Thyroid peroxidase (TPO), the enzyme responsible for thyroid hormone synthesis, is a key autoantigen in autoimmune thyroid disease. A new triterpenoid, squasapogenol (S-11), was isolated from the hydrolytic products of dilute ethanol (10%) extract of the roots of Glycyrrhiza squamulosa Franch, along with four known triterpenoids. The structure of S-11 was elucidated as olean-11,13(18)-diene-3 beta, 22 beta-diol on the basis of chemical and spectroscopic data. The four known compounds were identified as betulinic acid(S-8), methyl ester of macedonic acid(S-9), soyasapogenol B (S-10) and olean-13(18)-ene-22 alpha-Cl-3 beta, 24-diol(S-12).  相似文献

Pressure-volume-temperature (PVT) properties and vapor pressures of HFC125 (pentafuoroethane; CHF2CF3) have been experimentally obtained. Vapor pressures of HFC-125 have been measured in the range of temperatures from 223 to 338 K and pressures up to 3.54 M Pa with uncertainties of 5 mK and 2.5 kPa, respectively. The vapor pressure equation for this substance was correlated based on the present data. PVT properties of HFC-125 have been determined with a constant-volume apparatus in the range of temperatures from 280 to 473 K, pressures up to 17 M Pa, and densities up to 1145 kg · m–3 with uncertainties of 5 mK, 2.5 kPa, and 0.01%, respectively. BACKGROUND: Stable renal transplant recipients have an excess prevalence of hyperhomocysteinemia, which is a risk factor for arteriosclerosis. OBJECTIVE: To determine the effect of treatment with 1) vitamin B6 or 2) folic acid plus vitamin B12 on fasting and post-methionine-loading plasma total homocysteine levels in renal transplant recipients. DESIGN: Block-randomized, placebo-controlled, 2 x 2 factorial study. SETTING: University-affiliated transplantation program. PATIENTS: 29 clinically stable renal transplant recipients. INTERVENTION: Patients were randomly assigned to one of four regimens: placebo (n = 8); vitamin B6, 50 mg/d (n = 7); folic acid, 5 mg/d, and vitamin B12, 0.4 mg/d (n = 7); or vitamin B6, 50 mg/d, folic acid, 5 mg/d, and vitamin B12, 0.4 mg/d (n = 7). MEASUREMENTS: Fasting and 2-hour post-methionine-loading plasma total homocysteine levels. RESULTS: Vitamin B6 treatment resulted in a 22.1% reduction in geometric-mean post-methionine-loading increases in plasma total homocysteine levels (P = 0.042), and folic acid plus vitamin B12 treatment caused a 26.2% reduction in geometric-mean fasting plasma total homocysteine levels (P = 0.027). These results occurred after adjustment for age; sex; and pretreatment levels of total homocysteine, B vitamins, and creatinine. CONCLUSIONS: Vitamin B6 should be added to the combination of folic acid and vitamin B12 for effective reduction of both post-methionine-loading and fasting plasma total homocysteine levels in renal transplant recipients.  相似文献

The kinesin-related motor protein CHO1/MKLP1 was initially thought to be expressed only in mitotic cells, where it presumably transports oppositely oriented microtubules relative to one another in the spindle mid-zone. We have recently shown that CHO1/MKLP1 is also expressed in cultured neuronal cells, where it is enriched in developing dendrites [Sharp et al. (1997a) J. Cell Biol., 138, 833-843]. The putative function of CHO1/MKLP1 in these postmitotic cells is to intercalate minus-end-distal microtubules among oppositely oriented microtubules within developing dendrites, thereby establishing their non-uniform microtubule polarity pattern. Here we used in situ hybridization to determine whether CHO1/MKLP1 is expressed in a variety of rodent neurons both in vivo and in vitro. These analyses revealed that CHO1/MKLP1 is expressed within various neuronal populations of the brain including those in the cerebral cortex, hippocampus, olfactory bulb and cerebellum. The messenger ribonucleic acid (mRNA) levels are high within these neurons well after the completion of their terminal mitotic division and throughout the development of their dendrites. After this, the levels decrease and are relatively low within the adult brain. Parallel analyses on developing hippocampal neurons in culture indicate that the levels of expression increase dramatically just prior to dendritic development, and then decrease somewhat after the dendrites have differentiated. Dorsal root ganglion neurons, which generate axons but not dendrites, express significantly lower levels of mRNA for CHO1/MKLP1 than hippocampal or sympathetic neurons. These results are consistent with the proposed role of CHO1/MKLP1 in establishing the dendritic microtubule array.  相似文献
