Infants, whose growth and development are highly dependent upon the endocrine system, are particularly vulnerable to endocrine disruptor exposure. In our study, we examined whether or not alkylphenols, bisphenol A, benzophenone, di(2-ethylhexyl)adipate and 10 phthalates are present in containers of baby food. Furthermore, the migration into food-simulating solvents was also examined. Sixteen types of containers of baby food, obtained on the Japanese market in 2001, were examined: 8 glass bottle types, 1 plastic type, 1 metallic package (laminated with plastic) and 6 multilayered laminated film packages. Alkylphenols, benzophenone, bisphenol A and di(2-ethylhexyl)adipate were not detected in any of the samples. On the other hand, dibutylphthalate (DBP) was detected in 7 samples (1.2-9.1 micrograms/sample), and di(2-ethylhexyl)phthalate (DEHP) was detected in 15 samples (1.3-18 micrograms/sample). Migration tests were carried out for 5 samples, and the levels of DBP and DEHP that migrated into n-heptane were 0.68-1.7 micrograms/sample and 0.33-3.5 micrograms/sample, respectively. From these results, a baby's intake levels of DBP and DEHP were estimated to be about 1/2,500 and 1/90-300 of the tolerable daily intake (TDI), respectively. 相似文献

To assess the role of protein kinase Cbeta (PKCbeta) in human myometrial contractions during pregnancy, we evaluated the effect of a PKCbeta inhibitor (LY333531) on the pregnant and nonpregnant myometrial contractions and compared the level of PKCbeta in the pregnant myometrium with that in the nonpregnant myometrium. The effects of LY333531 on the myometrial contractions were examined by measuring contractile activity (frequency and amplitude). PKCbeta in human myometrium was assessed at mRNA level using real-time PCR method. The characteristics of contractile activity were different between the pregnant and the nonpregnant myometrium. The amplitude of rhythmic contractions in the preterm and term myometrium was increased 2- to 2.5-fold when compared with that in the nonpregnant myometrium, but the frequency of rhythmic contractions was decreased by about half. LY333531 (10(-6) M) reduced the increased amplitude in the preterm and term myometrium by about 50%, and the inhibitory effects of LY333531 in the pregnant myometrium were significantly greater than that in the nonpregnant myometrium (about 50 vs 25%). However, the frequency in the pregnant and nonpregnant myometrium was not influenced by LY333531. Real-time PCR revealed a significant, five- to sevenfold increase in the expression of PKCbeta mRNA in the preterm and term myometrium when compared with the nonpregnant myometrium. These findings suggest that the increased amplitude of human myometrial contractions during pregnancy is related to the increased level of PKCbeta. Daily intakes of 12 phenols which are possible endocrine disruptors were estimated in hospital meals from 2000 to 2001. 4-Nonylphenol (4-NP (mix)) and bisphenol A (BPA) were detected at levels of 5.0 to 19.4 ng/g and 0.2 to 1.1 ng/g, respectively. 4-tert-Butylphenol, 4-n-pentylphenol, 4 -tert-pentylphenol, 4-n-hexylphenol, 4-n-heptylphenol and 4-tert-octylphenol were detected at levels of 0.1 to 2.4 microg/g. The daily intakes of 4-NP (mix) and BPA were 5.8 microg/day and 0.42 microg/day, respectively. The daily intakes of other phenols were less than 1 microg/day. 相似文献

A monoacylglycerol lipase (MGL) was purified from Pseudomonas sp. LP7315 by ammonium sulfate precipitation, anion-exchange chromatography, and preparative electrophoresis. The purified enzyme was homogeneous on SDS-PAGE with a molecular mass of 59 kDa. Its hydrolytic activity was confirmed to be specific for monoglycerides: the enzyme did not hydrolyze di- and triglycerides. MGL was found to be stable even after 1-h incubation at 65 degrees C. The optimum pH for monopalmitin hydrolysis was approximately 8. The hydrolytic activity depended not only on temperature and pH but also on the type of monoglyceride used. MGL also catalyzed monoglyceride synthesis at 65 degrees C in a solvent-free two-phase system, in which fatty acid droplets were dispersed in the glycerol phase with a low water content. The synthetic reaction proceeded at a constant rate for approximately 24 h and approximately reached an equilibrium after 48 h of reaction. The initial rate and equilibrium yield of the synthetic reaction depended on the type of fatty acid used as the substrate. 相似文献

A method and results of static and dynamic analysis of Pascal programs are described. In order to investigate characteristics of large systems programs developed by the stepwise refinement programming approach and written in Pascal, several Pascal compilers written in Pascal were analysed from both static and dynamic points of view. As a main conclusion, procedures play an important role in the stepwise refinement approach and implementors of a compiler and designers of high level language machines for Pascal-like languages should pay careful attention to this point. Cholesterol oxidase (CHO) with high stability in detergents was found from an isolated strain, Y-134, belonging to the gamma-subclass of Proteobacteria. CHO production reached its maximum by incubation at 30 degrees C for 12 d. It was purified from cell-free extract prepared by mixing the cells with 0.4% Triton X-100. The absorption spectrum of the purified enzyme exhibited maxima at 274 and 410 nm, and a shoulder at 330 nm. The molecular mass was 115 kDa with two identical subunits of 58 kDa. The enzyme oxidized cholest-5-en-3beta-ol (cholesterol) and 5alpha-cholestan-3beta-ol (dihydrocholesterol) at a high reaction rate, and the K(m) value for cholesterol was 65 microM. The stability of the enzyme was higher than other CHOs in nonionic detergents with high values of hydrophilelipophile balance (HLB) such as Triton X-450 and sodium cholate. NH2-terminal sequence analysis showed a high similarity to CHO from Burkholderia cepacia, but not to CHOs from Streptomyces or Brevibacterium. 相似文献

Levels of p-t-butylphenol (PTBP), p-t-octylphenol (PTOP), p-nonylphenol (NP) and bisphenol A (BPA) in rubber products in food-contact use, infant-related use and other uses are reported. Of 60 rubber products tested, PTOP was found in three in a concentration range of 2.2-37 μg g-1, and NP in 15 in a concentration range of 2.6-513 μg g-1. PTBP and BPA were not found in any sample. PTOP and NP were not detected in any infant-related sample, regardless of the material type. In contrast, among the samples with food contact and with other uses, the percentages containing PTOP or NP were 33 and 43%, respectively. A migration test was performed on eight samples containing NP and used in food-contact environments. The migration level of NP from spatulas to n-heptane was 0.83-1.52 μg ml-1. The average migration rates to water, 20% ethanol and n-heptane were 0.03, 0.7 and 6.6%, respectively. 相似文献
