Two experiments were conducted to examine the effects of cooking parameters, including final internal temperature (65, 72·5 or 80°C), on the eating quality of pork. Assessments were made by a 10-member trained taste panel. In Experiment 1 on loin steaks (m. longissimus) from 28 carcasses weighing 66 ± 1·9 (SD) kg, increasing the final internal temperature from 65 to 80°C had the following effects on mean scores (1-8): tenderness -1·0; juiciness -1·5, pork flavour +0·6 and abnormal flavour -0·5. The medium temperature of 72·5°C was suggested as ideal. In Experiment 2 on leg roasts (m. gluteobiceps) from 96 carcasses weighing 65 + 2·7 (SD) kg, increasing the final internal temperature from 65 to 80°C had the following effects on mean scores (1-8): tenderness -0·2 (not significant), juiciness -0·6, pork flavour +0·3 and abnormal flavour -0·5. The effects of final internal temperature were therefore smaller in roasts and temperatures towards the upper end of the range would be recommended for ideal eating quality, balancing small losses in tenderness and juiciness against gains in flavour scores. Although the experiments were not designed to examine source and sex effects there was evidence that tenderness can be improved through the use of Duroc genes and pelvic suspension of carcasses and no evidence of differences in eating quality between entire males and females.  相似文献

Organic and conventional lamb loin chops, labelled as British lamb, were bought from three major UK supermarket chains (designated A, B and C) in the Bristol area on 10 occasions over a six week period. Samples (n=360) were from unknown production systems but representative of what is available to UK consumers. The nutritional quality of muscle was assessed in terms of its fatty acid composition and eating quality was assessed by a trained sensory panel. Lamb prices varied between £9 and £12.50 per kg, with a relatively modest price differential between organic and conventional lamb chops of £1.10, £1.88 and £1.16 £/kg for supermarkets A, B and C, respectively. On average, organic chops were 20g heavier than conventional chops. Chops were relatively lean, having just 14% of subcutaneous fat, approximately half that of a similar survey 10 years ago. Organic lamb had a better eating quality than conventional lamb in terms of juiciness (p<0.05), flavour (p<0.05) and overall liking (p<0.05) thus providing some evidence for the perception among consumers that organic products 'taste better'. Differences in juiciness were attributed to the higher intramuscular fat content of organic meat whilst differences in flavour were attributed to differences in fatty acid composition, in particular, the higher level of linolenic acid (18:3) and total n-3 PUFA in organic chops. Conventional chops had a higher percentage of linoleic acid (18:2). Chops from both productions systems had a favourable n-6:n-3 ratio. The most important difference between the three supermarkets was that lamb flavour was significantly lower in chops from supermarket A, probably due to differences in their 'display until' dates. The sequence has been determined of 80 888 bp of contiguous subtelomeric DNA, including the isp5 gene, from the right arm of chromosome I of Schizosaccharomyces pombe; 27 open reading frames (ORFs) longer than 100 codons are present, giving a density of one gene per 3.0 kb. Seven of the predicted proteins are members of the major facilitator superfamily (MFS) of transport proteins, including four amino acid permease homologues, bringing this family of amino acid permease sequences to 17 in Sz. pombe, and a phylogenetic analysis is presented. Also encoded is an allantoate permease homologue, a sulphate permease homologue and a probable urea active transporter. Predicted non-membrane proteins include a 1-aminocyclopropane-1-carboxylate deaminase (ACC deaminase), a class III aminotransferase, serine acetyltransferase, protein-L-isoaspartate O-methyltransferase, alpha-glucosidase, alpha-galactosidase, esterase/lipase, oxidoreductase of the short-chain dehydrogenase/reductase (SDR) family, aldehyde dehydrogenase, formamidase, amidase, flavohaemoprotein, a putative translation initiation inhibitor and a protein with similarity to a filamentous fungal conidiation-specific protein. The remaining six ORFs are likely to encode proteins, either because they have sequence similarity with hypothetical proteins or because they are known to be transcribed. Introns are scarce in the sequenced region: only three ORFs contain introns, with only one having multiple introns. The sequenced region also contains a single Tf1 transposon long terminal repeat (LTR). The sequence is derived from cosmid clones c869, c922 and c1039 and has been submitted to the EMBL database under entries SPAC869 (Accession No. AL132779), SPAC922 (AL133522) and SPAC1039 (AL133521).  相似文献

The Solvent Extraction Residual Biotreatment (SERB) technology was evaluated at a former dry cleaner site in Jacksonville, FL, where an area of tetrachloroethylene (PCE) contamination was identified. The SERB technology is a treatmenttrain approach for complete site restoration, which combines an active in situ dense nonaqueous-phase liquid (DNAPL) removal technology, cosolvent extraction, with a passive enhanced in situ bioremediation technology, reductive dechlorination. During the in situ cosolvent extraction test, approximately 34 kL of 95% ethanol/5% water (v:v) was flushed through the contaminated zone, which removed approximately 60% of the estimated PCE mass. Approximately 2.72 kL of ethanol was left in the subsurface, which provided electron donorfor enhancement of biological processes in the source zone and downgradient areas. Quarterly groundwater monitoring for over 3 yr showed decreasing concentrations of PCE in the source zone from initial values of 4-350 microM to less than 150 microM during the last sampling event. Initially there was little to no daughter product formation in the source zone, but after 3 yr, measured concentrations were 242 microM for cis-dichloroethylene (cis-DCE), 13 microM for vinyl chloride, and 0.43 microM for ethene. In conjunction with the production of dissolved methane and hydrogen and the removal of sulfate, these measurements indicate that in situ biotransformations were enhanced in areas exposed to the residual ethanol. First-order rate constants calculated from concentration data for individual wells ranged from -0.63 to -2.14 yr(-1) for PCE removal and from 0.88 to 2.39 yr(-1) for cis-DCE formation. First-order rate constants based on the change in total mass estimated from contour plots of the groundwater concentration data were 0.75 yr(-1) for cis-DCE, -0.50 yr(-1) for PCE, and -0.33 yr(-1) for ethanol. Although these attenuation rate constants include additional processes, such as sorption, dispersion, and advection, they provide an indication of the overall system dynamics. Evaluation of the groundwater data from the former dry cleaner site showed that cosolvent flushing systems can be designed and utilized to aid in the enhancement of biodegradation processes at DNAPL sites.  相似文献

通过混凝土经历10℃~-40℃、10℃~-80℃和10℃~-160℃等3种典型的超低温温度区间冻融循环作用试验,探讨不同的温度区间和冻融循环作用次数对混凝土弹性模量的影响。结果表明,随冻融循环作用次数的增加,上限温度时因冻融损伤累积使混凝土的弹性模量呈逐渐软化趋势,其中下限温度较低温度区间的变化趋势最为明显;下限温度时因混凝土内孔隙水结冰使混凝土弹性模量呈先增大后减小趋势,且较上限温度时的变化幅度更大。不同温度区间的混凝土相对弹性模量差以及上下限温度时单次冻融软化指标和累积冻融软化指标均存在明显的差异且变化规律较为复杂。该文还由试验结果拟合出混凝土相对弹性模量与超低温冻融循环作用的温度区间和次数间的关系式。所获得的这些结果可为LNG储罐类混凝土结构的设计和安全性能评估提供参考。  相似文献

This study concerns the effect of simple surface treatments on the nature of the oxide layer, of commercially pure titanium (cpTi) and Ti6Al4V alloy substrates and their effect on human osteoblast cells (HOBS). After treatment the surfaces were analyzed by X-ray photoelectron spectroscopy (XPS) in order to identify the surface groups responsible for the cell attachment process. The assessment of cell attachment was monitored by the Alamar blue assay (AB), measuring cell activity, in three types of media: phosphate-buffered saline (PBS), serum containing and serum-free Dulbecco's modified Eagle's cell culture medium (SER+ and SERF respectively). XPS analysis of the treated surfaces revealed consistent peaks representative of TiO2 on all surfaces and Ti0 and Ti2O3 on the non-heat-treated surfaces. The cell activity assays indicated that there were no significant differences in cellular activity caused by surface treatments, but the cellular activity compared between the three types of medium was greatest in the PBS over the initial stages of attachment.  相似文献
