Leucine modulates synthetic and degradative pathways in muscle, possibly providing metabolic benefits for both athletes and diseased populations. Leucine has become popular among athletes for improving performance and body composition, however little is known about the metabolic effects of the commonly consumed leucine‐derived metabolite β‐hydroxy‐β‐methyl butyrate (HMB). Our work measured the effects of HMB on metabolic protein expression, mitochondrial content and metabolism, as well as lipid content in skeletal muscle cells. Specifically, cultured C2C12 myotubes were treated with either a control or HMB ranging from 6.25 to 25 μM for 24 h and mRNA and/or protein expression, oxygen consumption, glucose uptake, and lipid content were measured. Contrary to leucine's stimulatory effect on metabolism, HMB‐treated cells exhibited significantly reduced regulators of lipid oxidation including peroxisome proliferator‐activated receptor alpha (PPARα) and PPARβ/δ, as well as downstream target carnitine palmitoyl transferase, without alterations in glucose or palmitate oxidation. Furthermore, HMB significantly inhibited activation of the master regulator of energetics, AMP‐activated protein kinase. As a result, HMB‐treated cells also displayed reduced total mitochondrial content compared with true control or cells equivocally treated with leucine. Additionally, HMB treatment amplified markers of lipid biosynthesis (PPARγ and fatty acid synthase) as well as consistently promoted elevated total lipid content versus control cells. Collectively, our results demonstrate that HMB did not improve mitochondrial metabolism or content, and may promote elevated cellular lipid content possibly through heightened PPARγ expression. These observations suggest that HMB may be most beneficial for populations interested in stimulating anabolic cellular processes.  相似文献

The bottromycins are a family of highly modified peptide natural products, which display potent antimicrobial activity against Gram‐positive bacteria, including methicillin‐resistant Staphylococcus aureus. Bottromycins have recently been shown to be ribosomally synthesized and post‐translationally modified peptides (RiPPs). Unique amongst RiPPs, the precursor peptide BotA contains a C‐terminal “follower” sequence, rather than the canonical N‐terminal “leader” sequence. We report herein the structural and biochemical characterization of BotP, a leucyl‐aminopeptidase‐like enzyme from the bottromycin pathway. We demonstrate that BotP is responsible for the removal of the N‐terminal methionine from the precursor peptide. Determining the crystal structures of both apo BotP and BotP in complex with Mn2+ allowed us to model a BotP/substrate complex and to rationalize substrate recognition. Our data represent the first step towards targeted compound modification to unlock the full antibiotic potential of bottro‐ mycin.  相似文献

A wave of recent cross-national research has pointed to the positive consequences for countries with high levels of “quality of government” (QoG), broadly defined, such as corruption, impartiality, and quality of public services. Yet the question of how QoG varies at the sub-national level is still widely overlooked, in particular with measures that are available over time. To address it, we present the third round of data from the regional European Quality of Government Index (EQI) survey corruption (D73), Europe (N44) governance (H11); sub-national (R50), time series (C22), collected in 2017 and built upon the opinions of 78,000 respondents in 193 regions from 21 European countries. The data provides several contributions to the literature. First, while the majority of QoG-type indices rely on expert assessments, the EQI relies on the assessments of citizens, who are the on-the-ground consumers of public services. Second, the data begins to show trends on QoG variation over time, as well as across European regions. Consequently, this data is the most comprehensive sub-national data to date; mapping of QoG within and across EU countries over the past decade. Building on previous rounds of data collected in 2010 and 2013, the 2017 EQI, which is published free for scholarly use, builds on both perceptions and experiences of citizens in public service areas such as health care, education, and law enforcement. This paper presents the results of the latest survey, improved with respect to the previous ones, discussion of trends across space and over time, as well as interesting avenues for future research that we detect across European regions.  相似文献

The enantioselective oxidation of 2° alcohols to ketones is an important reaction in synthetic chemistry, especially if it can be achieved using O2-driven alcohol oxidases under mild reaction conditions. However to date, oxidation of secondary alcohols using alcohol oxidases has focused on activated benzylic or allylic substrates, with unactivated secondary alcohols showing poor activity. Here we show that cholesterol oxidase (EC 1.1.3.6) could be engineered for activity towards a range of aliphatic, cyclic, acyclic, allylic and benzylic secondary alcohols. Additionally, since the variants demonstrated high (S)-selectivity, deracemisation reactions were performed in the presence of ammonia borane to obtain enantiopure (R)-alcohols.  相似文献

Biomaterials of either natural or synthetic origin are used to fabricate implantable devices, as carriers for bioactive molecules or as substrates to facilitate tissue regeneration. For the design of medical devices it is fundamental to use materials characterized by non-immunogenicity, biocompatibility, slow and/or controllable biodegradability, non-toxicity, and structural integrity. The success of biomaterial-derived biodevices tends to be based on the biomimetic architecture of the materials. Recently, proteins from natural precursors that are essentially structural and functional polymers, have gained popularity as biomaterials. The silks produced by silkworms or spiders are of particular interest as versatile protein polymers. These form the basis for diverse biomedical applications that exploit their unique biochemical nature, biocompatibility and high mechanical strength. This review discusses and summarizes the latest advances in the engineering of silk-based biomaterials, focusing specifically on the fabrication of diverse bio-mimetic structures such as films, hydrogels, scaffolds, nanofibers and nanoparticles; their functionalization and potential for biomedical applications.  相似文献

The goal of this work was to investigate the effect of two stage pressure molding on the compression molding of a sheet molding compound (SMC). It has been shown in previous studies that a rapid drop in pressure during SMC curing significantly reduced severity of sink marks. This study concentrated on a method of predicting the optimum time during curing to release pressure by examining material behavior through process data from in-mold sersors. A simple control scheme was them applied for automatic pressure release at the optimum time corresponding with the peak of the material expansion and the onset of the reaction exotherm.  相似文献

The present study aims to compare the oxidative stress biomarkers, pro-inflammatory cytokines, and histological changes induced by three cardiovascular risk factors, namely, hypertension, dyslipidemia, and type 1 diabetes mellitus. Hypertension was induced with 40 mg/kg body weight (b.w.) of N omega-nitro-L-arginine-methyl (L-NAME) administered orally. Dyslipidemia was induced by the administration of a diet with a high cholesterol (2%) content. Diabetes mellitus was induced by intraperitoneal administration of a single dose of streptozocin (65 mg/kg). Malondialdehyde (MDA) and total oxidative status (TOS) are increased by all three cardiovascular risk factors (up to 207%). The indirect assessment of NO synthesis (NOx) is observed to be reduced after L-NAME administration (43%), and dyslipidemia induction (16%), while type 1 diabetes mellitus is associated with the highest levels of NOx (increased 112%). Hypertension, dyslipidemia, and type 1 diabetes reduced the total antioxidative capacity (TAC) and total thiol (SH) levels (up to 57%). The values of evaluated pro-inflammatory cytokines, tumour necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-1β (IL-1β), assessed from the ascending aorta were elevated by all three cardiovascular risk factors, with the highest levels induced by type 1 diabetes mellitus (up to 259%). The histopathological examination of the ascending and descending aorta revealed reversible pro-atherogenic changes consisting of the accumulation of lipid droplets in the subendothelial connective tissue on rats with hypertension and dyslipidemia. Irreversible pro-atherogenic changes consisting of a reduction of the specific elasticity of the arteries were observed in rats with type 1 diabetes mellitus. Type 1 diabetes mellitus demonstrates an alteration of the oxidative stress parameters, the elevation of tissue levels of the pro-inflammatory cytokines and causing irreversible pro-atherogenic changes on the aortic wall.  相似文献

Tics manifest as brief, purposeless and unintentional movements or noises that, for many individuals, can be suppressed temporarily with effort. Previous work has hypothesized that the chaotic temporal nature of tics could possess an inherent fractality, that is, have neighbour-to-neighbour correlation at all levels of timescale. However, demonstrating this phenomenon has eluded researchers for more than two decades, primarily because of the challenges associated with estimating the scale-invariant, power law exponent—called the fractal dimension Df—from fractional Brownian noise. Here, we confirm this hypothesis and establish the fractality of tics by examining two tic time series datasets collected 6–12 months apart in children with tics, using random walk models and directional statistics. We find that Df is correlated with tic severity as measured by the YGTTS total tic score, and that Df is a sensitive parameter in examining the effect of several tic suppression conditions on the tic time series. Our findings pave the way for using the fractal nature of tics as a robust quantitative tool for estimating tic severity and treatment effectiveness, as well as a possible marker for differentiating typical from functional tics.  相似文献
