Nonstarter lactic acid bacteria isolated from Argentinean cheeses were identified and characterized by focusing on their resistance to biological barriers, along with other physiological features of potential interest, in the search for future probiotic organisms. Lactobacilli were enumerated and isolated from semihard and soft cheeses made with multistrain Streptococcus thermophilus starters. Lactobacilli counts in 1-week-old cheeses were between 10(5) and 10(7) CFU/g and then reached 10(7) CFU/ g in all 1-month samples, while streptococci were always above 10(9) CFU/g. A total number of 22 lactobacilli isolates were retained, identified, and characterized by in vitro tests. Species identity was determined by carbohydrate metabolism and species-specific PCR assays. Genetic diversity was explored by random amplified polymorphic DNA (RAPD) PCR analysis. The Lactobacillus strains were assigned to the species L. casei, L. plantarum, L. rhamnosus, L. curvatus, L. fermentum, and L. perolens. All the strains studied tolerated 25 ppm of lysozyme, and most of them showed resistance to 0.3% bile. After incubation in gastric solution (pH 2.0), counts decreased by several log units, ranging from 3.2 to 7.0. The strains were able to grow in the presence of bile salts, but only three isolates were capable of deconjugation. The nonstarter lactobacilli that were assayed fermented the prebiotic substrates (especially lactulose and inulin). Some strains showed high cell hydrophobicity and beta-galactosidase activity, as well as inhibitory activity against pathogenic bacteria. It was concluded that most of the lactobacilli isolated in this study demonstrated resistance to biological barriers and physiological characteristics compatible with probiotic properties, which make them suitable for further research in in vivo studies aimed at identifying new probiotic organisms. 相似文献

In light of the increasing interest in the development of functional food, several researches have focused on the production of food grade emulsifiers of nutritional interest, especially enriched in the eicosapentaenoic (EPA) and docosahexaenoic acids (DHA). The aim of this paper is to make a review of the production of monoacylglycerols (MAG) and diacylglycerols (DAG) obtained from different feedstock, mainly fish oil. A section of this paper is dedicated to the raw materials used as feedstock for these emulsifiers production. The health benefits of these partial acylglycerols are outlined. The chemical and enzymatic methods for producing these esters of glycerol are discussed, focusing on glycerolysis reactions. Recent advances on the lipase-catalyzed production of these partial acylglycerols in alternative reaction media and systems are also reviewed. 相似文献

The present study was conducted in Lima Metropolitana to evaluate the prevalence of Shiga toxin-producing Escherichia coli (STEC) O157:H7 in raw beef, raw ground beef, soft cheese and fresh vegetables, sampled at different markets in the city. Between October 2000 and February 2001, 407 food samples were collected from different markets in the 42 districts of Lima Metropolitana. Samples were assayed for E. coli O157 by selective enrichment in modified Tryptic Soy Broth containing novobiocin, followed by immunomagnetic separation (IMS) and plating onto sorbitol MacConkey agar supplemented with cefixime and potassium tellurite. Fifty (12.3%) of 407 food samples resulted positive for E. coli O157 isolation (23 of 102 ground beef; 15 of 102 beef meat; eight of 102 soft cheese and four of 101 fresh vegetables). Thirty-five E. coli O157 isolates were further analysed for the presence of virulence genes. All 35 were positive by PCR for O157 rfbE, fliCh7, eae-gamma1 and ehxA genes. In addition, genes encoding Shiga toxins were detected in 33 of 35 isolates, five isolates (14%) encoded stx(1), stx(2), and 28 (80%) stx2 only. The isolates were of seven different phage types (PT4, PT8, PT14, PT21, PT34, PT54, and PT87) with three phage types accounting for 80% of isolates: PT4 (15 isolates), PT14 (8 isolates), and PT21 (5 isolates). Interestingly, the majority (31 of 35; 89%) of E. coli O157:H7 isolates characterized in this study belonged mainly to the phage types previously found in STEC O157:H7 strains associated with severe human disease in Europe and Canada. Pulsed-field gel electrophoresis (PFGE) of 32 isolates revealed 14 XbaI-PFGE groups (I to XIV) of similarity >85%, with 23 (72%) isolates grouped in five clusters. Some isolates from different districts presented a high clonal relatedness. Thus, PFGE group VIII clustered eleven strains from nine different districts. The broad range of PFGE subtypes found in this study demonstrates the natural occurrence of many genetic variants among STEC O157:H7 spread in Lima. 相似文献

This work presents a new adsorption technique where the adsorbent (powdered activated carbon-PAC) is in the form of suspended flocs formed with water-soluble polymer flocculants. Thus, the adsorption of a typical dye, methylene blue (MB), was studied onto polyacrylamide flocs of PAC (PACF) in a fluidized bed reactor. The technique is based on the fact that the adsorption capacity of PAC does not decrease after flocculation because the adsorbed polymer occupies only a few surface sites, in the form of trains, loops, and tails. Moreover, the adsorption was found to proceed through a rapid mass transfer of MB to the adsorbing PAC flocs, in the same extent as onto PAC. Because of the rapid settling characteristics of the aggregates formed, the two phase separations, loaded PAC and solution, become easier. Thus, the technique offers the advantages of conducting simultaneously both adsorption and solid/liquid separation all in one single stage. Results obtained showed that high MB removal values can be attained in a fluidized bed reactor (>90%) and that PACF presents a much higher adsorption capacity (breakthrough points) than granulated activated carbon (GAC) in the same adsorbing bed. It is believed that this technique highly broadens the potential of the use of powdered activated carbon or other similar ultrafine adsorbents. 相似文献

The plums used to produce a traditional candied product, “Ameixa d’Elvas”, were obtained from two orchards, Vila Viçosa (VV) and Cano (CA). These orchards were selected because the fruits were behaving differently: (1) The day of harvesting for candying, established by the total soluble solids and titratable acidity, was one week earlier in VV; (2) VV yielded candied plums with good texture properties whereas CA gave poor processed fruits. In order to understand the origin of these differences, fruits from both orchards were harvested on the day established as the harvesting day for VV (day 1) and for CA (day 8). Comparable texture properties were obtained in firmness, rigidity and deformation work between the VV fresh plums harvested on day 1 and the CA plums harvested on day 8 but these were lower in CA when the flesh was analysed separately, in accordance with the activities of pectin methyl esterase (PME), polygalacturonase (PG) and cellulase (Cel). The increase of the intercellular area of parenchyma cells and the decrease in cell area caused by boiling, which resulted in a pronounced loss of textural properties, were partially recovered after the immersion of the fruits in sucrose syrup. The CA plums harvested on day 8 had a more pronounced degradation with boiling and lower recovery of cell shape, size and textural characteristics than had those of VV harvested on day 1. Upon candying, similar properties were observed for the fruits harvested on the same day: good candied products were observed for VV and CA fruits harvested on day 1 and poor candied products were observed for VV and CA fruits harvested on day 8. This work shows that the characteristics of the flesh of the fresh fruits are key parameters in defining the textural properties of the candied plum. The establishment of the harvesting moment for candying should take into account the changes that occur in the flesh of the plums during ripening. 相似文献

The macromolecular structure of coals thermally treated at 35°C, 100°C and 150°C was investigated by dynamic penetrant transport in thin coal sections. The effects of temperature, carbon content in coal, and penetrant type on the transport mechanism were investigated. Dynamic swelling studies showed that penetrant transport into coal is a function of the average molecular weight between crosslinks, Mc. The penetrant transport mechanism at low activity is Fickian diffusion. The transport mechanism deviates from Fickian diffusion to anomalous transport, when the carbon content in coal and penetrant activity increase. Variations of the diffusion coefficients and relaxation constants were determined using a diffusion/relaxation coupled model. 相似文献

The rheological behavior of micellar solutions is analyzed under nonhomogeneous velocity and stress flow conditions. The framework is based on the extended irreversible thermodynamics and the transient network formulation coupled to the underlying kinetics embodying two relevant processes: formation of wormlike chains from a free micellar solution through a thermally activated process and their flow induced degradation. The second kinetic process consists in the formation of entanglements from the free wormlike chains and their flow‐induced breakage. These processes are modeled in a coupled kinetic scheme constituted by a set of reversible kinetic equations describing the evolution in average of the three microstates (free short rod‐like micelles, free wormlike chains, and entangled wormlike chains) that reflect the complexity of macromolecular interactions. The contribution of fecal pollution from dogs in urbanized areas can be significant and is an often underestimated problem. Microbial source tracking methods (MST) utilizing quantitative PCR of dog-associated gene sequences encoding 16S rRNA of Bacteroidales are a useful tool to estimate these contributions. However, data about the performance of available assays are scarce. The results of a multi-laboratory study testing two assays for the determination of dog-associated Bacteroidales (DogBact and BacCan-UCD) on 64 single and mixed fecal source samples created from pooled fecal samples collected in California are presented here. Standardization of qPCR data treatment lowered inter-laboratory variability of sensitivity and specificity results. Both assays exhibited 100% sensitivity. Normalization methods are presented that eliminated random and confirmed non-target responses. The combination of standardized qPCR data treatment, use of normalization via a non-target specific Bacteroidales assay (GenBac3), and application of threshold criteria improved the calculated specificity significantly for both assays. Such measures would reasonably improve MST data interpretation not only for canine-associated assays, but for all qPCR assays used in identifying and monitoring fecal pollution in the environment. 相似文献

Anaerobic digestion enables waste (water) treatment and energy production in the form of biogas. The successful implementation of this process has lead to an increasing interest worldwide. However, anaerobic digestion is a complex biological process, where hundreds of microbial populations are involved, and whose start-up and operation are delicate issues. In order to better understand the process dynamics and to optimize the operating conditions, the availability of dynamic models is of paramount importance. Such models have to be inferred from prior knowledge and experimental data collected from real plants. Modeling and parameter identification are vast subjects, offering a realm of approaches and methods, which can be difficult to fully understand by scientists and engineers dedicated to the plant operation and improvements. This review article discusses existing modeling frameworks and methodologies for parameter estimation and model validation in the field of anaerobic digestion processes. Previous earthquakes, such as the 2010 Maule earthquake in Chile, have demonstrated the need to establish suitable predictors of compressive or tensile strains in concrete or steel in reinforced concrete shear walls, which can provide limit states or confinement requirements. Slender walls are commonly controlled by flexural deformations that can be divided into elastic and inelastic components. This study provides calibrated expressions for the elastic and inelastic components of flexural deformations using a fiber model for slender walls. These expressions are obtained for rectangular and T‐shaped walls. The elastic component is dependent on the axial load and the boundary steel reinforcement ratio. The impact of wall coupling is investigated, which requires a correction for the elastic component. The investigation of the inelastic component is based on a plastic hinge model, in which the length of the plastic hinge is a function of the lateral inelastic drift of the wall among other parameters. The traditional linear inelastic curvature distribution over the wall height is also modified for cases with steel reinforcement with a long yield plateau or low strain hardening, which results in a larger curvature at the wall base.
