Positron emission tomography (PET) of the cerebral glucose metabolism has shown to be useful in the presurgical evaluation of patients with epilepsy. Between seizures, PET images using fluorodeoxyglucose (FDG) show a decreased glucose metabolism in areas of the gray matter (GM) tissue that are associated with the epileptogenic region. However, detection of subtle hypo-metabolic regions is limited by noise in the projection data and the relatively small thickness of the GM tissue compared to the spatial resolution of the PET system. Therefore, we present an iterative maximum-a-posteriori based reconstruction algorithm, dedicated to the detection of hypo-metabolic regions in FDG-PET images of the brain of epilepsy patients. Anatomical information, derived from magnetic resonance imaging data, and pathophysiological knowledge was included in the reconstruction algorithm. Two Monte Carlo based brain software phantom experiments were used to examine the performance of the algorithm. In the first experiment, we used perfect, and in the second, imperfect anatomical knowledge during the reconstruction process. In both experiments, we measured signal-to-noise ratio (SNR), root mean squared (rms) bias and rms standard deviation. For both experiments, bias was reduced at matched noise levels, when compared to post-smoothed maximum-likelihood expectation-maximization (ML-EM) and maximum a posteriori reconstruction without anatomical priors. The SNR was similar to that of ML-EM with optimal post-smoothing, although the parameters of the prior distributions were not optimized. We can conclude that the use of anatomical information combined with prior information about the underlying pathology is very promising for the detection of subtle hypo-metabolic regions in the brain of patients with epilepsy.  相似文献

Short oligonucleotides can selectively recognize duplexes by binding in the major groove thereby forming triplexes. Based on the success of our recently developed strategy for furan‐based crosslinking in DNA duplexes, we here investigated for the first time the use of the furan‐oxidation crosslink methodology for the covalent locking of triplex structures by an interstrand crosslink. It was shown that in a triplex context, although crosslinking yields are surprisingly low (to nonexistent) when targeting fully complementary duplexes, selective crosslinking can be achieved towards mismatched duplex sites at the interface of triplex to duplex structures. We show the promising potential of furan‐containing probes for the selective detection of single‐stranded regions within nucleic acids containing a variety of structural motifs.  相似文献

This article deals with a specific support mechanism exempting offshore wind power generators partially from their balancing responsibilities by means of a tolerance margin. This specific support mechanism was enforced in Belgium as from 2009 and is defended by its proponents in view of the lower power output predictability at offshore locations. Although policies accommodating offshore developments may be seen as important to tap better wind resources, this contribution stresses the importance of full balancing responsibility for variable renewables. After a detailed evaluation of the support mechanism and its impact on the balancing costs for wind power generators, the use of current applied production support mechanisms is recommended. These can be used to acquire the same financial effect without increasing market complexity and harming the operation of the balancing market. The first part of the study deals with the specific implementation of the tolerance margin in the Belgian context. Secondly, its underlying motivation is quantitatively assessed, namely the relatively higher offshore prediction errors. Finally, the total offshore subsidy resulting from the measure is determined. Expressed in €/MWh, this subsidy is currently determined at €1.4–1.7/MWh, which represents the required increase of production support in order to replace the regulation.  相似文献

Palladium (Pd)‐loaded poly‐hydromethylsiloxane (PHMS) microparticles of tunable size and elasticity are prepared in a capillary‐based coaxial flow‐focusing microfluidic device constructed using off‐the‐shelf components. Simultaneous droplet formation and chemical cross‐linking processes are performed by tuning the dilution of the cross‐linking catalyst in the annular flow of the microreactor, resulting in PHMS microparticles synthesized in a single step. The size of the elastomeric microparticles can be tuned by adjusting the flow rate ratio of the polymer and cross‐linker mixture to water, while the elasticity can be tuned by the polymer to cross‐linker ratio as well as the flow rate ratio of the polymer mixture to cross‐linking catalyst mixture. Microparticle elasticity is characterized by the degree of solvent uptake. Application of the synthesized PHMS microparticles in organic synthesis is demonstrated by producing monodispersed Pd‐loaded microparticles and utilizing them as microreaction vessels for continuous Suzuki‐Miyaura cross‐coupling in a Pd‐loaded microparticle‐packed bed reactor. © 2018 American Institute of Chemical Engineers AIChE J, 64: 3188–3197, 2018  相似文献

This preliminary study is devoted to the application of front-face fluorescence spectroscopy to the study of egg yolks during storage. A total of 79 eggs stored for 1, 2, 3, 4, 5, 9, 10, 12, 16, 18, 23, 25 and 29 days at room temperature were analysed. The fluorescence emission spectra of tryptophan residues (excitation: 290 nm; emission: 305–430 nm) of proteins and the excitation spectra of vitamin A (emission: 410 nm; excitation: 270–350 nm) were recorded directly on egg yolk samples. Factorial discriminant analysis (FDA) was used to classify the eggs according to their date after they were laid. Using tryptophan fluorescence spectra, correct classification was observed for 57.1 and 51.9% for the calibration and the validation sets, respectively. Better classification (94.9 and 91.4% of the calibration and validation samples, respectively) was obtained from the vitamin A fluorescence spectra. The first five principal components (PCs) of the principal component analysis (PCA) extracted from each data set (tryptophan and vitamin A fluorescence spectra) were pooled (concatenated) into a single-matrix and analysed by FDA. Correct classifications were obtained for 97.5% of the calibration and 96.3.1% of the validation spectra. The discrimination of the investigated egg yolks according to their storage time was excellent. It was concluded that the concatenation of different fluorescence spectra might be considered as a promising indicator of shell egg freshness when they are used in egg products.  相似文献

Nitroxoline, a well-known antimicrobial agent, has been identified in several independent studies, and on different molecular targets, as a promising candidate to be repurposed for cancer treatment. One specific target of interest concerns cathepsin B, a lysosomal peptidase involved in the degradation of the extracellular matrix (ECM), leading to tumor invasion, metastasis and angiogenesis. However, dedicated optimization of the nitroxoline core is needed to actually deliver a nitroxoline-based antitumor drug candidate. Within that context, 34 novel nitroxoline analogs were synthesized and evaluated for their relative cathepsin B inhibitory activity, their antiproliferative properties and their antimicrobial activity. More than twenty analogs were shown to exert a similar or even slightly higher cathepsin B inhibitory activity compared to nitroxoline. The implemented modifications of the nitroxoline scaffold and the resulting SAR information can form an eligible basis for further optimization toward more potent cathepsin B inhibitors in the quest for a clinical nitroxoline-based antitumor agent.  相似文献

The direct electrochemical synthesis of NH3 from nitrogen and water vapor without the use of a fossil carbon source is highly desired. This synthesis is a viable option to store energy and produce fertilizer precursors. Here, a new Pt-free membrane electrode assembly is presented. An electrochemical membrane reactor demonstrates the feasibility of co-generating NH3 and H2 directly from nitrogen and water vapor at ambient conditions. An unprecedented high NH3-specific current efficiency of up to 27.5% using Ti as cathodic catalyst is reported. The co-generation can be tuned by the balance of process parameters.  相似文献

Pastry products are produced from heterogeneous multilayered dough systems. The main ingredients are flour, water, fat and sugar for puff pastry, and the same plus yeast for fermented pastry. Key aspects in pastry production are (i) building laminated dough containing alternating layers of dough and bakery fat and (ii) maintaining this multilayered structure during processing to allow for steam entrapment for proper dough lift during baking. Although most authors agree on the importance of gluten and fat for maintaining the integrity of the different layers, detailed studies on their specific function are lacking. The exact mechanism of steam entrapment during dough lift and the relative contribution of water set free from the fat phase during baking also remain unclear. This review brings together current knowledge on pastry products and the factors determining (intermediate) product quality. Its focus is on flour constituents, fat, water, and (where applicable) yeast during the different production stages of pastry products. Future research needs are addressed as the knowledge on biochemical and physical changes occurring in flour constituents and other ingredients during pastry production and their effect on product quality is currently inadequate.  相似文献

We show the synthesis of an in vivo stable mercury compound with functionality suitable for radiopharmaceuticals. The designed cyclic bisarylmercury was based on the water tolerance of organomercurials, higher bond dissociation energy of Hg−Ph to Hg−S, and the experimental evidence that acyclic structures suffer significant cleavage of one of the Hg−R bonds. The bispidine motif was chosen for its in vivo stability, chemical accessibility, and functionalization properties. Radionuclide production results in 197(m)HgCl2(aq), so the desired mercury compound was formed via a water-tolerant organotin transmetallation. The Hg-bispidine compound showed high chemical stability in tests with an excess of sulfur-containing competitors and high in vivo stability, without any observable protein interaction by human serum assay, and good organ clearance demonstrated by biodistribution and SPECT studies in rats. In particular, no retention in the kidneys was observed, typical of unstable mercury compounds. The natHg analogue allowed full characterization by NMR and HRMS.  相似文献
