BACKGROUND: Strawberries are nutritive fruits and a source of antioxidants. We evaluated antioxidant properties of ‘Camino Real’ strawberries grown in the Brazilian savannah, harvested in different seasons. Analytical and meteorological data were analyzed by partial least squares regression. RESULTS: Fruits from May showed the lowest contents of total phenolics (1789.78 mg kg?1 fresh weight (FW)), catechin (21.37 mg kg?1 FW), quercetins (4.89 mg kg?1 FW) and total ellagic acid (208.68 mg kg?1 FW) and the lowest antioxidant activity by 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) (11.39 mg Butylated hydroxytoluene (BHT) eq. g?1 FW) and ferric reducing antioxidant power (FRAP) (22.01 mg ferrous sulfate eq. g?1 FW) assays. Strawberries harvested in July presented the lowest concentrations of total (190.61 mg kg?1 FW) and individual anthocyanins (73.88 mg kg?1 FW and 5.96 mg kg?1 FW for pelargonidin‐3‐glucoside and cyanidin‐3‐glucoside, respectively), but the highest contents of vitamin C (685.47 mg kg?1 FW), DPPH (18.87 mg BHT eq. g?1 FW) and FRAP (39.30 mg ferrous sulfate eq. g?1 FW). The highest contents of free ellagic acid (26.11 mg kg?1 FW), pelargonidin‐3‐glucoside (291.82 mg kg?1 FW) and cyanidin‐3‐glucoside (11.84 mg kg?1 FW) were found in strawberries from September. Rain in the previous 30 days to harvest influenced negatively many phenolics and antioxidant activity of strawberries harvested in May. In July, longer photoperiod and lower temperature at 30 days previous to harvest probably led to higher antioxidant activity and vitamin C. Increased photoperiod and temperature at the final stage of maturation seem to raise pigments and free ellagic acid in strawberries. CONCLUSION: It was possible to observe significant relationships among meteorological and antioxidant variables for strawberries grown in the Brazilian savannah. We investigated the effect of increased glucose oxidase concentration as a technological option to decrease oxidative stress during the processing of probiotic yogurts. Probiotic yogurts were produced with increased concentrations of glucose oxidase (0, 250, 500, 750, or 1,000 mg/kg) and submitted to physicochemical and microbiological analysis at 1, 15, and 30 d of refrigerated storage. Higher concentrations of glucose oxidase (750 and 1,000 mg/kg) and a longer storage time were found to have an influence on the characteristics of the probiotic yogurt, contributing to more extensive postacidification, an increase in the dissolved oxygen level, and higher proteolysis. In addition, increased production of aroma compounds (diacetyl and acetaldehyde) and organic acids (mainly lactic acid) and a decrease in the probiotic bacteria count were reported. The use of glucose oxidase was a feasible option to minimize oxidative stress in probiotic yogurts. However, supplementation with excessive amounts of the enzyme may be ineffective, because insufficient substrate (glucose) is present for its action. Consumer tests should be performed to evaluate changes in the sensory attributes of the probiotic yogurts with increased supplementation of glucose oxidase. In addition, packaging systems with different permeability to oxygen should be evaluated.  相似文献

The present work reports the development and validation of an analytical method for lactic acid quantification in milk by ion exclusion-HPLC with UV detection. This method showed good precision (variation coefficient <7.0%) and good accuracy (97.6-99.6%). Calibration curves were linear (R(2)>0.996) at the tested range of 5.0-25.0mmolL(-1); and the detection and quantification limits were 0.5 and 1.0mmolL(-1), respectively. Analysis of lactic acid in fermented milk (FM) or NaOH neutralised fermented milk (FNM) was performed using samples obtained during 9h of fermentation. An ANOVA analysis combined with the Tukey test was used to show that the method was insensitive to the differences between FM and FNM. The proposed method therefore proved to be a useful quality control tool in laboratories because it was able to determine milk acidity even in adulterated samples.  相似文献

A differential amperometric method for the specific determination of ascorbic acid in honey was developed by association of a flow injection analysis (FIA) system and a tubular reactor containing the ascorbate oxidase enzyme immobilised. A gold electrode modified by electrochemical deposition of palladium was employed as working electrode. Ascorbic acid was quantified in seven samples of commercial honeys using a potential of +0.60 V vs. Ag/AgCl(sat). The linear dynamic range in ascorbic acid extends from 1 to 50 μmol L−1, at pH 7.0. At flow rate of 1.5 mL min−1 and injecting 250 μL sample volumes, a sampling frequency of 180 determinations per hour is afforded. The detection and quantification limit of this method are 0.14 and 0.49 μmol L−1, respectively. The samples analyses were compared with the volumetric method, and showed an excellent correlation between the methods.  相似文献

This study aimed at the optimization of the production of chitosanase in solid culture. Trichoderma koningii sp., an entomopathogenic fungus, was used to produce chitosanase under solid-state fermentation using a mixture of wheat bran and chitosan. The incubation period; addition of moistening water and culture medium composition were optimized. The protocol to extract the enzyme was also optimized. The optimal conditions for chitosanase production by T. koningii were obtained using a mixture of 3.0 g of wheat bran and 1.5 g of chitosan, with the addition of 2.5 mL of moistening water (pH 5.5) and of 2.5 mL of saline solution (pH 5.5) containing NaNO3 (1.0 g/L), (NH4)2HPO4 (1.0 g/L), MgSO4.7H2O (1.0 g/L), and NaCl (1.0 g/L). Optimal enzyme extraction was carried out adding 20 mL of sodium acetate buffer (200 mM, pH 5.5) at 30 °C under orbital agitation at 150 rpm for 6 min. In this paper we propose a new post-processing deblocking technique that is independent of the compression method used to encode the image. The development of this filter was motivated by the use of Multidimensional Multiscale Parser (MMP) algorithm, a generic lossy and lossless compression method. Since it employs an adaptive block size, it presents some impairments when using the deblocking techniques presented in the literature. This led us to the development of a new and more generic deblocking method, based on total variation and adaptive bilateral filtering.The proposed method was evaluated not only for still images, but also for video sequences, encoded using pattern matching and transform based compression methods. For all cases, both the objective and subjective quality of the reconstructed images were improved, showing the versatility of the proposed technique.  相似文献

Composite hydrogels of poly(acrylamide‐co‐acrylate) with rice husk ash (RHA) were synthesized and studies of the swelling variables were accomplished comparatively with commercial polyacrylamide gel and PAMACRYL, a poly(acrylamide‐co‐acrylate) hydrogel without RHA. FT‐IR and WAXS were the techniques employed for characterizing a series of hydrogel obtained by varying the percentage of RHA (1, 2, 5, 10, and 20 wt %) and the amount of crosslinking agent (0.05, 0.1, and 0.2 mol %) relative to sum of AAm and AAc. Superabsorbent hydrogel with Weq > 800 g H2O/g gel was obtained with percentage of 10 wt % of RHA and 0.1 of crosslinking agent mol %. The hydrogel showed to be sensitive to the pH variation and to the presence of salts. The hydrogels, even though submitted through cycles of drying and swelling, preserved their superabsorbent characteristics and demonstrated better water absorbance properties when compared with commercial polyacrylamide gel. The composite hydrogels of poly(acrylamide‐co‐acrylate) with RHA presented good characteristics to be applied as soil conditioner for using in agriculture. © 2011 Wiley Periodicals, Inc. Edessa meditabunda is a secondary pest within the piercing-sucking stink bug complex that attacks soybean crops in Brazil. The behavioral responses of males and females to aeration extracts from conspecifics suggested the presence of a male-produced sex pheromone. Gas chromatographic (GC) analysis of male and female aeration extracts revealed the presence of two male-specific compounds in a ratio of 92:8. Gas chromatographic -electroantennographic detection (GC-EAD) assays indicated that the major component is bioactive for females, supporting the behavioral data. Analysis of the mass and infrared spectra of the male-specific compounds suggested that they were both methyl-branched long-chain methyl esters. On the basis of the mass spectra of the respective hydrocarbons obtained by micro derivatizations, the structures of these methyl esters were proposed to be methyl 4,8,12-trimethylpentadecanoate (major) and methyl 4,8,12-trimethyltetradecanoate (minor). An 11 step synthetic route that was based on a sequence of Grignard reactions, starting from cyclopropyl methyl ketone, was developed to obtain synthetic standards with a 7.9 % overall yield for the major compound and a 9.9 % yield for the minor. The synthetic standards co-eluted with the natural pheromones on three different GC stationary phases. Y-tube olfactometer assays showed that the synthetic standards, including the major compound alone and a mixture of the major and minor compounds in the proportion found in natural extracts, were strongly attractive to females.  相似文献

Phthia picta is part of a complex of true bugs (Heteroptera) in Brazil that attack tomatoes, being particularly damaging because nymphs and adults feed on both leaves and fruit. Gas chromatography (GC) of aeration extracts of adult males vs. females revealed the presence of a male-specific compound. GC-electroantennographic detector experiments indicated that the antennae of females are highly sensitive to this male-specific compound. GC-mass spectrometry and GC-FTIR analyses suggested a methyl branched hydrocarbon structure for this compound. After synthesis of three different proposed structures, the natural product was indentified as 5,9,17-trimethylhenicosane, which was strongly attractive to females in Y-tube olfactometer bioassays. Analysis of dissected body parts of adults revealed that the pheromone is produced in the lateral accessory glands of the metathoracic scent gland of males only.  相似文献

Rice bran oil was obtained from rice bran by solvent extraction using ethanol. The influence of process variables, solvent hydration (0-24% of water, on mass basis), temperature (60-90 °C), solvent-to-rice bran mass ratio (2.5:1 to 4.5:1) and stirrer speed (100-250 rpm) were analysed using the response surface methodology.The extraction yield was highly affected by the solvent water content, and it varied from 8.56 to 20.05 g of oil/100 g of fresh rice bran (or 42.7-99.9% of the total oil available) depending on the experimental conditions. It was observed that oryzanol and tocols behave in different ways during the extraction process. A larger amount of tocols is extracted from the solid matrix in relation to γ-oryzanol. It was possible to obtain values from 123 to 271 mg of tocols/kg of fresh rice bran and 1527 to 4164 mg of oryzanol/kg of fresh rice bran, indicating that it is feasible to obtain enriched oil when this renewable solvent is used. No differences in the chemical composition of the extracted oils were observed when compared to the data cited in the literature.  相似文献
