BACKGROUND: The mass transfer of model drugs Lidocaine hydrochloride and Dihydroquercetin from hydrogels (the usual carriers for topical drugs), and hydrogels containing liposomes, as novel drug vehicles, was studied. Diffusion experiments were performed using a Franz diffusion cell. Experimental data were used to calculate drug diffusion coefficients across membranes, and their effective diffusion coefficients from hydrogels and liposome containing hydrogels. For the first time the diffusion resistance of all drug carriers was determined from corresponding diffusion coefficients. The main aim of this work was the study of drug diffusion coefficients from liposomes and their comparison with related diffusion coefficients from hydrogels to find how liposomes contribute to prolonged and controlled drug release. RESULTS: Drug diffusion coefficients were: 1.38 · 10?8m2 s?1 for Lidocaine hydrochloride and 5.96 · 10?9m2 s?1 for Dihydroquercetin, while corresponding effective diffusion coefficients from hydrogels were: 7.82 · 10?10m2 s?1 and 7.98 · 10?10m2 s?1, respectively. Effective diffusion coefficients from liposome‐containing hydrogels were:4.82 · 10?10m2 s?1 (Lidocaine hydrochloride) and 4.305 · 10?10m2 s?1 (Dihydroquercetin). Diffusion resistances for the two hydrogels were almost the same. Very similar values of diffusion resistances for all liposome dispersions were obtained. CONCLUSION: Calculated diffusion coefficients and resistances demonstrate that liposomes, as drug carriers, significantly affect diffusion rates. Prostate cancer (CaP) is the most common type of tumour disease in men. Early diagnosis of cancer of the prostate is very important, because the sooner the cancer is detected, the better it is treated. According to that fact, there is great interest in the finding of new markers including amino acids, proteins or nucleic acids. Prostate specific antigen (PSA) is commonly used and is the most important biomarker of CaP. This marker can only be detected in blood and its sensitivity is approximately 80%. Moreover, early stages cannot be diagnosed using this protein. Currently, there does not exist a test for diagnosis of early stages of prostate cancer. This fact motivates us to find markers sensitive to the early stages of CaP, which are easily detected in body fluids including urine. A potential is therefore attributed to the non-protein amino acid sarcosine, which is generated by glycine-N-methyltransferase in its biochemical cycle. In this review, we summarize analytical methods for quantification of sarcosine as a CaP marker. Moreover, pathways of the connection of synthesis of sarcosine and CaP development are discussed. 相似文献

Electrochemical detection of quantum dots (QDs) has already been used in numerous applications. However, QDs have not been well characterized using voltammetry, with respect to their characterization and quantification. Therefore, the main aim was to characterize CdTe QDs using cyclic and differential pulse voltammetry. The obtained peaks were identified and the detection limit (3 S/N) was estimated down to 100 fg/mL. Based on the convincing results, a new method for how to study stability and quantify the dots was suggested. Thus, the approach was further utilized for the testing of QDs stability. 相似文献

The unprecedented molecular alumoxane [{LAl(F)}2(μ-O)] (2) is easily accessible from the reaction between [{LAl(H)}2(μ-O)] (1) and the mild fluorinating agent Me3SnF. Recently, there has been an extension of three-dimensional (3D) printing technology of metal materials in the medical field. Additive technology has made it possible to manufacture customized implants. However, 3D printing products often require surface treatment. The possible treatments include acid etching. This study investigated the effect of surface etching on the corrosion resistance of Ti–6Al–4V alloy concerning biological applications. The samples were etched in a mixture of hydrofluoric acid and nitric acid. The corrosion behaviour was described by measuring the time dependence of polarization resistance in a saline solution and surface analysis. The results showed that etching creates a fluoride-rich layer on the surface, which negatively affects the corrosion behaviour of the material for up to 24 hr. Cytocompatibility tests showed that the resulting layer does not affect the biocompatibility of the material. 相似文献

pH sensitive, nontoxic, and biocompatible poly(methacrylic) acid (PMAA) based soft networks have been extensively used in the design of systems for targeted drug delivery. Still, their highly hydrophilic nature limits their potential to be used as a carrier of poorly water-soluble substances. With the aim to overcome this limitation, the present study details a new approach for modification of PMAA based carriers using two amphiphilic components: casein and liposomes. The FTIR analysis revealed structural features of each component as well as the synergetic effect that originated from the formation of specific interactions. Namely, hydrophobic interactions between the poorly water-soluble model drug (caffeine) and casein enabled caffeine encapsulation and controlled release, while addition of liposomes ensured better control of the release rate. The morphological properties of the carriers, swelling behavior, and release kinetics of caffeine were investigated depending on the variable synthesis parameters (neutralization degree of methacrylic acid, concentration of caffeine, presence/absence of liposomes) in two different media simulating the pH environment of human intestines and stomach. The data obtained from in vitro caffeine release were correlated and analyzed in detail using several mathematical models, indicating significant potential of investigated carriers for targeted delivery and controlled release of poorly water-soluble substances. 相似文献

BACKGROUND: The meat alternatives market offers a wide range of products resembling meat in taste, flavour or texture but based on vegetable protein sources. These high protein–low purine foods may find application in a low purine or purine‐free diet, which is sometimes suggested for subjects with increased serum urate levels, i.e. hyperuricaemia. RESULTS: We determined purine content (uric acid, adenine, guanine, hypoxanthine, xanthine) in 39 commercially available meat substitutes and evaluated them in relation to their protein content. Some of the products contained a comparable sum of adenine and hypoxanthine per protein as meat. Analysis of variance showed an influence of protein source used. Mycoprotein‐based products had significantly higher contents (2264 mg kg?1) of adenine and hypoxanthine per kg of 100% protein than soybean‐based products (1648 mg kg?1) or mixtures consisting of soybean protein and wheat protein (1239 mg kg?1). CONCLUSION: Protein‐rich vegetable‐based meat substitutes might be generally accepted as meat alternatives for individuals on special diets. The type of protein used to manufacture these products determines the total content of purines, which is relatively higher in the case of mycoprotein or soybean protein, while appearing lower in wheat protein and egg white‐based products. Electrochemical studies of both the a-Si:H based metal/oxide/semiconductor structures and a-Si:H modified Pt electrode as a part of two-electrode cell compartment with electrolyte were performed to obtain information on the nature of mobile hydrogen in a-Si:H. We assume, that the complex formed by a hydrogen molecule and a silicon dangling bond (the origin of Dh states) provides for mobile hydrogen. Information on the process of electron selfexchange (hopping) in a-Si:H driven by diffusion in the presence of a concentration gradient of redox centers is obtained. The capability of the electrochemical methods for studying the role of hydrogen in degradation of a-Si:H caused by bias annealing and/or illumination is demonstrated. Also, it was shown that because of the unique electrocatalytic features of the a-Si:H modified Pt electrode, the latter can act as a “programmable” and selective sensor of heavy metal ions and/or metal complexes. 相似文献

Mycotoxins are secondary metabolites produced by microfungi that are capable of causing disease and death in humans and other animals. This work was aimed at investigation of influence of mouldy wheat contaminated by pathogenic fungi producing mycotoxins on metallothionein levels in hepatic tissue of rats. The rats were administrating feed mixtures with different contents of vitamins or naturally mouldy wheat for 28 days. It was found that the wheat contained deoxynivalenol (80 ± 5 μg per kg of mouldy wheat), zearalenone (56 ± 3 μg/kg), T2-toxin (20 ± 2 μg/kg) and aflatoxins as a sum of B1, B2, G1 and G2 (3.9 ± 0.2 μg/kg). Rats were fed diets containing 0, 33, 66 and 100% naturally moulded wheat. Control group 0, 33, 66 and 100% contained vitamins according to Nutrient Requirements of Rats (NRC). Other four groups (control group with vitamins, vit33, vit66 and vit100%) were fed on the same levels of mouldy wheat, also vitamins at levels 100% higher than the previous mixtures. We determined weight, feed conversion and performed dissection to observe pathological processes. Changes between control group and experimental groups exposed to influence of mouldy wheat and experimental groups supplemented by higher concentration of vitamins and mouldy wheat were not observed. Livers were sampled and did not demonstrate significant changes in morphology compared to control either. In the following experiments the levels of metallothionein as a marker of oxidative stress was determined. We observed a quite surprising trend in metallothionein levels in animals supplemented with increased concentration of vitamins. Its level enhanced with increasing content of mouldy wheat. It was possible to determine a statistically significant decline (p<0.05) between control group and groups of animals fed with 33, 66 and 100% mouldy wheat. It is likely that some mycotoxins presented in mouldy wheat are able to block the mechanism of metallothionein synthesis. 相似文献
