Contaminated sites represent new ecological niches where historical pollution has originated an unusual microbial biodiversity. The knowledge of these microorganisms contributes to the discovery of new pathways and metabolic networks and may offer potential solutions for damaged areas. In the present work seven microbial consortia have been isolated from an abandoned mine of blend and galena (Ingurtosu, Italy) through a selection for resistance to zinc (tested up to 40 mM in solution). All the consortia were able to accumulate zinc and the best accumulator, named Ing5, has been studied for the following characteristics: resistance and accumulation of Zn, Cd, Hg, bioaccumulation mechanisms of Zn, and influence of Zn and Cd on the metabolic profile. The results indicate that the consortium Ing5 bears resistance systems for Cd and Hg as well as Zn and that, for some of the 5 isolates belonging to Ing5, the resistance thresholds are higher in consortium than in pure culture. The prevalent mechanism for zinc accumulation can be reasonably considered to be metabolism-dependent, inducible and regulated by metal concentrations. The study on the metabolic profile, carried out by the Biolog system, shows that Zn exerts a very low influence on the metabolic profile and that this influence can also be positive; Cd has a stronger negative influence but that, despite this, the consortium is able to maintain a wide metabolic potential in the presence of heavy metals. These features of Ing5 make it a good candidate for biotechnological applications and for further investigation of the degradation of organic pollutants in the presence of metals.  相似文献

The preparation of alginate films with suitable properties requires a two-step contact with reticulating agents: initially a weakly structured pre-film is formatted which is further crosslinked in a second stage by immersion in a more concentrated solution. The present work evaluated the effects of a combined crosslinking procedure using calcium and barium ions on the physical and morphological properties of alginate-based films containing natamycin as antimicrobial agent. The release behavior of natamycin in water was evaluated as well as the antimicrobial activity against four target microorganisms, which are common cheese product contaminants. Films attributes were affected by the type of ion used in the second stage while the natamycin release rate and the antimicrobial activity were influenced by the ion used in the first stage. Films crosslinked with Ba2+ in the first and Ca2+ in the second stage (Ba–Ca films) exhibited physical properties very similar to films crosslinked with calcium in both stages. Release kinetics of natamycin in water fitted well to Fick's second law diffusional model, with effective diffusivity values ranging from 0.40 × 10−11 to 1.74 × 10−11 cm2/s. Ba–Ca films presented the lowest natamycin diffusion coefficient and the smallest inhibition zone diameter against the four microorganisms tested.  相似文献

Tannin from “Acacia Mearnsii” usually called Mimosa, is largely employed in vegetal tanning of hide even if the quality of the leather is poorer than that obtained using chromium as tanning agents. However, taking into account the natural resources of this product and its reduced environmental impact, mimosa tannin has been modified to obtain leather having improved properties. Tannin was modified using formaldehyde, urea, a sulfonic acid and a sulfiting agent: The new products show a higher molecular weight than mimosa tannin but maintain a good solubility in water. Modified tannins were characterized through physicochemical and technological tests and evaluated as tanning agents. The properties of tanned leathers obtained using the new tannin products and mimosa were compared. Some of the polymers containing phenolsulfonic acid are good tannin agents, especially those with a suitable molecular weight. The technological and sensorial properties of the leather obtained using modified tannins (distension and strength of grain, tensile strength, elongation, and tear load) are better than those obtained using mimosa when an appropriate ratio of the reagents were employed. At the same time the feeling of the tanned leather was improved and the final products show fullness, elasticity, fluency, reactivity, and a thin and fine grain. Some of the tannins modified with naphtalensulfonic acid show poor tanning properties but good penetration in the leather suggesting their use as auxiliary sintans in the pretanning process. In this paper we tried to define the capillaroscopic pattern of anti phospholipid syndrome able to differentiate between the primary (PAPS) and the systemic lupus erythematosus-associated form (SLE-APS) and to be a predictive marker of thrombotic manifestations. Eight PAPS and five SLE-APS patients were studied. In each patient the evaluation was based on anti cardiolipin antibody levels, nailfold capillaroscopy, retinal fluorangiography and transcranial doppler sonography. Statistical analysis has been performed using chi 2 analysis. Morphological alterations of capillary loops, venular visibility and sludging of blood were often observed in both groups. While we found in higher prevalence a variability of capillary loop length in PAPS patients, the SLE-APL group significantly differed for the presence of microhaemorrhages (p < 0.001). When we evaluated the clinical history, a marked microcirculatory damage was related with the occurrence of thrombotic manifestations in the PAPS patients. Anti cardiolipin antibody levels, retinal fluorangiography and transcranial doppler sonography did not correlate with clinical history in either group. In conclusion, nailfold capillaroscopy can be usefully employed in the differentiation between primary and SLE-associated anti phospholipid syndrome, and it can help to identify the patients at higher risk of thrombotic disease.  相似文献

Waste stabilisation pond (WSP) technology offers some important advantages and interesting possibilities when viewed in the light of sustainable energy and carbon management. Pond systems stand out as having significant advantages due to simple construction; low (or zero) operating energy requirements; and the potential for bio-energy generation. Conventional WSP requires little or no electrical energy for aerobic treatment as a result of algal photosynthesis. Sunlight enables WSP to disinfect wastewaters very effectively without the need for any chemicals or electricity consumption and their associated CO(2) emissions. The energy and carbon emission savings gained over electromechanical treatment systems are immense. Furthermore, because algal photosynthesis consumes CO(2), WSP can be utilised as CO(2) scrubbers. The environmental and financial benefits of pond technology broaden further when considering the low-cost, energy production opportunities of anaerobic ponds and the potential of algae as a biofuel. As we assess future best practice in wastewater treatment technology, perhaps one of the greatest needs is an improved consideration of the carbon footprint and the implications of future increases in the cost of electricity and the value of biogas.  相似文献
