Aerosol matrix-assisted laser desorption/ionization (MALDI) has been combined with a reflectron time-of-flight mass spectrometer for improved mass resolution. A methanol solution of matrix and analyte was sprayed directly into a reflectron time-of-flight mass spectrometer, and the aerosol particles were irradiated and ionized with a frequency-tripled Nd:YAG laser at 355 nm. Mass resolution of over 300 was observed for the peptides bradykinin, angiotensin II, and gramicidin D and for vitamin B(12). This represents a resolution enhancement of approximately 10-fold over that previously reported for aerosol MALDI with a linear time-of-flight instrument. 相似文献

The design of a diffractive phase element (DPE) that simultaneously implements wavelength demultiplexing and focusing is carried out on the basis of the general theory of amplitude-phase retrieval. The designed DPE is fabricated with optical contact lithography. Three masks are needed to produce the surface-relief structure of the DPE with eight quantized levels in depths. Experiments demonstrate that the designed DPE can successfully implement both the functions of demultiplexing three different-wavelength beams and focusing each component at a predesignated position simultaneously. In the shadow-moiré system, the period of the grating is varied by rotation of the grating, so the phase of the moiré pattern is changed as well. By the selection of suitable rotation angles, three images at different positions of the grating are acquired to obtain the absolute distance from the object to the grating. A theoretical analysis is presented for the method, and some experiments have been done to verify the theoretical analysis. The results show that the method is fast and the accuracy is better than 10 μm. The measurable range is directly proportional to the period of the grating and inversely proportional to the angles at which the grating is rotated. 相似文献

Pancreatic cancer is a highly aggressive and treatment-refractory cancer. A clinically-relevant animal model is necessary to develop therapy for metastatic pancreatic cancer. In this study we evaluated the efficacy of mitomycin C (MMC) and 5-FU against the human pancreatic adenocarcinoma cell line PAN-12 in an orthotopic human metastatic pancreatic cancer nude mice model. The model is constructed by surgical orthotopic implantation (SOI) of histologically intact tumor tissue in the tail portion of the pancreas near the spleen. PAN-12 grew very aggressively in the control group of nude mice with extensive local invasion and distant metastasis to various organs with a propensity for the lung but to other organs as well, including the liver, kidney and regional and distant lymph nodes. In a striking effect none of the mice in the MMC-treated group developed tumor. Although mice in the 5-FU treated group survived statistically significantly longer than those in the untreated control, the overall incidence of metastasis in these mice was equivalent to those in the control. However no liver or kidney metastases were found in the 5-FU treated animals perhaps accounting in part for their longer survival. This "clinical" nude mouse model of highly metastatic pancreatic cancer can now be used to discover new effective agents for this disease. 相似文献

As (where 0.1 × 1) alloys on GaAs (001) and (110) were carried out using transmission electron microscopy (TEM) and high resolution X-ray diffraction (XRD). Three different strain relief mechanisms related to the formation of misfit dislocations (MDs) were observed. An experiment on fitness of one-piece casting osseointegrated implant frameworks was performed among three fixture abument positions where other conditions were controlled. The result revealed that there was statistically significant differences in dimensional change among the three fixture abument positions. In terms of the relation between abument position and fitness, the more backward the abument position was, the worse the osseointegrated framework fitness was. The in situ formed Al2O3, TiB2 and Al3Ti mixture-reinforced aluminium composites were successfully fabricated by the reaction sintering of the TiO2-B-Al system in a vacuum. With increasing boron content in the TiO2-B-Al system, the amount of generated TiB2 in the composites increased and Al3Ti content decreased. At the same time the distribution uniformity of the in situ formed Al2O3 and TiB2 particulates was obviously improved, and the size of the Al3Ti particles was reduced. The in situ Al2O3 and TiB2 particulates had sizes from 0.096–1.88 m. The interface between the in situ formed particulates and the aluminium matrix was clean, and no consistent crystallographic orientation relationship was found. The strength and elastic modulus of the composites was significantly improved by lowering the Al3Ti content. When the boron content in the TiO2-B-Al system rose, the morphology of the tensile fracture surface of the composites was changed from large fractured Al3Ti blocks and fine dimples, to fine dimples and pulled-out particulates. The track structure model of heavy ion cross sections was developed by Katz and co-workers in the 1960s. In this model the action cross section is evaluated by mapping the dose-response of a detector to gamma rays (modeled from biological target theory) onto the radial dose distribution from delta rays about the path of the ion. This is taken to yield the radial distribution of probability for a "hit" (an interaction leading to an observable end-point). Radial integration of the probability yields the cross section. When different response from ions of different Z having the same stopping power is observed this model may be indicated. Since the 1960s there have been several developments in the computation of the radial dose distribution, in the measurement of these distributions, and in new radiobiological data against which to test the model. The earliest model, by Butts and Katz made use of simplified delta ray distribution functions, of simplified electron range-energy relations, and neglected angular distributions. Nevertheless it made possible the calculation of cross sections for the inactivation of enzymes and viruses, and allowed extension to tracks in nuclear emulsions and other detectors and to biological cells. It set the pattern for models of observable effects in the matter through which the ion passed. Here we outline subsequent calculations of radial dose which make use of improved knowledge of the electron emission spectrum, the electron range-energy relation, the angular distribution, and some considerations of molecular excitation, of particular interest both close to the path of the ion and the outer limits of electron penetration. These are applied to the modeling of action cross sections for the inactivation of several strains of E-coli and B. subtilis spores where extensive measurements in the "thin-down" region have been made with heavy ion beams. Such calculations serve to test the radial dose calculations at the outer limit of electron penetration. We lack data from which to test these calculations in regions close to the path of the ion aside from our earliest work on latent tracks in plastics, though it appears that the criterion then suggested for the threshold of track formation, of a minimal dose at a minimal distance (of about 20 angstroms, in plastics), remains valid. 相似文献

Heterozygous mutations in the gene for the Kit transmembrane receptor have been identified recently in human piebaldism and mouse "dominant spotting." Interestingly, all of the 14 known missense mutations that cause depigmentation in these species map to the tyrosine kinase domain of the receptor, whereas none have involved the extracellular ligand-binding domain. In an attempt to detect these uncommon mutations, we screened the nine exons encoding the extracellular portion of Kit for single-strand conformation polymorphisms (SSCP) in eight piebald subjects previously reported to be negative for kinase mutations. Four of these eight kindreds proved to carry novel mutations. The first mutation, found in two apparently unrelated probands with mild piebaldism and English ancestry, substitutes an arginine for a highly conserved cysteine at codon 136. This substitution disrupts a putative disulfide bond required for formation of the second Ig-like (D2) loop of the Kit ligand-binding domain. The second mutation, detected in a piebald kindred characterized by unusually limited depigmentation, substitutes a threonine for an alanine at codon 178, a site just proximal to conserved cysteines at codons 183 and 186. The third mutation, occurring in a kindred with more extensive depigmentation, is a novel four-base insertion in exon 2 that results in a proximal frameshift and premature termination. The data strongly suggest that piebaldism can result from missense mutations in the Kit ligand-binding domain, although the resulting phenotype may be milder than that observed for null or kinase mutations. The apparent clustering of these uncommon mutations at or near the conserved cysteines for the D2 Ig-like loop further suggests a critical role for this region in Kit receptor function. 相似文献
