Patients with familial amyloidotic polyneuropathy (FAP) showed extremely low plasma apolipoprotein AII (apoAII) levels and apolipoprotein AII/AI (apoAII/AI) ratio while plasma levels of AI, B, CII, CIII, and E were all within normal ranges. To elucidate the reason for these phenomena, we investigated the percent of these proteins contained in high density lipoprotein (HDL) extracted from plasma of FAP patients by ultracentrifugation. The apoAII/AI ratio in extracted HDL was much lower in asymptomatic carriers of FAP as well as FAP patients than that in control subjects. Since a significant amount of apoAII as well as apoAI was recognized in the fraction of the density > 1.21 g/ml in the samples from asymptomatic carriers of FAP and FAP patients, decreased apoAII/AI ratio may result from the increased dissociation of apoAII from HDL during the process of the ultracentrifugation. Electrophoresis of HDL extracted by agarose gel revealed that HDL from asymptomatic carriers of FAP as well as FAP patients increased negative charge, suggesting that nature of HDL itself may change in these subjects. Although urinary excretion of apoAII was increased in carriers of FAP and FAP patients, there was no correlation between plasma and urinary apoAII levels and also no relationship between urinary total protein and apoAII levels. These results suggest that changed affinity of apoAII to HDL may cause the increased secretion of apoAII to the urine and the decreased plasma apoAII level in carriers of FAP and FAP patients.  相似文献

Combined neuropathological and neurochemical assessment of the nucleus of Meynert in senile dementia of Alzheimer type (SDAT) have demonstrated that the cholinergic biochemical activity, choline acetyltransferase, is more extensively reduced in the nucleus (over 90%) than the loss of putative cholinergic perikarya (35%). Acetylcholinesterase histochemical activity was however substantially retained in individual neurones in the nucleus although virtually absent from the neocortex in SDAT. These abnormalities are consistent with a primary degeneration of cholinergic axons projecting to the cortex and secondary loss of perikarya from the subcortical nucleus. In contrast, preliminary observations on cases of Parkinson's disease suggest that the neuronal loss from the nucleus of Meynert may be greater in this disease than in SDAT, and previous studies have not consistently demonstrated a reduction in cortical choline acetyltransferase activities in Parkinson's disease. These observations, together with major differences in the neuropathology of the nucleus in SDAT and Parkinson's disease (neurofibrillary tangle and Lewy body formation, respectively) suggest that the involvement of the cholinergic system may differ in the two disease processes.  相似文献

Complete analysis of retinal tissue is difficult because it consists of a thin neural tissue spread across the back of a hemispheric surface. Conventional sectioning in a plane parallel to a central axis of symmetry produces a large number of samples, each containing only a small amount of the tissue of interest. Consequently, quantitative comparison of any feature of interest typically uses a small fraction of the sections from each retina, because analysis of the entire collection of sections is too time consuming. Such a sampling process can lead to misleading or erroneous conclusions. We present a new method which allows complete analysis of the retina using a small number of samples produced by sectioning flattened retinas. This procedure is straightforward as illustrated using an antibody against proliferating cell nuclear antigen (PCNA) to locate dividing cells in the teleost fish retina. Immunocytochemical staining on flat-sectioned retinas was quantified using a computer-based image analysis system. When the cells of interest are randomly distributed, conventional sampling procedures can seriously under- or over-estimate their number. The new technique presented allows significantly more efficient examination and quantification of the entire retina as compared to conventional techniques.  相似文献

Four cases of papillary muscle rupture occurring in the setting of acute myocardial infarction are presented, which illustrate the following points: the diagnosis may not be apparent at presentation, a mitral regurgitant murmur may be absent, transesophageal echocardiography may establish the diagnosis when transthoracic echocardiography does not, and appropriate surgical correction can lead to excellent functional recovery.  相似文献

Nonanastomotic aneurysms of arterial prosthetic grafts are an infrequent but well-documented complication of arterial bypass surgery. Several cases of nonanastomotic aneurysm formation of the external velour-type Dacron graft have been reported. We present a case of an external velour ringed Dacron (Sauvage EXS) femoropopliteal bypass graft with multiple nonanastomotic aneurysms. An extensive review of the English literature shows this to be the first reported case of multiple aneurysms occurring in this type of prosthesis.  相似文献

Isometric peak twitch force (stimulation frequency 0.5/s; 29.5-30.5 degrees C) was correlated with sarcomere length in isolated papillary muscles of the rabbit. Sarcomere length was measured from photographic recordings (1.5 ms exposure time) performed at rest between contractions and at the time of isometric peak twitch force. The sarcomere length at rest was found to be relatively uniform throughout the preparation and to be linearly related to the overall muscle length within the range Lmax-0.85Lmax. The distribution of sarcomere lengths increased considerably as the muscle went from rest to activity. Studies of surface markers showed different degrees of shortening (or elongation) of individual segments along the length of the preparation. The mean resting sarcomere length at Lmax (the optimum muscle length for force production) was 2.44 +/- 0.01 micron (grand mean +/- S.E., 7 muscles). The means active sarcomere length at Lmax was 2.29 +/- 0.04 micron. Active force declined steeply as the muscle length was reduced below Lmax. At a resting sarcomere length of 2.0 micron, active force was approximately 1/3 of the maximum. The observed differences between the length-tension relat-onships in myocardium (twitch responses) and skeletal muscle (tetanic contractions) are discussed on the basis of a length dependency of the activation process in cardiac muscle.  相似文献
