The molecular mechanism of discogenic low back pain (LBP) involves nonphysiological nerve invasion into a degenerated intervertebral disc (IVD), induced by nerve growth factor (NGF). Selective cyclooxygenase (COX)-2 inhibitors are mainly used in the treatment of LBP, and act by suppressing the inflammatory mediator prostaglandin E2 (PGE2), which is induced by inflammatory stimuli, such as interleukin-1β (IL-1β). However, in our previous in vitro study using cultured human IVD cells, we demonstrated that the induction of NGF by IL-1β is augmented by a selective COX-2 inhibitor, and that PGE2 and PGE1 suppress NGF expression. Therefore, in this study, to elucidate the mechanism of NGF suppression by PGE2 and PGE1, we focused on mitogen-activated protein kinases (MAPKs) and its phosphatase, dual-specificity phosphatase (DUSP)-1. IL-1β-induced NGF expression was altered in human IVD cells by MAPK pathway inhibitors. PGE2 and PGE1 enhanced IL-1β-induced DUSP-1 expression, and suppressed the phosphorylation of MAPKs in human IVD cells. In DUSP-1 knockdown cells established using small interfering RNA, IL-1β-induced phosphorylation of MAPKs was enhanced and prolonged, and NGF expression was significantly enhanced. These results suggest that PGE2 and PGE1 suppress IL-1β-induced NGF expression by suppression of the MAPK signaling pathway, accompanied by increased DUSP-1 expression.  相似文献

Hydroxyapatite (HAp) deposition onto positively charged surfaces (i.e., self-assembled monolayers (SAMs) terminated with NH2 head groups) and negatively charged surfaces (i.e., OH-SAMs (weak) and COOH-SAMs (strong)) soaked at 50°C in aqueous supersaturated solutions (1.5 SBF, pH 7.0–7.6; SBF = simulated body fluid) was investigated using a quartz-crystal microbalance. The results revealed that the solution conditions greatly influenced the formation of HAp on the charged surfaces. In a stable supersaturated solution of simulated body fluid (1.5 SBF, pH <7.4), more strongly negative surfaces had a more powerful induction capability for the heterogeneous nucleation of HAp (COOH > OH), whereas nucleation was obviously prohibited on a positive surface (NH2-SAM). On the other hand, after the calcium phosphate particles had nucleated homogeneously in an unstable soaking solution (1.5 SBF, pH ≧7.4), adhesion of the HAp microparticles to the NH2-SAM was observed. A two-step interaction is conceivable to describe the formation of HAp on the positive NH2-SAM: At the first stage, electrostatic interaction dominates the adhesion of HAp microparticles; at the second stage, hydrogen bonds possibly form between the HAp microparticles and the amino head groups of the NH2-SAM, for a firm bonding with the substrate, and the microparticles grow progressively into a thin film. The electrophoretic behaviors of the HAp microparticles confirmed this hypothesis.  相似文献

The biodegradability of sodium alkylpoly(oxyalkylene)sulfates was studied under aerobic conditions by oxygen consumption, total organic corbon (TOC) and methylene blue active substance (MBAS) measurements. MBAS of linear alkylpoly(oxyalkylene)sulfates with propylene oxide (APS) or ethylene oxide (AES) disappeared within 5 days, whereas AES with branched alkyl chains were degraded less than linear AES. APS with propylene oxide from 1 to 3 mol showed BOD/ThOD values of more than 40% after 6 days. Therefore, these surfactants are considered to be readily biodegradable. In comparison to the biodegradability of APS and AES, the existence of propylene oxide groups resulted in a slight decreasing in oxygen consumption and TOC removal. Linear APS with PO of 1~3 mol were degraded according to Swisher's distance principle up to a C16 alkyl chain length. That is, increasing distance principle up to a C16 alkyl chain length. That is, increasing distance between sulfate and chain end increased the rate of biodegradation of these surfactants. Furthermore, from the biodegradation test of35S-C12E3S, it is suggested that the initial step of biodegradation is attack on the terminal methyl group.  相似文献

Diarylacetylene monomers having trimethylsilyl groups and other substituents (substituted biphenyl, 1a and 1b; trimethylsilylmethylphenyl, 1c-e) were synthesized and polymerized with TaCl5-n-Bu4Sn catalyst to produce the corresponding poly(diarylacetylene)s (2a-d). Polymers 2a-c had high molecular weights and were soluble in common organic solvents. Free-standing membranes of 2a-c as well as previously reported 2f-h were prepared by the solution-casting method. Desilylation of these Si-containing polymer membranes was carried out with trifluoroacetic acid to afford 3a, 3b, and 3f-h. Upon desilylation, biphenyl-containing membranes became less permeable (3a, b), whereas fluorene-containing membranes became more permeable (3f-h). In particular, 3h exhibited extremely high gas permeability (PO2 = 9800 barrers), which is about the same as that of poly(1-trimethylsilyl-1-propyne). Desilylated membranes 3a and 3f-h showed different gas permeability from that of polymers 2i-k which have the identical chemical structures and obtained directly by the polymerization of the corresponding monomers.  相似文献

Ultra-wide-band tellurite-based fiber Raman amplifier   总被引:2,自引:0,他引:2  
We describe the first wide-band tellurite-based fiber Raman amplifier (T-FRA) for application to seamless ultra-large-capacity dense wavelength-division multiplexing (WDM) systems. First, we confirmed that the Raman scattering characteristics of the tellurite-based fiber has so large a gain coefficient and Stokes shift that we can achieve a wide-band tellurite-based fiber Raman amplifier with a shorter fiber length than when using silica-based fiber. Second, we investigated the small signal gain and the signal transmission characteristics for a high gain and high output power operation with a single-stage amplifier. Focusing on double Rayleigh scattering, we compared the high gain limit of tellurite- and silica-based fibers. We then studied the impact of nonlinear effects by measuring the bit error rate (BER) when using a two-stage amplifier with a high output power of 18.8 dBm in which we simultaneously amplified eight channel signals in the L-band located on the ITU 100-GHz grid. Finally, we designed a wide-band tellurite-based fiber Raman amplifier with a multiwavelength band pumping scheme. We constructed this amplifier with a tellurite-based fiber only 250 m in length pumped by four-wavelength-channel laser diodes, and it provided a 160-nm bandwidth with a gain of over 10 dB and a noise figure below 10 dB from 1490 to 1650 nm. We also measured the BER to confirm the transmission characteristics of the amplifier for single channel operation over the whole signal wavelength range of 160 nm. We thus confirmed that the amplifier could be employed in ultra-high-capacity WDM systems.  相似文献

South-East Asian population is daily exposed to strong sunlight. As a result, the majority of population will have darker, ethnic skin. Moreover, many people suffer from dark spots, hyperpigmentation, which is considered to be a skin disorder and causes psychological disturbance. To treat dark spots, most of the population will still rely on traditionally used crude drugs, knowledge about which is transferred from generation to generation. Fifty-two crude drugs were selected based on the survey performed among local healers and beauticians of different ethnic origin. These crude drugs were screened for mushroom tyrosinase inhibitory activity, as tyrosinase inhibitors are becoming increasingly important as cosmetic and medicinal products, primarily to control hyperpigmentation. Among the tested crude drugs, methanolic extracts of Glycyrrhiza glabra, Morus alba, Syzygium aromaticum, Citrus aurantifolia, Cypreae moneta, Punica granatum and Citrus aurantium, at the final concentration of 50 microg mL(-1), showed mushroom tyrosinase inhibitory activity of 78.9%, 71.0%, 69.4%, 59.0%, 56.0%, 53.4 and 51.9%, respectively, with 91.4% inhibitory activity of kojic acid taken as positive control. To our knowledge, this is the first report that extracts of Cypreae moneta shell and Syzygium aromaticum flowering bud have tyrosinase inhibitory activity. These potent extracts were further evaluated at different concentration. The final concentration of the extracts in reaction mixtures was 50, 25 and 5 microg mL(-1) for the initial concentration of 1000, 500 and 100 microg mL(-1), respectively. They showed concentration-dependent inhibition of mushroom tyrosinase. Those extracts expressing relatively weak tyrosinase inhibitory activity may act through different inhibition pathway which is not based on tyrosinase activity. Further evaluation of the most potent tyrosinase inhibitors in in vivo conditions would be recommended.  相似文献

The receptor-binding assay (RBA) method for the detection of paralytic shellfish poisoning (PSP) toxins was evaluated for its overall performance in comparison with the mouse bioassay (MBA). An initial study to evaluate the effects of filtering shellfish extracts prior to running the RBA indicated no significant difference between filtered and unfiltered extracts on the determined saxitoxin (STX) concentrations. Next, we tested the RBA assay on 295 naturally contaminated mussel tissue samples, ranging in concentrations from 320 µg STX equiv. kg?1 to 13,000 µg STX equiv. kg?1 by MBA. An overall trend was observed with the RBA giving higher results (256 µg STX equiv. kg?1 on average) than the MBA; however, at low concentrations (< 500 µg STX equiv. kg?1) the RBA results were marginally lower. A third study was conducted using spiked mussel tissue analysed by three independent laboratories, two of which performed the RBA and one the MBA. This multi-laboratory study again showed the RBA to give higher results than the MBA; however, it also revealed that STX determination was accurate by the RBA, unlike the MBA. To optimise the assay for efficient usage under regulatory practice, three suggestions have been made: the use of an initial screening plate to separate those samples that exceed the alert level; use of rapid PSP test kits in the field and in the laboratory for screening negative samples and for early detection of toxicity; and use of an alternate commercially available porcine membrane in place of the laboratory-prepared rat membrane homogenate. The large number of samples analysed and the diversity of the tests conducted in this study further support the RBA as an affordable rapid method for STX detection that is also free of the routine sacrifice of live animals.  相似文献

We manufactured a small-scale hydrogen liquefier with a two-stage 10 K Gifford–McMahon cycle (GM) refrigerator. It had a hydrogen tank with the volume of 30 L that was surrounded by a radiation shield. This liquefier continuously liquefied gaseous hydrogen with the volumetric flow rate of 12.1 NL/min. It corresponds to the liquefaction rate of 19.9 L/day for liquid hydrogen. We proposed a simple estimation method for the liquefaction rate and confirmed that the estimation method well explained the experimental result. To evaluate the estimation method, we applied the estimation method to other liquefiers. In case of a liquefier with the GM refrigerator, we confirmed the estimation method was available for predicting the liquefaction rate. However, in case of a liquefier with the pulse tube refrigerator, the results of the estimation indicated small values as compared with the experimental data. We discuss the details about the estimation method of the liquefaction rate for the small-scale liquefiers.  相似文献

Elasto‐plastic shear deformation and fracture behaviors of short steel stanchions. Short steel stanchions with various width‐to‐thickness ratios of flange and web under axial compression are clarified experimentally as well as analytically. There exists a critical value of shear span ratio Gr(H/h) between shear and bending yield of wide flange steel stanchions. Shear yielding of short steel columns shows no Lüders's lines but very many small protuberance particles up to parallelogram deformed web surfaces. There exists a fundamental large difference between shear and bending yield behaviors of wide flange steel stanchions. Finally the critical shear span ratio of wide flange steel profile will be derived analytically and through experiment proved.  相似文献

Scratch behavior in neat and talc‐filled high crystallinity polypropylene (PP) copolymers containing various additives is investigated using a cosntant load scratch test and two types of indentation tests (Rockwell and Shore D). The talcfilled PP copolymer exhibits high scratch visibilityand scratch depth. The addition of a nucleating agent or lubricant improves the scratch resistance of the talc‐filled PP copolymer. Differential scanning calorimetry, scanning electron microscopy and attenuated total reflectance Fourier transform infrared spectroscopy are used to characterize crystallinity morphology and scratch mechanisms in PP systems. It is found that the scratch resistance of the PP copolymer systems investigated, in terms of scratch depth and scratch visibility, depends mainly on the fracture features generated during the scratch process. The influences of talc, nucleating agent and lubricant on the scratch behavior of PP are discussed.  相似文献
