Failures of earth dam embankments and river dikes, which are constructed of sandy soils with low dry densities, have been observed to occur during rising of water levels. In this paper, a large-scale physical model test was conducted in order to investigate the behavior of a small dam embankment as water levels rose. The test results were simulated by use of a consolidation analysis method coupled with an elastoplastic model for unsaturated soils. All parameters used in the simulation were obtained from element tests; oedometer, triaxial compression, soil water retention, and permeability tests. First, in order to verify the parameters identified, direct shear tests were simulated using the consolidation analysis method; results of the simulation and the tests were consistent. From the embankment model test it was found that the crest of the embankment moved upstream at the first stage of impounding and then moved back. After a seepage surface appeared on the downstream slope, tension cracks occurred on the downstream slope, and sliding occurred through the crack and the downstream toe. The results of the simulation were consistent with those of the model test. This consolidation analysis method could be used to simulate the complex deformations induced by saturation collapse and shear strains and even failure behavior. Old embankments constructed with loose densities might have histories in which cracks occurred on the downstream slopes when the reservoir water level rose, and their stabilities might have decreased. 相似文献

Hypoeutectic Al-Si alloys consist of primary α-Al and Al-Si eutectic phases and show typical elasticplastic fracture. To understand their fracture behavior, fracture processes
were simulated using an elastic-plastic finite-element method. The validity of the J-integral-based criterion was verified and applied to the simulations. A complicated model was used to simulate the fracture
in an idealized dendritic microstructure, and four simplified models were intended to more clearly understand the interaction
between a crack and individual α phases. Results show that the crack is attracted to the soft α phase when passing by the α phase, whereas it is repelled when the α phase is close in front of or behind the crack tip. The presence of α phase close in front of or behind the crack tip leads to an amplification of the driving force. However, the α phase beside the tip reduces the driving force. Furthermore, the fracture behavior is mainly affected by the adjacent α phase on one side around the crack tip, while the remote α phase on the opposite side has an offsetting effect. The local stress-strain fields were examined to analyze the simulated

For the development of immunotherapy using MAGE peptides, the identification of additional tumor antigens is required. Because HLA-A24 is the most common allele in Japanese and is also frequently present in Caucasians, MAGE-3-encoded synthetic peptides with binding affinity for HLA-A24 were thus tested for the induction of specific CTLs from the peripheral blood mononuclear cells (PBMCs) of HLA-A24 healthy donors using a simplified method. By using a peptide with a sequence of IMPKAGLLI (amino acid position in MAGE-3 195-203), the CTL responses could thus be induced from unseparated PBMCs by stimulation with freshly isolated, peptide-pulsed PBMCs as antigen-presenting cells (APCs) and by also using interleukin 7 and keyhole limpet hemocyanin for a primary culture. The induced CTLs could lyse HLA-A24 carcinoma cells expressing MAGE-3, as well as the peptide-pulsed target cells, in an HLA class-I restricted manner. The identification of the MAGE-3/HLA-A24 peptide, IMPKAGLLI, may thus potentially offer the opportunities to design peptide-based immunotherapeutic approaches that might prove to be effective in treating patients with MAGE-3-positive malignant tumors. 相似文献

A high-response gamma-ray densitometer was developed for the measurement of void fraction caused by the flashing vaporization of high-pressure and -temperature water under an instantaneous pipe break accident of a boiling water reactor, BWR. The initial conditions of the water were 6.86 MPa in pressure and saturation temperature. In order to prove the reliability and accuracy, the calibration test by dropping the acrylic void simulators and the air injection test into the cold water filled in the pipe were conducted.The following results are obtained in the experiments: (1) the cone slit method is very useful to increase the measuring accuracy, (2) it is clearly observed that the apparent increase of the void fraction occurs after the rarefaction wave passes, (3) the first maximum of the void fraction occurs with some delay time after break. Specimens with weld lines were produced via conventional and film insert molding techniques using two types of materials as the substrate resin, i.e. a polycarbonate/acrylonitrile‐butadiene‐styrene (PC/ABS) blend and glass fiber‐filled polycarbonate (PC‐gf). The formation and morphology of the weld line region was assessed with and without the presence of 0.5‐mm‐thick PC film inserts. The weld line formation and characteristics were found to be dependent on the extent of interaction between the injected resin and the mold surface or the film insert. Better interfacial interaction between the substrate and film led to the distortion of the weld line orientation, which significantly enhanced the mechanical properties of the weld line. The incorporation of glass fibers into the substrate resin would usually reduce the resistance of the weld line towards tensile, flexural and impact loadings. However, with the attachment of film inserts, the mechanical properties of the weld line region have significantly improved, even with the presence of rigid fibers. Upon examination of tensile and impact fracture surfaces of film insert specimens, a unique orientation of fibers across the weld line (parallel to the flow direction and perpendicular to the weld line) could be observed at regions directly under the film. The combination of favorable properties from the unique fiber orientation and distortion of the weld line, as well as the ability of the film to effectively dissipate forces towards a larger area, have synergistically contributed towards the mechanical property enhancement of the weld line region in film insert moldings.
