A versatile solid‐phase approach based on peptide chemistry was used to construct four classes of structurally diverse polyamines with modified backbones: linear, partially constrained, branched, and cyclic. Their effects on DNA duplex stability and structure were examined. The polyamines showed distinct activities, thus highlighting the importance of polyamine backbone structure. Interestingly, the rank order of polyamine ability for DNA compaction was different to that for their effects on circular dichroism and melting temperature, thus indicating that these polyamines have distinct effects on secondary and higher‐order structures of DNA.  相似文献

Structure‐guided protein engineering achieved a variant of the unique racemase AMDase G74C, with 40‐fold increased activity in the racemisation of several arylaliphatic carboxylic acids. Substrate binding during catalysis was investigated by saturation‐transfer‐difference NMR (STD‐NMR) spectroscopy. All atoms of the substrate showed interactions with the enzyme. STD‐NMR measurements revealed distinct nuclear Overhauser effects in experiments with and without molecular conversion. The spectroscopic analysis led to the identification of several amino acid residues whose substitutions increased the activity of G74C. Single amino acid exchanges increased the activity moderately; structure‐guided saturation mutagenesis yielded a quadruple mutant with a 40 times higher reaction rate. This study presents STD‐NMR as versatile tool for the analysis of enzyme–substrate interactions in catalytically competent systems and for the guidance of protein engineering.  相似文献

The detailed reaction mechanism of Di-Air, which showed the unprecedented high deNOx performance at high temperature conditions, was studied in this work. Since the Di-Air phenomenon occurs with continuous short pulse injections of hydrocarbons over NSR (NOx storage and reduction) catalyst, this study focused on the specific function of HC as an effective reductant for NOx reduction reaction. As a first step, the deNOx performance was compared with three different reductant gases including CO, H2 and C3H6 in a modified synthetic gas bench equipped with a gas injector which enables continuous small amount of injections. At inlet gas temperature of 450 °C C3H6 showed the best deNOx performance whereas H2 was the best at 150 °C. Moreover the result of temperature programmed desorption showed that intermediate species represented by –NCO (isocyanates) produced from C3H6 was thermally more stable than that produced from CO. These results confirmed that the injected HC reacts with adsorbed NOx on NSR catalyst generating thermally stable intermediates, which could contribute the high deNOx performance at high temperature conditions.  相似文献

Sunflower seeds ((Helianthus annuus were roasted for 6, 12, 20 or 30 min at a frequency of 2450 MHz using a domestic microwave oven. After the kernels were separated from the sunflower seeds, the quality characteristics and the compositions of the oils were investigated in relation to their tocopherol distributions, and they were further evaluated as compared with an unroasted oil sample. Only minor increases (p < 0.05) in chemical and physical changes of the oils, such as the carbonyl value, the p‐anisidine value and the color development, occurred at a prolonged roasting period. Significant decrease (p < 0.05) was observed in the amounts of phospholipids in the oils after microwave roasting. Nevertheless, compared to the original level, more than 92 wt‐% tocopherols still remained after 30 min of roasting. With a few exceptions, these results indicate that the exposure of sunflower seeds to microwaves for 12 min caused no significant (p < 0.05) loss or change in the content of tocopherols and polyunsaturated fatty acids in the kernels.  相似文献

Morphology and fracture mechanisms in two nanoclay‐filled epoxy systems were investigated using both microscopy and spectroscopy tools. Clay exfoliation was achieved using a series of sample preparation steps, and confirmed using wide angle X‐ray diffraction (XRD) and transmission electron microscopy (TEM) techniques. Significant improvement in modulus was obtained when clay exfoliation was achieved. Incorporation of core‐shell rubber (CSR) in both caly‐filled epoxy systems leads to greatly enhanced fracture toughness. Optical microscopy and TEM observations of the CSR‐modified nanocomposites suggest that CSR cavitation. shear yielding of the matrix, clay layer delamination. CSR bridging, crack bifurcation. and crack deflection are among the operative toughening mechanisms observed, depending on the nature of the epoxy matrix utilized.  相似文献

Ceramic photonic crystals with diamond structure were fabricated using stereolithography and successive sintering. The green body of an epoxy resin incorporating 10 vol% TiO2–SiO2 was formed by stereolithography and then heated in air at 1100°–1400°C for 2 h. The sintered products maintained the diamond structure with a linear shrinkage ratio of about 57% and a porosity of 38%. The ceramic photonic crystal with eight unit cells showed a photonic band gap at the center frequency of 23.5 GHz. This fabrication method of three-dimensional (3D) ceramic photonic crystals is applicable to other 3D structural ceramics and does not require any molding techniques.  相似文献

The present study focused on the design and synthesis of covalent DNA dendrons bearing multivalent cytosine-phosphate-guanine oligodeoxynucleotides (CpG ODNs) that can stimulate the immune system through the activation of TLR9. These dendrons were synthesized using branching trebler phosphoramidite containing three identical protecting groups that enabled the simultaneous synthesis of multiple strands on a single molecule. Compared with linear ODNs, covalent DNA dendrons were found to be more resistant to nuclease degradation and were more efficiently taken up by macrophage-like RAW264.7 cells. Cellular uptake was suggested to be mediated by macrophage scavenger receptors. The covalent DNA dendrons composed of multivalent immunostimulatory branches enhanced the secretion of proinflammatory cytokines TNF-α and IL-6 from RAW264.7 cells, and 9-branched DNA dendrons showed the highest enhancement. Given their enhanced efficacy, we expect covalent DNA dendrons to be useful structures of oligonucleotide medicines.  相似文献

Beneficial effects of dietary phytic acid (myo-inositol hexaphosphate; IP6) have often been explained by its strong iron ion-chelating ability, which possibly suppresses iron ion-induced oxidative damage in the gastrointestinal tract. Because phytic acid is hydrolyzed during digestion, this work aimed to know whether its hydrolysis products (IP2′ IP3′, IP4′ and IP5) could still prevent iron ion-induced lipid peroxidation. Studies using liposomal membranes demonstrated that hydrolysis products containing three or more phosphate groups are able to inhibit iron ion-induced lipid peroxidation although their effectiveness decreased with dephosphorylation. Similarly, they also prevented iron ion-induced decomposition of phosphatidylcholine hydroperoxide. These results demonstrate that intermediate products of phytic acid hydrolysis still possess iron ion-chelating ability, and thus they can probably prevent iron ion-induced lipid peroxidation in biological systems.  相似文献

This series of articles provides, from an international perspective, an analysis of the future of the automotive industry and aluminum’s role in automobile weight reduction. Part I briefly described technological, environmental, and societal issues that are mandating vehicular weight reduction for, among other reasons, purposes of fuel economy. Part II described the status of the weight-reduction technologies that are currently available and projects how they may evolve in the coming years. Carbon-matrix composites with self-assembly interconnected carbon nano-ribbon networks were fabricated using natural and inexpensive rice husks by impregnating the rice husks with transitional metal solutions and sintering. The nano-structure of the composite was characterized using transmission electron microscopy. The electromagnetic interference shielding effectiveness of the composite was evaluated in the frequency range of 30 kHz–1.5 GHz. The electrical conductivity was measured as a function of the sintering temperature. These characteristics were compared with those of carbon-matrix composites without nano-ribbon networks.  相似文献
