The effect of in situ autolysis on cerebral mitochondrial structure and function has been investigated. Mice (n = 9) were sacrificed and stored for up to 24 h under unfavorable post-mortem conditions at 25 degrees C. At different time intervals groups of three animals were submitted to post-mortem dissection and tissue from different regions of the brain was used for the preparation of "free" and synaptosomal mitochondria. On electron microscopic examination, the post-mortem period had no significant influence on mitochondrial morphology and enzymatic activities of complexes I-V of the mitochondrial oxidative phosphorylation system were still present in all the mitochondrial preparations from different regions of the brain, albeit at a reduced levels. Degradation of mitochondrial DNA was virtually absent from mitochondrial preparations during the 24-h period of autolysis, as shown by the presence of intact DNA by Southern blot and PCR analysis. Based on these results, alterations in mitochondrial DNA and deficiencies of mitochondrial respiratory complexes I-V can be recognized in cerebral tissue even after 24 h of unfavorable post-mortem storage conditions.  相似文献

Sometimes, we find decision situations in which it is difficult to express some preferences by means of concrete preference degrees. In this paper, we present a consensus model for group decision making problems in which the experts use linguistic interval fuzzy preference relations to represent their preferences. This model is based on two consensus criteria, a consensus measure and a proximity measure, and on the concept of coincidence among preferences. We compute both consensus criteria in the three representation levels of a preference relation and design an automatic feedback mechanism to guide experts in the consensus reaching process.  相似文献

Tool wear detection is a key issue for tool condition monitoring. The maximization of useful tool life is frequently related with the optimization of machining processes. This paper presents two model-based approaches for tool wear monitoring on the basis of neuro-fuzzy techniques. The use of a neuro-fuzzy hybridization to design a tool wear monitoring system is aiming at exploiting the synergy of neural networks and fuzzy logic, by combining human reasoning with learning and connectionist structure. The turning process that is a well-known machining process is selected for this case study. A four-input (i.e., time, cutting forces, vibrations and acoustic emissions signals) single-output (tool wear rate) model is designed and implemented on the basis of three neuro-fuzzy approaches (inductive, transductive and evolving neuro-fuzzy systems). The tool wear model is then used for monitoring the turning process. The comparative study demonstrates that the transductive neuro-fuzzy model provides better error-based performance indices for detecting tool wear than the inductive neuro-fuzzy model and than the evolving neuro-fuzzy model.  相似文献

Analysis of the cyclic short-term variation of indoor power line channels   总被引:1,自引:0,他引:1  
The purpose of this paper is to analyze the properties of indoor power line communications channels when they are used for broadband transmission. It will be shown that these channels exhibit a short-term variation, due to the fact that high-frequency parameters of electrical devices depend on the instantaneous amplitude of mains voltage. This phenomenon leads to a channel model proposal based on a linear periodically time-varying system and cyclostationary random noise. A measurement system, designed to capture these time variations in real power networks, and some of the most significant results obtained with it, are presented. In addition, a statistical analysis of the measurements performed in several indoor power line scenarios is included, so that the relevance of time variations in actual channels can be evaluated.  相似文献

The presence of ferrite/pearlite bands in dual phase and TRIP assisted steels is a consequence of microchemical segregation which causes mechanical properties anisotropy. Such inhomogeneous phase distribution produces a lowering of the mechanical properties such as fracture behaviour. This anisotropy is commonly not accounted in micromechanics computations which often assume a random distribution of phases in the solid. The present paper deals with an integral model for this undesirable band formation accounting for the solute segregation caused by solidification, microcomponent diffusion present in the austenitisation process, and the nucleation of the transformed phase in segregated regions. In the present work, the model was applied to two industrial grade dual phase steels and two TRIP assisted steels. The influence of such parameters on band formation is summarised in a number of “band prevention plots”, which are aimed at providing the optimum processing conditions for ferrite/pearlite band prevention.  相似文献

Two experiments with human participants are presented that differentiate renewal from other behavioral effects that can produce a response after extinction. Participants played a video game and learned to suppress their behavior when sensor stimuli predicted an attack. Contexts (A, B, & C) were provided by fictitious galaxies where the game play took place. In Experiment 1, participants who received conditioning in A, extinction in B, and testing in A showed some context specificity of conditioning during extinction and a recovery of suppression on test. Experiment 2 demonstrated recovery of extinguished responding when participants were conditioned in A, extinguished in B, and tested in C, a third, neutral context. The experiment also demonstrated that the context of extinction did not control performance by becoming inhibitory. Results are discussed in terms of mechanisms that can produce a response recovery after extinction. The experiments demonstrated a renewal effect: a response recovery that was not attributable to the contexts acting as simple conditioned stimuli and is the first work with human participants to conclusively do so. Molecular screening for pathogenic mutations in sudden cardiac death (SCD)-related genes is common practice for SCD cases. However, test results may lead to uncertainty because of the identification of variants of unknown significance (VUS) occurring in up to 70% of total identified variants due to a lack of experimental studies. Genetic variants affecting potential splice site variants are among the most difficult to interpret. The aim of this study was to examine rare intronic variants identified in the exonic flanking sequence to meet two main objectives: first, to validate that canonical intronic variants produce aberrant splicing; second, to determine whether rare intronic variants predicted as VUS may affect the splicing product. To achieve these objectives, 28 heart samples of cases of SCD carrying rare intronic variants were studied. Samples were analyzed using 85 SCD genes in custom panel sequencing. Our results showed that rare intronic variants affecting the most canonical splice sites displayed in 100% of cases that they would affect the splicing product, possibly causing aberrant isoforms. However, 25% of these cases (1/4) showed normal splicing, contradicting the in silico results. On the contrary, in silico results predicted an effect in 0% of cases, and experimental results showed >20% (3/14) unpredicted aberrant splicing. Thus, deep intron variants are likely predicted to not have an effect, which, based on our results, might be an underestimation of their effect and, therefore, of their pathogenicity classification and family members’ follow-up.  相似文献

Ecdysteroids are widely investigated for their role during the molting cascade in insects; however, they are also involved in the development of the female reproductive system. Ecdysteroids are synthesized from cholesterol, which is further converted via a series of enzymatic steps into the main molting hormone, 20-hydoxyecdysone. Most of these biosynthetic conversion steps involve the activity of cytochrome P450 (CYP) hydroxylases, which are encoded by the Halloween genes. Three of these genes, spook (spo), phantom (phm) and shade (shd), were previously characterized in the desert locust, Schistocerca gregaria. Based on recent sequencing data, we have now identified the sequences of disembodied (dib) and shadow (sad), for which we also analyzed spatiotemporal expression profiles using qRT-PCR. Furthermore, we investigated the possible role(s) of five different Halloween genes in the oogenesis process by means of RNA interference mediated knockdown experiments. Our results showed that depleting the expression of SchgrSpo, SchgrSad and SchgrShd had a significant impact on oocyte development, oviposition and hatching of the eggs. Moreover, the shape of the growing oocytes, as well as the deposited eggs, was very drastically altered by the experimental treatments. Consequently, it can be proposed that these three enzymes play an important role in oogenesis.  相似文献

Various treatments based on drug administration and radiotherapy have been devoted to preventing, palliating, and defeating cancer, showing high efficiency against the progression of this disease. Recently, in this process, malignant cells have been found which are capable of triggering specific molecular mechanisms against current treatments, with negative consequences in the prognosis of the disease. It is therefore fundamental to understand the underlying mechanisms, including the genes—and their signaling pathway regulators—involved in the process, in order to fight tumor cells. Long non-coding RNAs, H19 in particular, have been revealed as powerful protective factors in various types of cancer. However, they have also evidenced their oncogenic role in multiple carcinomas, enhancing tumor cell proliferation, migration, and invasion. In this review, we analyze the role of lncRNA H19 impairing chemo and radiotherapy in tumorigenesis, including breast cancer, lung adenocarcinoma, glioma, and colorectal carcinoma.  相似文献
