In this work, a study on the influence of morphologic texture on the residual stress determination by diffraction in metallic materials with cubic and hexagonal symmetry is proposed. To this end, elastic self-consistent model has been developed to properly take into account the morphologic texture. Extreme crystallites morphologies (sphere, disc and fibre) were studied, and coupled with the crystallographic texture to reflect the combined effect of morphologic and crystallographic texture in elasticity. In the case of morphologic texture, a stronger influence than the crystallographic texture on the estimated residual stresses (several tens of MPa difference) was observed. We propose a methodology through a scale transition model to take into account the influence of these different morphologies in the stress analysis by diffraction methods. The main purpose of this work was to make the best choice for lattice planes (hkl) used for residual or internal stress analysis, in elasticity, depending on the morphologic (and crystallographic) texture of the polycrystal, especially when the usual X-ray Elasticity Constants (XECs) are used instead of the stress factors.  相似文献

Stably transfected cell lines are widely used in drug discovery and biological research to produce recombinant proteins. Generation of these cell lines requires the isolation of multiple clones, using time‐consuming dilution methods, to evaluate the expression levels of the gene of interest. A new and efficient method is described for the generation of monoclonal cell lines, without the need for dilution cloning. In this new method, arrays of patterned cell colonies and single cell transfection are employed to deliver a plasmid coding for a reporter gene and conferring resistance to an antibiotic. Using a nanofountain probe electroporation system, probe positioning is achieved through a micromanipulator with sub‐micron resolution and resistance‐based feedback control. The array of patterned cell colonies allows for rapid selection of numerous stably transfected clonal cell lines located on the same culture well, conferring a significant advantage over slower and labor‐intensive traditional methods. In addition to plasmid integration, this methodology can be seamlessly combined with CRISPR/Cas9 gene editing, paving the way for advanced cell engineering.  相似文献

A straightforward analytical method has been developed for the determination of glyphosate and aminomethylphosphonic acid (AMPA), its major metabolite in cereals. This method entails a rapid extraction and derivatization with 9-fluorenylmethyl chloroformate followed by separation with a conventional reversed-phase rapid chromatography used in daily routine analysis and detection by electrospray ionization tandem mass spectrometry. To overcome matrix effects and compensate for any analyte losses during sample treatment, an isotopic dilution approach was used. Since 2010, the monitoring of cereals for the widely used herbicide glyphosate is obligatory to all European Union (EU)-member states, laid down in Commission Regulation (EC) No. 1213/2008. Hence, there is definitively a need for a reliable and easy-to-handle analytical method for monitoring of this compound. The proposed method can be run without having to make time-consuming changes on the equipment used for daily routine analysis. The analytical performance of the method was evaluated according to SANCO/10684/2009 criteria and demonstrated that this method is rugged and cost-effective, therefore suitable for monitoring purposes as well as legislative enforcements within the EU. The apparent recoveries of both analytes were between 97% and 113% with relative standard deviations less than 20%. The limits of quantification of glyphosate and AMPA were 0.02 mg/kg in cereal matrices. Finally, the accuracy of the method was assessed by analyzing a proficiency test material which was available from a previous round (EUPT-C4).  相似文献

Despite of its complex multicomponent organization and its compact architecture, the Stratum corneum (SC) is not completely impermeable to substances directly applied on the skin surface. A huge number of works have been dedicated to the understanding of the mechanisms involved in substance permeation by exploring deeper layers than the SC itself. Surprisingly, there is a poor interest in studies relating to interactions which may occur in the near-surface region (i.e. approximately 1 nm depth) of the SC. In this work, equilibrium proton-transfer reactions have been used as probes to define in a fundamental point of view the nature of the SC interactions with its environment. Such titration curves are investigated on 'in vitro' SC (isolated SC from abdominal skin tissue) and on 'in vivo' volar forearm (a sebum poor area). The results are discussed in term of work of adhesion and surface pKa values. Because SC can 'reconstruct' under heating, influence of the temperature on titration curves is investigated and the role of the different components is discussed. Different sigmoidal transitions were observed. Two common pKa values (pKa(1) = 4 and pKa(2) = 11.5) were clearly identified in both cases and associated to an acid-base character. By playing with the temperature of 'in vitro' SC, the 'accessibility' of polar functions was increased, thus refining the results by revealing an amphoteric character with an acid-to-base transition at pH 3.5 and two acid transitions at pH = 6.5 and pH = 11.5. Adhesion forces between an Atomic Force Microscopy (AFM) tip and a single isolated corneocyte through buffered liquid media were also investigated to better understand the role of the individual corneocytes.  相似文献

Irrigation of rice fields in Bangladesh with arsenic-contaminated groundwater transfers tens of cubic kilometers of water and thousands of tons of arsenic from aquifers to rice fields each year. Here we combine observations of infiltration patterns with measurements of porewater chemical composition from our field site in Munshiganj Bangladesh to characterize the mobility of arsenic in soils beneath rice fields. We find that very little arsenic delivered by irrigation returns to the aquifer, and that recharging water mobilizes little, if any, arsenic from rice field subsoils. Arsenic from irrigation water is deposited on surface soils and sequestered along flow paths that pass through bunds, the raised soil boundaries around fields. Additionally, timing of flow into bunds limits the transport of biologically available organic carbon from rice fields into the subsurface where it could stimulate reduction processes that mobilize arsenic from soils and sediments. Together, these results explain why groundwater irrigated rice fields act as net sinks of arsenic from groundwater.  相似文献

Experiments were conducted to determine whether formation of quinone methide intermediates from lignin occurs during ruminal fermentation of corn stover, as indicated by nucleophilic addition reaction with sulphur-containing reducing agents. Corn stover leaf and stem fractions harvested at full maturity were incubated in buffered ruminal fluid without reducing agents or with (NH4)2SO4 (S-control), Na2S.9H2O, cysteine-HCl (cysHCl), or cysHCl plus Na2S.9H2O; and in only buffer with or without cysHCl plus Na2S.9H2O. Mixed reducing agents (cysHCl plus Na2S.9H2O) enhanced ( P< 0.001) in vitro fibre degradation after 48 h, tended to increase solubilisation of fibre ( P =0.07) and dry matter ( P =0.06) in buffer alone, and elevated ( P< 0.001) S-content of residual fibre. In vitro incorporation of S into the undegraded fibre was determined for corn stover fractions of varying lignin compositions that were harvested at two maturities (early dent and full maturity) in 2 years. Extent of fibre degradation was correlated with extent of S-incorporation ( r =-0.54, P< 0.001), and with lignin methoxyl content ( r =-0.84, P< 0.001). The negative association of lignin methoxyl content with digestibility is explained by the relative likelihood of quinone methide intermediate formation from guaiacyl and syringyl units in lignin.  相似文献

The aim of the present work was to study the effect of dietary supplementation of Euphorbia heterophylla on the quality of the Guinea pig meat. Forty guinea pigs were divided into two groups fed ad libitum during 46 days a Panicum maximum diet (Panicum diet) or a mixed diet (75% Panicum maximum + 25% Euphorbia heterophylla) (Paneuphorbia diet) to compare their effects on performances and on the composition of guinea pig tissues and carcass. Daily weight gain, liver weight, carcass yield, and the lipid content of both the carcass and the perirenal fat were significantly increased by the Paneuphorbia diet. Feeding Paneuphorbia diet increased (P < 0.05) the n-3 PUFA content in perirenal fat, muscle, liver and in the carcass and decreased (P < 0.05) the n-6/n-3 ratios in all these tissues and the carcass. In conclusion, this study shows that Euphorbia heterophylla is a source of n-3 fatty acids which can improve significantly the n-3 PUFA content of Guinea pig meat and carcass.  相似文献

Changes in volatile compounds were monitored in whiting (Merlangius merlangus), cod (Gadus morhua) and mackerel (Scomber scombrus) and related to spoilage. Data are presented from headspace/mass spectrometric (HS/MS) analysis and solid‐phase microextraction/gas chromatographic/mass spectrometric (SPME/GC/MS) analysis at two time points (day 0 and day 10) during storage at 4 °C. HS/MS revealed 24 ions that could be used as markers of spoilage. SPME/GC/MS identified 86 compounds, 20 of which could perhaps be used to characterize freshness: principally alcohols, ketones, aldehydes and C2–C11 esters.
