Intravenous inoculation of Salmonella enterica serovar Typhimurium into mice is a prime experimental model of invasive salmonellosis. The use of wild-type isogenic tagged strains (WITS) in this system has revealed that bacteria undergo independent bottlenecks in the liver and spleen before establishing a systemic infection. We recently showed that those bacteria that survived the bottleneck exhibited enhanced growth when transferred to naive mice. In this study, we set out to disentangle the components of this in vivo adaptation by inoculating mice with WITS grown either in vitro or in vivo. We developed an original method to estimate the replication and killing rates of bacteria from experimental data, which involved solving the probability-generating function of a non-homogeneous birth–death–immigration process. This revealed a low initial mortality in bacteria obtained from a donor animal. Next, an analysis of WITS distributions in the livers and spleens of recipient animals indicated that in vivo-passaged bacteria started spreading between organs earlier than in vitro-grown bacteria. These results further our understanding of the influence of passage in a host on the fitness and virulence of Salmonella enterica and represent an advance in the power of investigation on the patterns and mechanisms of host–pathogen interactions.  相似文献

When advising farmers on how to control Johne's disease in an infected herd, one of the main recommendations is to avoid feeding waste milk to calves and instead feed calf milk replacer (CMR). This advice is based on the assumption that CMR is free of viable Mycobacterium avium ssp. paratuberculosis (MAP) cells, an assumption that has not previously been challenged. We tested commercial CMR products (n = 83) obtained from dairy farms around the United States by the peptide-mediated magnetic separation (PMS)-phage assay, PMS followed by liquid culture (PMS-culture), and direct IS900 quantitative PCR (qPCR). Conventional microbiological analyses for total mesophilic bacterial counts, coliforms, Salmonella, coagulase-negative staphylococci, streptococci, nonhemolytic Corynebacterium spp., and Bacillus spp. were also performed to assess the overall microbiological quality of the CMR. Twenty-six (31.3%) of the 83 CMR samples showed evidence of the presence of MAP. Seventeen (20.5%) tested positive for viable MAP by the PMS-phage assay, with plaque counts ranging from 6 to 1,212 pfu/50 mL of reconstituted CMR (average 248.5 pfu/50 mL). Twelve (14.5%) CMR samples tested positive for viable MAP by PMS-culture; isolates from all 12 of these samples were subsequently confirmed by whole-genome sequencing to be different cattle strains of MAP. Seven (8.4%) CMR samples tested positive for MAP DNA by IS900 qPCR. Four CMR samples tested positive by both PMS-based tests and 5 CMR samples tested positive by IS900 qPCR plus one or other of the PMS-based tests, but only one CMR sample tested positive by all 3 MAP detection tests applied. All conventional microbiology results were within current standards for whole milk powders. A significant association existed between higher total bacterial counts and presence of viable MAP indicated by either of the PMS-based assays. This represents the first published report of the isolation of viable MAP from CMR. Our findings raise concerns about the potential ability of MAP to survive manufacture of dried milk-based products.  相似文献

Re-establishing a functional endothelium following endovascular treatment is an important factor in arresting neointimal proliferation. In this study, both histology (in vivo) and computational simulations (in silico) are used to evaluate neointimal growth patterns within coronary arteries along the axial direction of the stent. Comparison of the growth configurations in vivo and in silico was undertaken to identify candidate mechanisms for endothelial repair. Stent, lumen and neointimal areas were measured from histological sections obtained from eight right coronary stented porcine arteries. Two re-endothelialization scenarios (endothelial cell (EC) random seeding and EC growth from proximal and distal ends) were implemented in silico to evaluate their influence on the morphology of the simulated lesions. Subject to the assumptions made in the current simulations, comparison between in vivo and in silico results suggests that endothelial growth does not occur from the proximal and distal ends alone, but is more consistent with the assumption of a random seeding process. This may occur either from the patches of endothelium which survive following stent implantation or from attachment of circulating endothelial progenitor cells.  相似文献

The selection of product portfolios, processing routes and the combination of technologies to obtain a sustainable biorefinery design according to economic and environmental criteria represents a challenge to process engineering. The aim of this research is to generate a robust methodology that assists process engineers to conceptually optimise the environmental and economic performances of biorefinery systems. A novel economic value and environmental impact (EVEI) analysis methodology is presented in this paper. The EVEI analysis is a tool that emerges from the combination of the value analysis method for the evaluation of economic potential with environmental footprinting for impact analysis. The methodology has been effectively demonstrated by providing insights into the performance of a bioethanol plant as a case study. The systematisation of the methodology allowed its implementation and integration into a computer-aided process engineering (CAPE) tool in the spreadsheet environment.  相似文献

Soybean agglutinin (SBA) protein, also known as soybean lectin, is regarded as an anti‐nutrient due to its negative effect on the ability of monogastric animals to gain weight following consumption of raw soybean seed. Historically, SBA has been measured using a time‐consuming and cumbersome hemagglutination procedure. The objective of our research was to obtain a validated methodology that is precise and accurate in the measurement of SBA while allowing minimally equipped laboratories to effectively conduct the analysis, thus our focus was on evaluating an existing commercially available ELISA, an enzyme‐linked‐lectin‐assay (ELLA), and a hybrid ELISA/ELLA. A new ELLA technique that can detect and quantify lectins was chosen and modified specifically for the quantitation of SBA in soybean seed. The proposed ELLA methodology is similar to a standard sandwich ELISA, and uses polyacrylamide‐linked N‐acetylgalactosamine (Gal–NAc–PAA) for a capture phase and the biotinylated version (Gal–NAc–PAA–Biotin) for detection. Based upon the validation data, the ELLA method can precisely and accurately determine soybean lectin levels in soybean seed. The validated ELLA method was used to quantify SBA in nine commercial soybean varieties introduced between 1972 and 2008 and demonstrated that the natural variability of SBA is subject to the effects of genotype and environment.  相似文献

Sugar beet (Beta vulgaris L cv Great Western Sugar) was grown using the nutrient film technique with a half-strength modified Hoagland nutrient solution to determine its biomass yield and nutritional quality. After 6 months, storage root and foliage weights per plant were 493·1 g and 551·0 g, respectively. Sucrose content in the fresh storage root was 118·4 g kg−1 but was less than 10 g kg−1 in the fresh leaves and petioles. Some nutrients in the leaves and petioles were analysed to evaluate their potential as a leafy vegetable. Fresh leaf protein, total dietary fibre, mineral (Ca, Mg, Zn, Fe and K), vitamin (carotene, ascorbic acid and thiamine) and oxalic acid concentrations were similar to those of consumer-accepted green vegetables.  相似文献

To predict the response of polyethylene thin films subjected to stress for a long time, it is necessary to understand the influence of stress on either the relaxation modulus or creep compliance. Extensive testing has been conducted on 20-micron-thick samples of a particular linear low-density polyethylene film at temperatures from 23°C to −50°C. When reduced to creep compliance and compared with results from dynamic mechanical analysis (DMA), the influence of nonlinearities in the response function is apparent. However, the use of a two-step loading procedure has produced sufficient data to discriminate between the effect of stress on amplitude and time on the creep compliance. It has been found that a master curve of compliance generated by DMA equipment may be used in conjunction with certain nonlinear functions to accurately predict the response of the polyethylene. Perhaps of more importance is the observation that the principles of simple time-temperature superposition, commonly used with linear viscoelastic characterization, are insufficient for use with polyethylene films at most stress levels of interest.  相似文献
