Using the human lung epithelial WI26VA4 cell line, we investigated the capacity of fenspiride, an anti-inflammatory drug with anti-bronchoconstrictor properties, to interfere with histamine-induced intracellular Ca2+ increase and eicosanoid formation. Histamine and a histamine H1 receptor agonist elicited a rapid and transient intracellular Ca2+ increase (0-60 s) in fluo 3-loaded WI26VA4 cells. This response was antagonized by the histamine H1 receptor antagonist, diphenhydramine, the histamine H2 receptor antagonist, cimetidine, having no effect. Fenspiride (10(-7)-10(-5) M) inhibited the histamine H1 receptor-induced Ca2+ increase. In addition, histamine induced a biphasic increase in arachidonic acid release. The initial rise (0-30 s), a rapid and transient arachidonic acid release, was responsible for the histamine-induced intracellular Ca2+ increase. In the second phase release (15-60 min), a sustained arachidonic acid release appeared to be associated with the formation of cyclooxygenase and lipoxygenase metabolites. Fenspiride (10(-5) M) abolished both phases of histamine-induced arachidonic acid release. These results suggest that anti-inflammatory and antibronchoconstrictor properties of fenspiride may result from the inhibition of these effects of histamine.  相似文献

In this study, total coliforms, thermotolerant coliforms, Escherichia coli and groundwater nitrate concentration were monitored at 127 groundwater sampling points (only 62 water points for bacteriological parameters) located in the southern part of Abidjan District. Each water sampling location was sampled in March and July 2007, representing respectively the long dry season and the long wet season. Geostatistical methods were used to analyze the spatial variability of nitrates and the groundwater nitrate pollution risk. The maximum seasonal content of total coliforms and thermotolerant coliforms ranged from 400 to 1000 CFU/100 mL and from 200 to 500 CFU/100 mL respectively. Moreover, 94% of these locations presented traces of bacteriological contamination. This contamination was mainly recorded during the rainy period. The degree of correlation between bacterial abundance and chemical parameters is variable. Nitrites, ammonium and potassium favoured coliform abundance. In the 127 water points, groundwater nitrate concentrations ranged from 4 to 198 mg L?1 and were log-normally distributed in the study area. The groundwater contamination risk map indicated that the strongly urbanized west side of the site presented a high probability of exceeding the WHO drinking water standard (50 mg L?1NO3?).  相似文献

This paper presents a compact analytical model for the organic field-effect transistors(OFETs), which describes two main aspects, the first one is related to the behavior in above threshold regime, while the other corresponds to the below threshold regime.The total drain current in the OFET device is calculated as the sum of two components, with the inclusion of a smooth transition function in order to take into account both regions using a single expression. A genetic algorithm based approach(GA) is investigated as a parameter extraction tool in the case of the compact OFET model to find the parameters values from experimental data such as:mobility enhancement factor γ, threshold voltage VT h, subthreshold swing S, channel length modulation λ, and knee region sharpness m. The comparison of the developed current model with the experimental data shows a good agreement in terms of the transfer and the output characteristics. Therefore, the GA based approach can be considered as a competitive candidate compared to the direct method.  相似文献

Conjugation of the bioactive apelin-17 peptide with a fluorocarbon chain results in self-organization of the peptide into micelles. Fluorine NMR spectroscopy studies show that the fluoropeptide‘s micelles are monodisperse, while proton NMR indicates that the peptide moiety remains largely disordered despite micellization. A very fast exchange rate is measured between the free and micellar states of the peptide which enables the number of molecules present in the micelle to be estimated as 200, in agreement with values found by dynamic light scattering measurements.  相似文献

TiAl coupons were dipped in a low-concentration phosphoric acid solution, air dried, then heated up to 700 °C or 800 °C before discontinuous oxidation under laboratory air at these temperatures. At the end of the heating ramp, surfaces of TiAl samples were covered by a layer made of a pyrophosphate compound, resulting from reaction between the deposited acid and oxidized titanium. This layer strongly adhered to the substrate surface and allowed to greatly improve the resistance of TiAl to oxidation. After a certain time, a transition period occurred during which pyrophosphate compound disappeared to be changed into TiO2 rutile. For experiments carried on at 800 °C, it was shown that the phosphorus coming from the pyrophosphate group was present in this titanium oxide and that sample mass gain was still lowered. Performed analyses (XRD, SEM, EDS, μ-Raman spectroscopy) allowed to propose an overall mechanism to account for the “phosphorus effect”.  相似文献

The Gate All Around (GAA) MOSFET is considered as one of the most promising devices for downscaling below 50?nm. By surrounding the channel completely, the gate gains increased electrostatic control of the channel and short-channel-effects (SCEs) can be drastically suppressed. However, challenges still remain to resolve the important issues particularly concerning hot-carrier reliability and accurate device models for nanoscale circuit designs. Hot-carrier effects have been the major issues in the long-term stability of subthreshold performances in a nanoscale MOS transistor. In this paper we present a two-dimensional analytical analysis of the subthreshold behavior, subthreshold current and subthreshold swing, including the interfacial hot-carrier effects. The calculated results of the proposed approach match well with those of the 2-D numerical device simulator. The present work provides valuable design insights in the performance of nanoscale CMOS-based devices including hot-carrier degradation effects.  相似文献

A simple, fast and ecological analytical method using a semi-automatic fat extractor and HPLC-FLD (fluorescence detection) for determination of polycyclic aromatic hydrocarbon markers i.e. benzo(a)anthracene (BaA), chrysene (Chr), benzo(a)pyrene (BaP) and benzo(b)fluoranthene (BbF) in cocoa butter has been validated. Validation’s procedure performed out in concordance with French standard NF V03-110 (2010) was based on existing polycyclic aromatic hydrocarbon (PAH) determination methods in various smoked foodstuffs and edible vegetable oils. Determination of correlation coefficients for specific PAHs ranged from 0.9992 to 0.9998. Respective values of limits of detection were 0.010, 0.011, 0.033 and 0.029 μg kg?1 and those of quantification were 0.035, 0.038, 0.111 and 0.098 μg kg?1 for BaA, Chr, BbF and BaP. Both values of repeatability and intermediary precision tests coefficients of variation were less than 5%. Recovery scores of four PAH markers matched EU standard 836/2011 recommendations. Sum of four PAH markers (BaA, Chr, BbF, BaP) contents varied from 5.42?±?0.58 to 11.37?±?0.01 μg kg?1 whereas those of BaP was comprised between 0.26?±?0.00 and 1.75?±?0.13 μg kg?1 in 20 cocoa butter samples extracted from raw cocoa bean stored at Ivorian cocoa farmer levels.  相似文献

Cocoa beans are the principal raw material for chocolate manufacture. Moulds have an important place in the change in the quality of cocoa beans due to their role in the production of free fatty acids and mycotoxins, namely ochratoxin A (OTA). This study investigated the impact of the key post-harvest treatments, namely the fermentation and drying methods on OTA contamination of raw cocoa beans. Analytical methods for OTA detection were based on solid–liquid extraction, clean-up using an immunoaffinity column, and identification by reversed-phase HPLC with fluorescence detection. Of a total of 104 randomly selected cocoa samples analysed, 32% had OTA contents above 2 µg kg–1. Cocoa sourced from pods in a bad state of health had a maximum OTA content of 39.2 µg kg1, while that obtained from healthy pods recorded 11.2 µg kg–1. The production of OTA in cocoa beans increased according to the pod-opening delay and reached 39.2 µg kg–1 after an opening delay of 7 days after harvest, while 6.1 and 11.2 µg kg–1 were observed when pods were opened after 0 and 4 days. OTA production also seemed to depend considerably to the cocoa fermentation materials. When using plastic boxes for bean fermentation, the OTA production was enhanced and reached an average OTA content of about 4.9 µg kg1, while the raw cocoa treated in banana leaves and wooden boxes recorded 1.6 and 2.2 µg kg–1 on average respectively. In parallel, the OTA production was not really influenced by either the mixing or the duration of the fermentation or the drying materials.  相似文献
