simulation results and experimental results.  相似文献

Recombinant adeno-associated virus vectors (AAV) were prepared in high titer (10(12) to 10(13) particles/mL) for the expression of human factor IX after in vivo transduction of murine hepatocytes. Injection of AAV-CMV-F.IX (expression from the human cytomegalovirus IE enhancer/promoter) into the portal vein of adult mice resulted in no detectable human factor IX in plasma, but in mice injected intravenously as newborns with the same vector, expression was initially 55 to 110 ng/mL. The expression in the liver was mostly transient, and plasma levels decreased to undetectable levels within 5 weeks. However, long-term expression of human F.IX was detected by immunofluorescence staining in 0.25% of hepatocytes 8 to 10 months postinjection. The loss of expression was likely caused by suppression of the CMV promoter, because polymerase chain reaction data showed no substantial loss of vector DNA in mouse liver. A second vector in which F.IX expression was controlled by the human EF1alpha promoter was constructed and injected into the portal vein of adult C57BL/6 mice at a dose of 6.3 x 10(10) particles. This resulted in therapeutic plasma levels (200 to 320 ng/mL) for a period of at least 6 months, whereas no human F.IX was detected in plasma of mice injected with AAV-CMV-F.IX. Doses of AAV-EF1alpha-F. IX of 2.7 x 10(11) particles resulted in plasma levels of 700 to 3, 200 ng/mL. Liver-derived expression of human F.IX from the AAV-EF1alpha-F.IX vector was confirmed by immunofluorescence staining. We conclude that recombinant AAV can efficiently transduce hepatocytes and direct stable expression of an F.IX transgene in mouse liver, but sustained expression is critically dependent on the choice of promoter.  相似文献

As basic research for the solution spinning of the high Tc oxide superconductor, the effect of organic acid on the complex formation of poly(vinyl alcohol) (PVA) with the copper [Cu(11)] ion was explored with various carboxylic acids, such as formic acid, acetic acid, n-butylic acid, and lactic acid. The effects of these acids on the interaction between PVA and Cu(II) acetate in aqueous solution were nonexistent, while a chelation of the Cu(II) ion and lactic acid was detected. In the solid state, a linear complex of PVA with the Cu(II) ion was formed by adding organic acid. The ligand of the linear complex was highly dependent on the kind of acid used. © 1994 John Wiley & Sons, Inc.  相似文献

Electrically evoked auditory brainstem responses (EABR) were recorded in 31 postlingually deafened adults, who had recently received cochlear implants (mini-system, Cochlear Ltd). The wave consisted of three distinct positive peaks labeled P1, P2, and P3 with latency of 1.35 (+/- 0.14), 2.17 (+/- 0.18) and 4.08 (+/- 0.31) ms, respectively. The P3 threshold (EABR-T) and slope (EABR-S) were 0.9 (+/- 0.47) mA and 0.6 (+/- 0.28) muv/mA, respectively. The relationships between the EABR parameters (EABR T and -S of the P3 wave) and age, duration of deafness, promontory test and subjective response (T and C-level) were investigated. The scattergram showed a strong negative linear relationship between EABR-S and subjective T-level. This finding suggests that EABR-S is good measure of postoperative perception.  相似文献

The applicability of a powerful but still easy to use technique, based on a phenomenological theory of viscoelasticity, for processing and analyzing dynamic mechanical data of some rice gels was investigated. Based on this theory a continuous relaxation spectra was generated by application of Tikhonov regularization procedure on continuous Maxwell model. Interpretation of relaxation spectra in terms of number of peaks, its peak intensity H(λ) and appearance of its main distribution peak and magnitude of equilibrium elasticity modulus (Ge) of continuous Maxwell model was found to appropriately reflect main peculiarities of the viscoelastic behavior of rice starch gels. An increase in number of peaks in the relaxation spectra was observed for starch gels having higher amylose content indicating the creation of more heterogeneous structure. H(λ) and Ge values also increased with increase in amylose content demonstrating a transition of the system to more stable state like a gel.  相似文献

Hepatocyte multicellular aggregates (spheroids), which maintain high expression of liver functions, have been advocated as a useful culture technique for various cell-based assays. In this study, we investigated the drug metabolic function of a hepatocyte spheroid microarray (HSM) chip, which contained an array of 672 spheroids of primary rat hepatocytes within a 100-mm2 region in the center of a poly(methylmethacrylate) plate (24 × 24 mm) and used an alkoxyresorufin (ethoxy-, methoxy-, pentoxy- and benzyloxyresorufin) O-dealkylase assay system. Ethoxyresorufin O-dealkylase (EROD) activity of the HSM chip initiated by 3-methylcholanthrene (3-MC), an inducer of cytochrome P450 enzymes, was 5- to 10-fold higher than that of monolayer hepatocytes, with activity being maintained for at least 2 weeks. We also demonstrated that 3-MC induced EROD, methoxyresorufin O-dealkylase (MROD) and benzyloxyresorufin O-dealkylase (BROD) activities in the HSM chip, while sodium phenobarbital (P450 inducer) induced pentoxyresorufin O-dealkylase (PROD), BROD, EROD and MROD activities. Induction of these activities was confirmed by increased gene expression of the related P450 enzymes. These results showed that the HSM chip had a good response to P450 inducers and that function was maintained for long periods of time. The HSM chip therefore may be a promising cellular platform for drug metabolic assays using hepatocytes.  相似文献

Specification tests defined in the Japanese Food Sanitation Law were conducted on 7 polylactic acid food-contact products. Moreover, the content and migration of other compounds were examined by means of ICP-AES, GC/MS and mutagenicity tests. All products met their specifications, and migration levels of heavy metals were negligible. No notable peak was observed in GC/MS analysis. Moreover, all products gave negative results in both rec-assay and the umu-test. An increase in the β-galactosidase activity in the umu-test observed with the migration solution of soup bowl was due not to polylactic acid, but to the polyurethane coating.  相似文献

Diabetic patients are prone to severe bacterial infections. The functional alterations of neutrophils by hyperglycemia are thought to be partially responsible for such infections. In this study, we investigated the functional changes of neutrophil-like differentiated cell lines (dHL-60, dTHP-1, and dNB-4) by treatment with 5.5 mM, 11 mM, or 35 mM of glucose. In dHL-60 cells, the incubation with high glucose (35 mM) resulted in the enhancement of cell aggregation, the suppression of cellular fragility, the induction of reactive-oxygen species (ROS) production by phorbol myristate acetate (PMA) stimulation, and the impairment of phagocytosis. In dTHP-1 cells, the treatment with higher glucose generated the suppression of cellular fragility and extremely impaired phagocytosis (by 35 mM), and induced ROS production due to PMA stimulation (by 11 mM). Furthermore, the higher glucose exposure to dNB-4 cells enlarged intracellular vacuoles (by 35 mM) and induced ROS production due to PMA stimulation (by 11 mM). Since the ROS generation of those cells was enhanced only after PMA stimulation under the higher glucose conditions, glucose may have a priming effect rather than a triggering effect. These extraordinary sensitivities caused by the higher glucose treatments may reflect the dysfunction or overactivation of neutrophils.  相似文献

Effect of cooking on the antioxidant properties of coloured peppers   总被引:1,自引:0,他引:1  
Pepper (Capsicum annum L.) has long been recognized as an excellent source of antioxidants, being rich in ascorbic acid and other phytochemicals. This study was conducted to investigate the effect of different cooking methods on the antioxidant properties of coloured peppers. Six varieties of peppers were subjected to different cooking methods, such as microwave heating, stir-frying and boiling in water, for 5 min individually. The cooked and raw peppers were analyzed for radical-scavenging activity (RSA) and total polyphenol content (TP) using 1,1-diphenyl-2-picrylhydrazyl–high-pressure liquid chromatography (DPPH)–HPLC and Folin-Ciocalteu methods, respectively. The samples were also evaluated for ascorbic acid content (AsA) by HPLC. Total carotenoid content was determined spectrophotometrically. Results suggest that there is no significant (P > 0.05) difference in RSA, TP, AsA and total carotenoid contents between the cooked and raw peppers when processed for 5 min. However, the cooked peppers show marked differences (P < 0.05) in the RSA, TP and AsA when cooked for 5 min in boiling water with further reduction observed after boiling for 30 min. This may be due to the leaching of antioxidant compounds from the pepper into the cooking water during the prolonged exposure to water and heat. Therefore, it is vital to use less water and cooking time and also to consume the water used for boiling so as to obtain the optimum benefits of bioactive compounds present in peppers. It is concluded that microwave heating and stir-frying without using water are more suitable cooking methods for pepper, to ensure the maximum retention of antioxidant molecules.  相似文献
