The effective life of artificial joints is approximately 15 years and it is necessary to extend this effective life. Previously, the use of graft-polymerized 2-methacryloyloxyethyl phosphorylcholine (MPC) was proposed for this purpose. However, the mechanism underlying the wear reduction in ultra-high molecular weight polythene (UHMWPE) by hydrophilic polymers such as MPC polymers remains unexplained. Therefore, this study investigates the characteristics of the MPC polymer membrane grafted onto the surface of a UHMWPE plate. The investigation clearly revealed that the MPC polymer membrane has a finely porous inner structure with a diameter of approximately 100 nm, and hence the membrane has a high water-retention capacity. Furthermore, a 2-axis pin-on-plate sliding test of a UHMWPE pin sample with an MPC polymer grafted on its surface revealed that the water-retention capacity of the porous MPC membrane plays an important role in wear reduction.  相似文献

Glycosylation is the most widespread posttranslational modification in eukaryotes; however, the role of oligosaccharides attached to proteins has been little studied because of the lack of a sensitive and easy analytical method for oligosaccharide structures. Recently, tandem mass spectrometric techniques have been revealing that oligosaccharides might have characteristic signal intensity profiles. We describe here a strategy for the rapid and accurate identification of the oligosaccharide structures on glycoproteins using only mass spectrometry. It is based on a comparison of the signal intensity profiles of multistage tandem mass (MSn) spectra between the analyte and a library of observational mass spectra acquired from structurally defined oligosaccharides prepared using glycosyltransferases. To smartly identify the oligosaccharides released from biological materials, a computer suggests which ion among the fragment ions in the MS/MS spectrum should yield the most informative MS3 spectrum to distinguish similar oligosaccharides. Using this strategy, we were able to identify the structure of N-linked oligosaccharides in immunoglobulin G as an example.  相似文献

The respiratory activity of Escherichia coli K-12 was inhibited by high NaCl concentrations. The addition of compatible solutes such as proline and ectoine led to the recovery of the respiration of E. coli K-12 inhibited by 1 M NaCl to a similar extent as did the addition of glycine betaine. Glucose, an exogenous substrate for the stimulation of respiratory activity, was not taken up by the cells in the presence of 1 M NaCl, but active uptake of glucose was observed following the addition of compatible solutes. As a result, we obtained a good correlation between respiratory activities and glucose uptake rates, suggesting that the glucose uptake activity inhibited by high NaCl concentrations was facilitated by these solutes. The addition of proline did not lead to cell proliferation in the minimal medium containing 1 M NaCl, although the cells took up proline quite efficiently under high osmolarity. On the other hand, cell growth occurred after a lag time in the medium containing glycine betaine or ectoine in place of proline. Similar actions of the compatible solutes mentioned above were observed for E. coli ATCC 9637.  相似文献

An accumulation of evidence shows that endogenous neural stem/progenitor cells (NSPCs) are activated following brain injury such as that suffered during ischemic stroke. To understand the expression patterns of these cells, researchers have developed mice that express an NSPC marker, Nestin, which is detectable by specific reporters such as green fluorescent protein (GFP), i.e., Nestin-GFP mice. However, the genetic background of most transgenic mice, including Nestin-GFP mice, comes from the C57BL/6 strain. Because mice from this background strain have many cerebral arterial branches and collateral vessels, they are accompanied by several major problems including variable ischemic areas and high mortality when subjected to ischemic stroke by occluding the middle cerebral artery (MCA). In contrast, CB-17 wild-type mice are free from these problems. Therefore, with the aim of overcoming the aforementioned defects, we first crossed Nestin-GFP mice (C57BL/6 background) with CB-17 wild-type mice and then developed Nestin-GFP mice (CB-17 background) by further backcrossing the generated hybrid mice with CB-17 wild-type mice. Subsequently, we investigated the phenotypes of the established Nestin-GFP mice (CB-17 background) following MCA occlusion; these mice had fewer blood vessels around the MCA compared with the number of blood vessels in Nestin-GFP mice (C57BL/6 background). In addition, TTC staining showed that infarcted volume was variable in Nestin-GFP mice (C57BL/6 background) but highly reproducible in Nestin-GFP mice (CB-17 background). In a further investigation of mice survival rates up to 28 days after MCA occlusion, all Nestin-GFP mice (CB-17 background) survived the period, whereas Nestin-GFP mice (C57BL/6 background) frequently died within 1 week and exhibited a higher mortality rate. Immunohistochemistry analysis of Nestin-GFP mice (CB-17 background) showed that GFP+ cells were mainly obverted in not only conventional neurogenic areas, including the subventricular zone (SVZ), but also ischemic areas. In vitro, cells isolated from the ischemic areas and the SVZ formed GFP+ neurosphere-like cell clusters that gave rise to various neural lineages including neurons, astrocytes, and oligodendrocytes. However, microarray analysis of these cells and genetic mapping experiments by Nestin-CreERT2 Line4 mice crossed with yellow fluorescent protein (YFP) reporter mice (Nestin promoter-driven YFP-expressing mice) indicated that cells with NSPC activities in the ischemic areas and the SVZ had different characteristics and origins. These results show that the expression patterns and fate of GFP+ cells with NSPC activities can be precisely investigated over a long period in Nestin-GFP mice (CB-17 background), which is not necessarily possible with Nestin-GFP mice (C57BL/6 background). Thus, Nestin-GFP mice (CB-17 background) could become a useful tool with which to investigate the mechanism of neurogenesis via the aforementioned cells under pathological conditions such as following ischemic stroke.  相似文献

Recently, the electric utility industry worldwide has been facing pressure to be deregulated or restructured in order to increase its efficiency, to reduce operational cost, or to give consumers more alternatives. For this aspect, a great deal more research is needed to achieve a better, intelligent knowledge process. The present centralized system for power system control, operation, and planning must be remodeled to cope with these situations. With the promotion for the deregulation of the electric power system, the definition of the objective function for the optimization problem such as outage work operation for electric power systems is becoming critical. Currently, agents are an intense focus in many subfields of computer science and artificial intelligence. Agents are being used in an increasingly wide variety of applications. In this paper, we developed a power system normal operation application by multiagent architecture. Our multiagent system consists of several facilitator agents, equipment agents, and switch‐box agents. Facilitator agent acts as a manager for negotiation process between agents. Equipment agent corresponds to the element of the electric power system, such as bus, transformer, and transmission line, while switch‐box agent is the pseudo object which consists of neighboring current breakers and disconnecting switches. The proposed system realizes the appropriate switching operations by interacting with corresponding agents. The proposed approach is applied to a simple network, and the results show that the proposed multiagent system is an efficient decentralized approach for solving power system normal operations. © 2004 Wiley Periodicals, Inc. Uniaxially strained SiGe layers were fabricated with a newly developed selective-ion-implantation technique. The SiGe layer was grown on the Si substrate, into which laterally selective ion-implantation with stripe pattern was carried out prior to the SiGe growth. A strain-relaxation of the SiGe layer was largely enhanced due to ion-implantation-induced defects selectively in the ion-implanted area while it was hardly enhanced in the neighboring unimplanted area. However, micro-Raman mapping and X-ray diffraction reciprocal space mapping measurements obviously revealed that the relaxed SiGe in the implanted area remarkably influenced a strain state of the neighboring strained SiGe in the unimplanted area, that is, the strain along the stripe line direction was highly relieved due to the stress caused by the neighboring relaxed SiGe while the strain in the direction perpendicular to the line was well maintained. As a result, highly asymmetric strain state, that is, uniaxial strain was realized, where 4 times different relaxation ratios in the two directions were observed. These results indicate that the selective-ion-implantation technique developed in this study has a high potential to realize uniaxially strained Si/Ge channel devices with high mobility.  相似文献

The effective life of artificial joints is approximately 15 years. A smooth metal sliding surface is presumably the most suitable when manufacturing artificial joints; however, the relationship between the characteristics of metal sliding surface and ultra high molecular polyethylene (UHMWPE) wear has not been confirmed. Further, there is no apparent proof that a smooth surface is the optimal option for the improvement in the wear resistance of artificial joints. In this study, we investigated the mechanism of UHMWPE wear and proved that scratch marks caused by a sliding motion against the metal surface are the prime cause of UHMWPE wear. Furthermore, we used a micro-dimpled surface as an effective sliding surface to reduce the UHMWPE wear. A 2-axes pin-on-plate sliding test proved that the life of artificial joints can be extended to approximately 35 years by using a micro-dimpled surface with 1-μm deep dimples.  相似文献

We prepared and characterized multi-walled carbon nanotube (MWCNT) solids by de-fluorination of fluorinated MWCNTs using spark plasma sintering. In order to investigate the influence of the structure of MWCNT on the mechanical strength of the resulting solids, MWCNTs used in this experiment were as-defected MWCNTs modified by nitric acid and as-purified ones as non-defected nanotubes by comparison. The bend strength and modulus of the resulting solids were lower than those of de-fluorinated MWCNT solids derived from as-purified MWCNTs. The mechanical properties of the resulting MWCNT solids were found to be strongly dependent on the structure of the MWCNT.  相似文献

An antimicrobial peptide, temporin L, and its derivative (TL-A2) were employed as anchor peptides and displayed streptavidin on a bacterial magnetic particle (BMP) membrane. The ribotoxin L3 loop (L3) and the arginine-chain peptide (R(12)), which are carrier peptides permeable to eukaryotic cell membranes, were also used. The peptides were labeled with a fluorescent dye, 4-fluoro-7-nitrobenzofurazan (NBD), at the N-terminal region (NBD-peptides) and mixed with BMPs. A specific integration of NBD-temporin L into a BMP membrane was observed. The basic amino acids in temporin L played an important role in the integration into BMPs. Biotin conjugated to the N-terminus of temporin L was integrated into a BMP membrane. The C-terminus of temporin L was incorporated into a BMP membrane, and the N-terminus was located on the BMP membrane surface. The present study shows that temporin L is a stable molecular anchor on BMPs by the binding of soluble protein to the N-terminus.  相似文献
