Pattern recognition receptors (PRRs) play a crucial role in inducing inflammatory responses; they recognize pathogen-associated molecular patterns, damage-associated molecular patterns, and environmental factors. Nucleotide-binding oligomerization domain-leucine-rich repeat-containing receptors (NLRs) are part of the PRR family; they form a large multiple-protein complex called the inflammasome in the cytosol. In mammals, the inflammasome consists of an NLR, used as a sensor molecule, apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC) as an adaptor protein, and pro-caspase1 (Casp1). Inflammasome activation induces Casp1 activation, promoting the maturation of proinflammatory cytokines, such as interleukin (IL)-1β and IL-18, and the induction of inflammatory cell death called pyroptosis via gasdermin D cleavage in mammals. Inflammasome activation and pyroptosis in mammals play important roles in protecting the host from pathogen infection. Recently, numerous inflammasome-related genes in teleosts have been identified, and their conservation and/or differentiation between their expression in mammals and teleosts have also been elucidated. In this review, we summarize the current knowledge of the molecular structure and machinery of the inflammasomes and the ASC-spec to induce pyroptosis; moreover, we explore the protective role of the inflammasome against pathogenic infection in teleosts. 相似文献

Control of magnetization direction is essential for the wide application of ferromagnets; it defines the signal size of memory and sensor. However, the magnetization itself causes a dilemma. While its size matters to obtain strong responses upon its reversal, the large magnetization concomitantly suppresses the range of its directional control because of the demagnetizing field. On the other hand, realization of the desired magnetic anisotropy requires careful engineering of crystalline and interfacial effects to overcome the demagnetization barrier. Thus, it would be ideal if one could find alternative magnets that carry no magnetization but strong responses. The discovery of a topological metallic state in the antiferromagnet Mn3Sn is significant; they host a large Berry curvature in momentum space, enabling the observation of disproportionately large transverse responses such as anomalous Hall and Nernst effects, the key functionalities for replacing ferromagnets in the magnetic devices. Here, the experimental realization of omnidirectional control of the large responses in an antiferromagnet is reported. In particular, it is demonstrated that the absence of shape anisotropy enables the omnidirectional control, and lifts the shape constraint in designing the magnetic devices. This work lays the technological foundation for developing simple-structured high-performance devices including multi-level memory and heat flux sensor. 相似文献

This paper describes a characterization method of disturbance force in a linear drive system supported by high-precision rolling guideways. Disturbance force is divided into the friction component and other disturbance force. Experimental characterization method to analyze effect of global position, effect of micro-position and effect of table velocity is suggested. With stop-dwell-go and reversal motions, characterization experiment and analysis are conducted on a positioning system consists of a linear motor and rolling guideways. Firstly, global disturbance force depends on global position and stick and slip friction components are characterized. Secondly, stick friction force is separated from stick and slip friction force in the micro-region. Thirdly, influence of the velocity is analyzed. Finally, a modified single bristle model, which describes friction characteristics in the micro-regime, is shown to simulate the measured disturbance force. 相似文献

Antibodies specific to a particular target molecule can be used as analytical reagents, not only for in vitro immunoassays but also for noninvasive in vivo imaging, e.g., immunoscintigraphies. In the latter case, it is important to reduce the size of antibody molecules in order to achieve suitable in vivo "diagnostic kinetics" and generate higher-resolution images. For these purposes, single-chain Fv fragments (scFvs; M(r) < 30 kDa) have greater potential than intact immunoglobulins (~150 kDa) or Fab (or Fab') fragments (~50 kDa). Our recent observation of enhanced tenascin-C (Tnc) expression at sites of cardiac repair after myocardial infarction prompted us to develop a radiolabeled scFv against Tnc for in vivo imaging of heart disease. We cloned the genes encoding the heavy and light chain variable domains of the mouse anti-Tnc monoclonal antibody 4F10, and combined them to create a single gene. The resulting scFv-4F10 gene was expressed in E. coli cells to produce soluble scFv proteins. scFv-4F10 has an affinity for Tnc (K(a) = 3.5 × 10(7) M(-1)), similar to the Fab fragment of antibody 4F10 (K(a) = 1.3 × 10(7) M(-1)) and high enough to be of practical use. A cysteine residue was then added to the C-terminus to achieve site-specific (111)In labeling via a chelating group. The resulting (111)In-labeled scFv was administered to a rat model of acute myocardial infarction. Biodistribution and quantitative autoradiographic studies indicated higher uptake of the radioactivity at the infarcted myocardium than the noninfarcted one. Single photon emission computed tomography (SPECT) provided in vivo cardiac images that coincided with the ex vivo observations. Our results will promote advances in diagnostic strategies for heart disease. 相似文献

Crystallization of an amorphous solid is usually accompanied by a significant change of transport properties, such as an increase in thermal and electrical conductivity. This fact underlines the importance of crystalline order for the transport of charge and heat. Phase‐change materials, however, reveal a remarkably low thermal conductivity in the crystalline state. The small change in this conductivity upon crystallization points to unique lattice properties. The present investigation reveals that the thermal properties of the amorphous and crystalline state of phase‐change materials show remarkable differences such as higher thermal displacements and a more pronounced anharmonic behavior in the crystalline phase. These findings are related to the change of bonding upon crystallization, which leads to an increase of the sound velocity and a softening of the optical phonon modes at the same time. 相似文献

To improve the balance of the electrical conductivity and mechanical strength for dilute Cu-Ti alloys by aging in a hydrogen
atmosphere, the influence of aging temperature ranging from 673 K to 773 K (400 °C to 500 °C) on the properties of Cu-3 at. pct
Ti alloy was studied. The Vickers hardness increases steadily with aging time and starts to fall at 3 hours at 773 K (500 °C),
10 hours at 723 K (450 °C), or over 620 hours at 673 K (400 °C), which is the same as the case of conventional aging in vacuum.
The maximum hardness increases from 220 to 236 with the decrease of aging temperature, which is slightly lower than aging
at the same temperature in vacuum. The electrical conductivity at the maximum hardness also increases from 18 to 32 pct of
pure copper with the decrease of the temperature, which is enhanced by a factor of 1.3 to 1.5 in comparison to aging in vacuum.
Thus, aging at 673 K (400 °C) in a hydrogen atmosphere renders fairly good balance of strength and conductivity, although
it takes nearly a month to achieve. The microstructural changes during aging were examined by transmission electron microscopy
(TEM) and atom-probe tomography (APT), and it was confirmed that precipitation of the Cu4Ti phase occurs first and then particles of TiH2 form as the third phase, thereby efficiently removing the Ti solutes in the matrix. 相似文献

Although the inter-layer coupling in layered materials has attracted considerable interest due to its importance in determining physical properties of two-dimensional systems, studies on the inter-layer coupling in one-dimensional systems have so far been limited. Double-wall carbon nanotubes (DWCNTs) are one of the most fundamental and ideal model systems to study the inter-layer coupling in one-dimensional systems. In this work, Rayleigh scattering spectroscopy and transmission electron microscopy are used to characterize the electronic transition between inner-and outer-nanotubes of the exactly same individual DWCNT. We find that the inter-layer coupling is strong, leading to downshifts in most of the optical transition energies (up to ~0.2 eV) compared to isolated CNTs. We also find that the presence of metallic tubes lead to stronger shifts. The inter-layer screening of Coulomb interactions is one of the key factors in explaining the observed results. 相似文献

We measured atmospheric opacity at 220 GHz at the summit of Mt. Fuji (alt. 3776 m) about one year in order to explore a feasibility of submillimeter-wave astronomical observations. For this purpose, a 220 GHz radiometer system enclosed in a radome (51×51×62 cm3) has been developed. The 220 GHz opacity was lower than 0.06 for a significant fraction (~ 45 %) of time from November 1994 to March 1995. Diurnal variation of the opacity at the summit of Mt. Fuji is so small that continuous observation at submillimeter-wave is possible through day and night. Yearly variation of the opacity is studied from water vapor pressure data measured at the weather station for the past 3 years. To prevent accumulation of ice and snow on the Gore-Tex membrane in the radome, the outer membrane was supplied with a thermal flux of 0.63 kW m?2 and the adjacent metal radome surfaces were supplied with a flux of ~0.9 kW m?2. We evaluate from the 220 GHz transmission data that this heat flux is sufficient to keep the membrane on the radome free of ice and snow during 83 % of the time in 5 winter months. The summit of Mt. Fuji appears to be a promising site for submillimeter-wave observations. 相似文献

X-ray absorption near-edge structure spectroscopy (XANES) was successfully employed to determine the gold valence in the metal-reducing bacterium Shewanella algae after exposure to a 1 mM aqueous HAuCl4 solution for 10-120 min. XANES spectra revealed the oxidation state of gold in the bacterial cells to be Au(0) without any contribution from Au(III), demonstrating that S. algae cells can reduce AuCl4- ions to elemental gold. Transmission electron microscopy (TEM) and energy dispersive X-ray (EDX) analysis confirmed that gold nanoparticles 5-15 nm in size were deposited in the periplasmic space of the bacterial cells; a preferable, cell surface location for the easy recovery of biogenic nanoparticles. 相似文献
