Non‐destructive X‐ray diffraction techniques were applied in order to monitor the influence of mechanical and shock‐loading on the microstructure of the plastic‐bonded high explosive KS32. The investigations uncovered damage to embedded coarse HMX crystals and to the binder system HTPB‐IPDI. Damage to the crystals occurred already during the kneading process in terms of deformation twinning. On higher loading between 400 MPa (static) and 480 MPa (dynamic) also crystal fracture was observed. The change in the binder structure was found after both static and dynamic loading, but not in the cured, differently kneaded samples. Moreover, the change in binder structure after dynamic loading was verified by dynamic mechanical analysis, and interpreted as a partial damage of the binder rubber shell around the explosive particles. Spliceosome-mediated RNA trans-splicing has become an emergent tool for the repair of mutated pre-mRNAs in the treatment of genetic diseases. RNA trans-splicing molecules (RTMs) are designed to induce a specific trans-splicing reaction via a binding domain for a respective target pre-mRNA region. A previously established reporter-based screening system allows us to analyze the impact of various factors on the RTM trans-splicing efficiency in vitro. Using this system, we are further able to investigate the potential of antisense RNAs (AS RNAs), presuming to improve the trans-splicing efficiency of a selected RTM, specific for intron 102 of COL7A1. Mutations in the COL7A1 gene underlie the dystrophic subtype of the skin blistering disease epidermolysis bullosa (DEB). We have shown that co-transfections of the RTM and a selected AS RNA, interfering with competitive splicing elements on a COL7A1-minigene (COL7A1-MG), lead to a significant increase of the RNA trans-splicing efficiency. Thereby, accurate trans-splicing between the RTM and the COL7A1-MG is represented by the restoration of full-length green fluorescent protein GFP on mRNA and protein level. This mechanism can be crucial for the improvement of an RTM-mediated correction, especially in cases where a high trans-splicing efficiency is required. 相似文献

DNA-templated self-assembly is an emerging strategy for generating functional supramolecular systems, which requires the identification of potent multi-point binding ligands. In this line, we recently showed that bis-functionalized guanidinium compounds can interact with ssDNA and generate a supramolecular complex through the recognition of the phosphodiester backbone of DNA. In order to probe the importance of secondary interactions and to identify side groups that stabilize these DNA-templated self-assemblies, we report herein the implementation of a dynamic combinatorial approach. We used an in situ fragment assembly process based on reductive amination and tested various side groups, including amino acids. The results reveal that aromatic and cationic side groups participate in secondary supramolecular interactions that stabilize the complexes formed with ssDNA. 相似文献

Structured triglycerides (ST) that contain medium- and long-chain fatty acids were synthesized by lipase-catalyzed interesterification
between tricaprylin and peanut oil. To select appropriate enzymes, we investigated nine commercial lipase preparations for
their ability to hydrolyze pure triglycerides as well as natural oils. Three microbial lipases from Rhizomucor miehei (RML), Candida sp. (CSL), and Chromobacterium viscosum (CVL) gave good results, and immobilized preparations were used in the interesterification. RML gave the highest yields of
ST (73%, 40°C), although its hydrolytic activity toward triolein was low. As the temperature was raised to 50°C, the yield
of ST increased to 79%. After 120 h reaction time, remaining activities were high for CSL (71%), moderate for CVL (48%), and
low for RML (20%).

Sedimentation superimposed on industrial cake filtration leads to longer filtration times and often has a detrimental effect on subsequent process steps such as washing and demoisturing. The influence of sedimentation is seldom recognised in laboratory filtration experiments. Methods are presented for evaluation of pressure filter experiments with superimposed sedimentation which avoid the error made in the usual evaluation methods. For the case of zone sedimentation the article presents a graphical evaluation and a numerical method of modelling permitting scale up to any desired cake height. In superimposed classifying sedimentation simultaneous measurement of filtrate volume and cake height provide information about local variation in filter cake resistance. It is shown for a model system that, owing to sedimentation, the cake resistance shows a minimum at mean cake height and increases rapidly towards greater heights. The local cake resistance corelates with particle size distributions measured for layers of a horizontally cut filter cake. The method of evaluation presented permints determination of the flow resistance of the uppermost layers of a cake and hence estimation of the gas pressure necessary for demoisturing. The profiles of local filter cake resistance show that the relative cake layering is largely independent of the level of suspension filling. A scale-up model is presented for use in those cases where classifying sedimentation cannot be suppressed. 相似文献

Optically active poly(amide‐imide) oligomers were synthesized by direct polycondensation between an aromatic diamine and a dicarboxylic acid both containing a diphenylsilylene unit. The reaction was carried out using triphenyl phosphite/pyridine in the presence of CaCl2 and N‐methyl‐2‐pyrrolidone as solvent. Oligomers were obtained in good yields and showed high solubility in common aprotic polar solvents. The precursors, monomers and poly(amide‐imide) oligomers were characterized using elemental analysis and Fourier transform infrared and NMR (1H, 13C, 29Si) spectroscopy. Additionally, the main vibrations of the functional groups (C?O, C?C or N? H) in the oligomers with respect to temperature were characterized using Raman spectroscopy. The glass transition temperature was determined by studying the Raman spectra and corroborated using differential scanning calorimetry. The thermal stability was studied using thermogravimetric analysis. The molecular mass of the compounds was obtained from matrix‐assisted laser desorption ionization time‐of‐flight mass spectrometry and their optical properties were analyzed using UV‐visible diode array spectrophotometry. The electronic properties of the oligomers as well as the delocalization of charge carriers within their structures were analyzed using conductance‐voltage curves, which showed that these materials are excellent candidates for integrated optoelectronic applications. Polyesters were synthesized by direct polycondensation of thiophene‐2,5‐dicarboxylic acid and five different silarylene‐containing diphenols using a tosyl chloride/pyridine/N,N‐dimethylformamide system as a condensing agent. Polymers were obtained in good yields and were characterized using Fourier transform infrared and NMR (1H, 13C, 135‐DEPT and 29Si) spectroscopy and elemental analysis. All polymers were completely soluble in aprotic organic polar solvents such as dimethylformamide, dimethylsulfoxide and N‐methyl‐2‐pyrrolidone. The range of effective mass of the polymers (m/z) was 1 × 105–2 × 105, determined using electrospray ionization mass spectrometry. Asymmetry and steric hindrance prevented dense packing of the polymeric chains, showing glass transition temperatures between ? 78 and ? 51 °C and loss of thermal stability at 177–199 °C (10% weight loss). Additionally, the melting points of the polyesters were found to be in the range 62–67 °C. Because of this, the samples were semi‐solid at room temperature. The optical band gaps of the polymers were observed between 4.54 and 4.48 eV, corresponding in all cases to insulator behavior. The molecular structure of the samples was studied using X‐ray diffraction, showing a degree of order that was associated with two monoclinic lattices. Additionally, the conductivity was studied using a two‐point method with contacts on top of polymer films. Prior to the electrical measurement, the samples were polarized in an external electric field of 0.8 to 6.4 V cm?1, and the alignment of the dipoles increased the electrical conductivity. A flexible method is presented, which enables the fabrication of porous as well as dense Si3N4/nano-SiC components by using Si3N4 powder and a preceramic polymer (polycarbosilazane) as alternative ceramic forming binder. The SiCN polymer benefits consolidation as well as shaping of the green body and partially fills the interstices between the Si3N4 particles. Cross-linking of the precursor at 300 °C increases the mechanical stability of the green bodies and facilitates near net shape machining. At first, pyrolysis leads to porous ceramic bodies. Finally, subsequent gas pressure sintering results in dense Si3N4/nano-SiC ceramics. Due to the high ceramic yield of the polycarbosilazane binder, the shrinkage during sintering is significantly reduced from 20 to 15 lin.%. Investigations of the sintered ceramics reveal, that the microstructure of the Si3N4 ceramic contains approx. 6 vol.% nano-scaled SiC segregations, which are located both at the grain boundaries and as inclusions in the Si3N4 grains. 相似文献

Several million tonnes of oil sands coke are generated each year in Alberta, Canada as a by-product of bitumen upgrading. Due to its high carbon content, oil sands coke can be a suitable precursor for the preparation of activated carbon. In this study, delayed and fluid oil sands coke were physically activated in a muffle furnace under select conditions of activation time (2–6 h), temperature (800–900 °C), steam rate (0.3–0.5 mL/min), and activation atmosphere (CO2, CO2 + steam, and N2 + steam). The activated products were characterized using thermogravimetric analysis, X-ray diffraction, scanning electron microscopy, nitrogen adsorption, iodine and methylene blue tests. An increase in activation time and temperature resulted in higher surface areas in both delayed and fluid coke due to an enhanced etching of pores. An increase in steam rate led to the production of the highest specific surface area (577 m2/g) and iodine number (670 mg/g) within delayed coke; whereas, a lower steam rate resulted in the production of the highest specific surface area (533 m2/g) and iodine number (530 mg/g) in activated fluid coke samples. 相似文献
