Kedarcidin is a potent antitumor antibiotic chromoprotein, composed of an enediyne-containing chromophore embedded in a highly acidic single chain polypeptide. The chromophore was shown to cleave duplex DNA site-specifically in a single-stranded manner. Herein, we report that in vitro, the kedarcidin apoprotein, which lacks any detectable chromophore, cleaves proteins selectively. Histones that are the most opposite in net charge to the apoprotein are cleaved most readily. Our findings imply that the potency of kedarcidin results from the combination of a DNA damaging-chromophore and a protease-like apoprotein.  相似文献

Numerical and experimental studies were performed to assess the transient impact response of 11 T-beams with various dimensions and aspect ratios. Numerical modeling was performed using a three stage finite-element modeling procedure which included modal analysis, resonant analysis, and three-dimensional transient dynamic analysis. The response at impact locations on both the top centerline of the flange and the bottom centerline of the web was investigated. Physical models of three of the beams were constructed in the laboratory to determine the physical response of the beams when subjected to a transient impact and to verify the numerical results. Relationships between the fundamental frequencies and the frequencies of higher cross-sectional modes of vibration were established for the various aspect ratios. Shape factors were derived from the numerical and experimental results. The practical significance of the results is demonstrated for a project where impact-echo testing was used to nondestructively assess the condition of a decommissioned concrete T-shaped girder.  相似文献

A complex diffusion cell was proposed to determine the amount of drug released from topical formulations. The released drug is automatically removed from the diffusion layer by a prestaltic pump to a secondary reservior where it is diluted and could be spectrophoto-metrically assayed and then returned to the reservoir. Permeable acrylic resins were used as efficient retarding materials to prepare controlled release salbutamol sulphate molded tablets. The formulation is simple, efficient, economic and is easily shaped into molded tablets. The effects of two types of acrylic resins, namely: Eudragit RL100 ad Eudragit RS100 in concentrations 1, 2 and 5% w/w on the physical characteristics as well as on the in vitro release patterns of salbutamol sulphate from molded tablets prepared with either polyethylene glycol (PEG) 4000 or 6000 were studied. It was revealed that, as the molecular weight of the PEG increased, the hardness of the tablets increased. Considerable retardation in the drug release was observed by using Eudragit RS100 as compared to Eudragit RL100. The formulation prepared with PEG 6000 and 5% Eudragit RS100 produced much more release time prolongation than the other tested formulations. On the other hand, tablets prepared by the direct compression technique produced a faster release of salbutamol sulphate than those prepared by molding.  相似文献

This paper reports on the in vitro cytotoxicity, bioactivity behaviour and mechanical properties of novel injectable calcium phosphate cement filled with hydroxylated multi-walled carbon nanotubes and bovine serum albumin (CPC/MWCNT-OH/BSA). To predict the in vitro bioactivity of the calcium phosphate composites, we investigated apatite formation on CPC/MWCNT-OH/BSA composites after soaking in simulated body fluid (SBF) for up to 28 days. Compressive strength tests, scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and cell culture experiments with human CCD-18Co fibroblasts cell lines were performed to evaluate the effect of SBF pre-treatment on the mechanical, structural and biological properties of the CPC/MWCNT-OH/BSA composites. Although apatite formation increased significantly with SBF immersion period, the results showed that all soaked CPC/MWCNT-OH/BSA composites exhibited up to 2.5 times lower compressive strength (13–20 MPa), which were however higher than values reported for the strength of trabecular bone (2–12 MPa). Cell culture experiments showed that low concentrations (6.25 and 12.5 μg/ml) of bio-mineralised CPC/MWCNT-OH/BSA composites led to cell proliferative rather than cytotoxic effects on fibroblasts, evidenced by high cell viabilities (104–113%). The novel CPC/MWCNT-OH/BSA composites presented in this study showed favourable cytocompatible and bioactive behaviour along with high compressive strength (13–32 MPa) and are therefore considered as an attractive bone filling material.  相似文献

The efficiency of catalytic ozonation with homogeneous (containing dissolved ions of Fe2+, Mn2+, Cu2+, Ni2+, Co2+, V5+, Cr3+, Mo6+) and heterogeneous (MnO2, Ni2O3, Fe2O3, CuO, Al2O3, CoO, V2O5, Cr2O3, MoO3, TiO2) catalysts and non-accompanied ozonation was compared for degradation of m-dinitrobenzene (m-DNB). Several transition metals in homogeneous and heterogeneous form improved significantly the ozone performance for degradation of m-DNB. This improvement was found to be due to supplementary formation of reactive species (hydroxyl radicals) and better ozone utilization. The effects observed were found to be strongly dependent on the treatment conditions.  相似文献
