We have reported recently that the dual specificity mitogen-activated protein kinase phosphatase-3 (MKP-3) elicits highly selective inactivation of the extracellular signal-regulated kinase (ERK) class of mitogen-activated protein (MAP) kinases (Muda, M., Theodosiou, A., Rodrigues, N., Boschert, U., Camps, M., Gillieron, C., Davies, K., Ashworth, A., and Arkinstall, S. (1996) J. Biol. Chem. 271, 27205-27208). We now show that MKP-3 enzymatic specificity is paralleled by tight binding to both ERK1 and ERK2 while, in contrast, little or no interaction with either c-Jun N-terminal kinase/stress activated protein kinase (JNK/SAPK) or p38 MAP kinases was detected. Further study revealed that the N-terminal noncatalytic domain of MKP-3 (MKP-3DeltaC) binds both ERK1 and ERK2, while the C-terminal MKP-3 catalytic core (MKP-3DeltaN) fails to precipitate either of these MAP kinases. A chimera consisting of the N-terminal half of MKP-3 with the C-terminal catalytic core of M3-6 also bound tightly to ERK1 but not to JNK3/SAPKbeta. Consistent with a role for N-terminal binding in determining MKP-3 specificity, at least 10-fold higher concentrations of purified MKP-3DeltaN than full-length MKP-3 is required to inhibit ERK2 activity. In contrast, both MKP-3DeltaN and full-length MKP-3 inactivate JNK/SAPK and p38 MAP kinases at similarly high concentrations. Also, a chimera of the M3-6 N terminus with the MKP-3 catalytic core which fails to bind ERK elicits non selective inactivation of ERK1 and JNK3/SAPKbeta. Together, these observations suggest that the physiological specificity of MKP-3 for inactivation of ERK family MAP kinases reflects tight substrate binding by its N-terminal domain. 相似文献

A typical groundwater remedation problem is studied by using a combined simulation-optimization model. The management procedure employs groundwater flow and contaminant transport simulation models in conjunction with linear and quadratic programming techniques. The methodology is applied to the hydrodynamic control of a contaminant plume that has to be stabilized and removed by a system of pumping wells. The paper focuses mainly upon a sensitivity analysis to the aquifer transmissivity. The effect of changes in the transmissivities of a zoned aquifer upon the optimal solutions of the management problem is examined by considering the optimal pumping rates, the time to remediation and the pumped groundwater volume as the key output variables of the remediation strategies. In addition, the influence of the dispersivities and the imposed hydraulic gradient upon the same output variables is critically evaluated. The results of the study illustrate the need for uncertainty reduction in the knowledge of the hydrogeologic parameters. 相似文献

The application of green roofs on urban buildings is considered to have a positive impact on their thermal behaviour and local microclimatic conditions. According to the literature, their ability for attenuation of storm water run-off as well as their contribution to the building's thermal protection is among the most important benefits of this technique. However, despite the development of computer models that can assist towards analysing the nature of their behaviour, there is still a relative gap in measured data representing long-term period thermal performance. In this paper, the results of a long-term experimental analysis are presented, which attempt to identify the thermal behaviour of a green roof in comparison with a bare flat roof. The results show that in Mediterranean countries, a green roof can contribute substantially to building's energy conservation mainly during the warm period of the year, while its influence during the cold period is negligible. 相似文献

The impact of thermal bridges on the energy demand of buildings with double brick wall constructions
The implementation of the European Directive on the Energy Performance of Buildings (EPBD) is a milestone towards the improvement of energy efficiency in the building sector. However, even in cases where impressive measures can be implemented in the densely built urban environment, the less glamorous measure of building's envelope thermal insulation remains a prerequisite towards the improvement of the building's energy efficiency. Despite the insulation requirements specified by national regulations, thermal bridges in the building's envelope remain a weak spot in the constructions. Moreover, in many countries construction practices tend to implement only partially the insulation measures foreseen by regulations. As a result, thermal losses are in practice greater than those predicted during the design stage. This paper presents a study on representative wall thermal insulation configurations used in Greek buildings, in order to investigate the impact of the thermal bridges on the energy consumption. The double wall construction, used widely in Greece and not only there, is rather susceptible to the occurrence of thermal bridges, in contrast to a typical thermal insulating façade, like the one applied in Central Europe. Schools are the most suitable type of building for the application of energy efficiency and good indoor air quality measures. This is justified by the fact that such measures can promote sustainability to the future citizens, and even more, ensure a comfortable and healthy environment for educational purposes. Unfortunately, in practice school buildings face the same, or even more intense, energy performance and indoor air quality problems as any other building. The purpose of this study is to investigate the energy efficiency, thermal environment and indoor air quality in public nursery and elementary school buildings in the city of Kozani, located at the cold climatic zone of Greece. The survey, conducted both by in-field measurements and by questionnaires, reveals the main parameters affecting the overall performance of the investigated buildings. The problematic building envelope, the improper control of heating and lighting systems, the absence of proper legislative measures and, above all, the lack of interest concerning the efficiency of such buildings are the main factors in the reported efficiency. 相似文献

We present a comprehensive set of theoretical values for differential, total, and momentum transfer cross sections for elastic scattering of electrons by inert-gas atoms, for energies below 1 keV. In addition, we also provide Sherman function values for the xenon atom at various energies. The present work includes a critical analysis of most available experimental and theoretical data, and systematic investigations on the effect of various choices for the atomic central, exchange, and polarization potential that we used in our calculations. The objective of the present work is to employ a relatively simple and semiempirical approach with a minimum number of adjustable parameters, which could generate a reliable set of cross sections for a wide range of energies.
