A series of pyrazolo[3,4-d]pyrimidines, previously found to be Src inhibitors, was tested for their ability to inhibit proliferation of three Bcr-Abl-positive human leukemia cell lines (K-562, KU-812, and MEG-01), on the basis of the experimental evidence that various Src inhibitors are also active against Bcr-Abl kinase (the so called dual Src/Abl inhibitors). They reduce Bcr-Abl tyrosine phosphorylation and promote apoptosis of the Bcr-Abl-expressing cells. A cell-free enzymatic assay on isolated c-Abl confirmed that such compounds directly inhibit Abl activity. Finally, molecular modeling simulations were also performed to hypothesize the binding mode of the compounds into the Abl binding site.  相似文献

A computational fluid dynamic (CFD) model for the pressure vessel of the evolutionary pressurized reactor (EPR™) was developed and validated. The aim of this model is the simulation of transients where three-dimensional effects play a strong role, such as boron dilution and main steam line break (MSLB) scenarios. First, a full solid (CAD) model has been built, that includes all details of the reactor pressure vessel (RPV) and the internals which are important for fluid dynamic analyses. The solid model has then been used as basis for the generation of the computational mesh necessary to carry out CFD simulations. Both a hexahedral and a polyhedral mesh have been created. The CFD model has been validated against experimental results of the JULIETTE facility, a 1:5 scaled mock-up of the EPR™ reactor RPV built by AREVA and equipped with advanced instrumentation.The performances of the hexahedral and the polyhedral meshes are investigated in relation to the agreement with experimental data, convergence and CPU requirements. In addition, the effect of the cold-leg swirls on the velocity field inside the RPV is investigated. These swirls mimic the effects of the main coolant recirculation pumps on the flow field at the entrance of the RPV. It is shown that the CFD model is able to capture the shift of the maximum velocity in the downcomer annulus observed in the experimental results. Good qualitative as well as quantitative agreement with the experimental data is achieved.  相似文献

We provide a validated and rapid protocol for the solubilization of amyloid β-peptide (Aβ). This procedure involves sequential solubilization using structure-breaking organic solvents hexafluoroisopropanol and DMSO followed by column purification. The low solubility and tendency of Aβ to aggregate considerably impede the in vitro handling and biophysical or biological investigation of Aβ, despite the interest in this peptide because of its implication in Alzheimer's disease. The main advantage of the proposed protocol over others is that it results in standardized aggregate-free Aβ peptide samples that are biocompatible for cell culture studies and yield reproducible aggregation kinetics and cytotoxicities. This three-step protocol also enables the co-solubilization of the longer Aβ42 variant with Aβ40 in ratios relevant to Alzheimer's disease.  相似文献

The development of fluorescent biolabels for specific targeting and controlled drug release is of paramount importance in biological applications due to their potential in the generation of novel tools for simultaneous diagnosis and treatment of diseases. Dopamine is a neurotransmitter involved in several neurological diseases, such as Parkinson's disease and attention deficit hyperactivity disorder (ADHD), and the controlled delivery of its agonists already proved to have beneficial effects both in vitro and in vivo. Here, we report the synthesis and multiple functionalization of highly fluorescent CdSe/CdS quantum rods for specific biolabeling and controlled drug release. After being transferred into aqueous media, the nanocrystals were made highly biocompatible through PEG conjugation and covered by a carbohydrate shell, which allowed specific GLUT-1 recognition. Controlled attachment of dopamine through an ester bond also allowed hydrolysis by esterases, yielding a smart nanotool for specific biolabeling and controlled drug release.  相似文献

The full characterization of the optical properties of nanofluids consisting of single-wall carbon nanohorns of different morphologies in aqueous suspensions is carried out using a novel spectrophotometric technique. Information on the nanofluid scattering and absorption spectral characteristics is obtained by analyzing the data within the single scattering theory and validating the method by comparison with previous monochromatic measurements performed with a different technique. The high absorption coefficient measured joint to the very low scattering albedo opens promising application perspectives for single-wall carbon nanohorn-based fluid or solid suspensions. The proposed approximate approach can be extended also to other low-scattering turbid media.  相似文献

In 2001, samples of liver (n = 38) and perirenal fat (n = 25) (only in males) were taken from European roe deer (Capreolus capreolus) populating a hilly, wooded protected area in the Emilia-Romagna region of Northern Italy. Samples were analyzed for organochlorine pesticides dichlorodiphenyltrichloroethane (DDT) and its metabolites, hexachlorobenzene (HCB), hexachlorocycloexane alpha-, beta- and gamma-isomers (alpha-, beta- and gamma-HCH), alpha-endosulfan, aldrin, dieldrin and endrin, and the seven PCB 'indicator' congeners IUPAC Nos. 28, 52, 101, 118, 138, 153 and 180, in order to investigate the current state of contamination of the area and to assess the possible toxicological risks for the monitored species. p,p'-DDE was detected in 95% of the liver and perirenal fat samples (n = 63), at concentrations ranging from < dl to 629.3 ng/g lipid weight, which were much higher than those found for HCB and gamma-HCH. The concentrations of the other measured organochlorine pesticides and metabolites were below the detection limit in all the samples. PCBs were found in 97% of the liver and perirenal fat samples, at total levels ranging between < dl to 414.5 ng/g lipid weight. The hepta- and hexa-chlorinated congeners 180, 153 and 138 were predominant and comprised more than 70% of the total seven PCBs in both the liver and perirenal fat samples. In the male roe deer, significantly higher concentrations of HCB, p,p'-DDE and PCBs were found in the liver than in the perirenal fat (P < 0.001). Hepatic levels of p,p'-DDE and PCBs were significantly higher in males than in females (P < 0.05 and P < 0.001, respectively). No significant differences in organochlorine concentrations were observed between juvenile and adult males in either tissue sample (P > 0.05). Females were not statistically tested because they were all adults. The levels of organochlorine contaminants were well below those associated with adverse reproductive effects and lethality in mammals.  相似文献

The adaptation of inkjet printing technology for the realisation of controlled micro- and nano-scaled biological structures is of great potential in tissue and biomaterial engineering. In this paper we present the Olivetti BioJet system and its applications in tissue engineering and cell printing. BioJet, which employs a thermal inkjet cartridge, was used to print biomolecules and living cells. It is well known that high stresses and forces are developed during the inkjet printing process. When printing living particles (i.e., cell suspensions) the mechanical loading profile can dramatically damage the processed cells. Therefore computational models were developed to predict the velocity profile and the mechanical load acting on a droplet during the printing process. The model was used to investigate the role of the stiffness of the deposition substrate during droplet impact and compared with experimental investigations on cell viability after printing on different materials. The computational model and the experimental results confirm that impact forces are highly dependent on the deposition substrate and that soft and viscous surfaces can reduce the forces acting on the droplet, preventing cell damage. These results have high relevance for cell bioprinting; substrates should be designed to have a good compromise between substrate stiffness to conserve spatial patterning without droplet coalescence but soft enough to absorb the kinetic energy of droplets in order to maintain cell viability.  相似文献

The study is aimed at developing a new cereal-based product, with increased nutritional quality, by using natural fermentation of blends of chestnut and rye flour. In spite of the remarkable similarity, the technological potential of combinations of both flours has never been explored before. Three spontaneous chestnut/rye sourdough fermentations were performed over a period of twelve days with daily back-slopping. Samples taken at all refreshment steps were used for culture-dependent and culture-independent evaluation of the microbiota present. Dominant species basically overlapped to those associated to sourdoughs strengthened with chestnut flour, such as Pediococcus pentosaceus or Weissella paramesenteroides. Microstructures, evaluated by means of Scanning Electron Microscopy, revealed the presence in chestnut sourdoughs of a distinguishable network surrounding starch granules, while rye flour-added sourdoughs showed a less structured matrix. By gas chromatography coupled to mass spectrometry, 51 volatile organic compounds were identified at 24 h and after prolonged fermentation. Within volatile organic compounds, alcohols, esters, acids, aldehydes and ketones, all well-known flavour compounds in sourdough fermentation, appeared as dominant. The PCA discriminated the sourdoughs into three distinct clusters and highlighted a clear influence of fermentation time on the volatile composition of sourdoughs.  相似文献

This study investigated the possibility of using yeast strains in fermented milks to obtain products with high Angiotensin I-converting enzyme (ACE) inhibitory activity and low bitter taste. Ninety-three yeast strains isolated from Colombian Kumis in different geographic regions were molecularly identified, and their milk fermentation performances were determined. Molecular identification evidenced that Galactomyces geotrichum, Pichia kudriavzevii, Clavispora lusitaniae and Candida tropicalis, were the dominant species. Eighteen out of 93 strains produced fermented milk with ACE-inhibitory (ACEI) activity values ranging from 8.69 to 88.19%. Digestion of fermented milk samples by pepsin and pancreatin demonstrated an increase in ACEI activity, with C. lusitaniae KL4A as the best producer of ACEI peptides. Moreover, sensory analysis of the products containing the major ACE-inhibitory activity pointed out that P. kudriavzevii KL84A and Kluyveromyces marxianus KL26A could be selected as potential adjunct starter cultures in Kumis, since they made a considerable contribution to the ACE inhibitory activity and produced fermented milk without bitter taste. In this study we observed that Colombian Kumis can be an excellent vehicle for the isolation of yeasts with a potential to enhance bioactive peptides produced during milk fermentation.  相似文献
