The preparation of thinned lamellae from bulk samples for transmission electron microscopy (TEM) analysis has been possible in the focussed ion beam scanning electron microscope (FIB‐SEM) for over 20 years via the in situ lift‐out method. Lift‐out offers a fast and site specific preparation method for TEM analysis, typically in the field of materials science. More recently it has been applied to a low‐water content biological sample (Rubino 2012). This work presents the successful lift‐out of high‐water content lamellae, under cryogenic conditions (cryo‐FIB lift‐out) and using a nanomanipulator retaining its full range of motion, which are advances on the work previously done by Rubino (2012). Strategies are explored for maintaining cryogenic conditions, grid attachment using cryo‐condensation of water and protection of the lamella when transferring to the TEM. Microsc. Res. Distributed real-time applications have presented the need to extend the Object Management Group's Common Object Request Broker Architecture (CORBA) standard to support real-time. This paper describes a Dynamic Real-Time CORBA system, which supports the expression and enforcement of end-to-end timing constraints as an extension to a commercial CORBA system. The paper also describes performance tests that demonstrate the system's ability to enforce expressed timing constraints.  相似文献

Milk fat fractions from supercritical carbon dioxide (SC-CO2) extraction were compared with commercial melt crystallization (MC) fractions for their physical and chemical properties. The fractions were analyzed for fatty acids, triacylglycerols, cholesterol, total carotenoid content, and volatile compounds. The fractions were also evaluated for solid fat content (SFC) by pulsed nuclear magnetic resonance and thermal profiles by differential scanning calorimeter (DSC). The distribution of fatty acids and triacylglycerols in the fractions depended on the fractionation technique used. SC-CO2 separated fractions based on molecular weight rather than on melting point, which is the driving force for the MC process. The differences among the fractions were quantified from their SFC and DSC curves. Triacylglycerol profiles by high-performance liquid chromatography showed that the SC-CO2 fractions were distinctly different from each other and from MC fractions. The SC-CO2 solid fraction (super stearin) was the most unique. It had a high concentration of long-chain, unsaturated fatty acid-containing triacylglycerols in a narrow range of high molecular weight, indicating a homogeneity of this fraction that has not been attainable by other techniques. It was also enriched in β-carotene and was devoid of volatile compounds. As compared to liquid MC fractions, the liquid SC-CO2 fraction had a high concentration of low-melting triacylglycerols and was enriched in volatile compounds. With SC-CO2, it is thus possible to simultaneously fractionate and produce a flavor-rich concentrate at no extra processing cost.  相似文献

The environmental support hypothesis postulates that it may be possible to reduce older adults' deficits in episodic memory by providing environmental support at the encoding and/or retrieval phases. To examine the validity of this hypothesis, we varied the amount of retrieval support by manipulating data-driven processes. Young and older adults performed a word-stem cued recall task under a low data-driven condition (LDDC) in which the retrieval cue comprised 3 letters, and a higher data-driven condition (HDDC) in which the cue comprised 4 letters. Older adults benefitted more than younger adults from the additional support. Older adults exhibited a large deficit relative to younger adults in the LDDC condition but no age differences were found in the HDDC condition. These findings demonstrate that age-related memory deficits can be reduced by increasing the environmental support at retrieval associated with the data-driven component of retrieval processing. The utility of liquid chromatography coupled to the isotope ratio mass spectrometry technique (LC-IRMS) has already been established through a variety of successful applications. However, the analytical constraint related to the use of aqueous mobile phases limits the LC separation mechanism. We report here a new strategy for high-precision (13)C isotopic analyses based on temperature-programmed LC-IRMS using aqueous mobile phases. Under these conditions, the isotopic precision and accuracy were studied. On one hand, experiments were carried out with phenolic acids using isothermal LC conditions at high temperature (170 degrees C); on the other hand, several experiments were performed by ramping the temperature, as conventionally used in a gas chromatography-based method with hydrosoluble fatty acids and pulses of CO 2 reference gas. In isothermal conditions at 170 degrees C, despite the increase of the CO 2 background, p-coumaric acid and its glucuronide conjugate gave reliable isotopic ratios compared to flow injection analysis-isotopic ratio mass spectrometry (FIA-IRMS) analyses (isotopic precision and accuracy are lower than 0.3 per thousand). On the opposite, for its sulfate conjugate, the isotopic accuracy is affected by its coelution with p-coumaric acid. Not surprisingly, this study also demonstrates that at high temperature (170 degrees C), a compound eluting with long residence time (i.e., ferulic acid) is degraded, affecting thus the delta (13)C (drift of 3 per thousand) and the peak area (compared to FIA-IRMS analysis at room temperature). Quantitation is also reported in isothermal conditions for p-coumaric acid in the range of 10-400 ng/mL and with benzoic acid as an internal standard. For temperature gradient LC-IRMS, in the area of the LC gradient (set up at 20 degrees C/min), the drift of the background observed produces a nonlinearity of SD (delta (13)C) approximately 0.01 per thousand/mV. To circumvent this drift, which impacts severely the precision and accuracy, an alternative approach, i.e., eluting the compound on the plateau of temperature studied was reported here. Other experiments with temperature-programmed LC-IRMS experiments are also reported with the presence of methanol in the injected solution to mimic residual solvent originating from the sample preparation or to slightly increase the solubility of the targeted compound for high-precision measurement.  相似文献

Surface coal mining is the dominant form of land cover change in Central Appalachia, yet the extent to which surface coal mine runoff is polluting regional rivers is currently unknown. We mapped surface mining from 1976 to 2005 for a 19,581 km(2) area of southern West Virginia and linked these maps with water quality and biological data for 223 streams. The extent of surface mining within catchments is highly correlated with the ionic strength and sulfate concentrations of receiving streams. Generalized additive models were used to estimate the amount of watershed mining, stream ionic strength, or sulfate concentrations beyond which biological impairment (based on state biocriteria) is likely. We find this threshold is reached once surface coal mines occupy >5.4% of their contributing watershed area, ionic strength exceeds 308 μS cm(-1), or sulfate concentrations exceed 50 mg L(-1). Significant losses of many intolerant macroinvertebrate taxa occur when as little as 2.2% of contributing catchments are mined. As of 2005, 5% of the land area of southern WV was converted to surface mines, 6% of regional streams were buried in valley fills, and 22% of the regional stream network length drained watersheds with >5.4% of their surface area converted to mines.  相似文献

The performance of InGaP-based pHEMTs as a function of gate length has been examined experimentally. The direct-current and microwave performance of pHEMTs with gate lengths ranging from 1.0-0.2 μm has been evaluated. Extrinsic transconductances from 341 mS/mm for 1.0 μm gate lengths to 456 mS/mm for 0.5 μm gate lengths were obtained. High-speed device operation has been verified, with ft of 93 GHz and fmax of 130 GHz for 0.2 μm gate lengths. The dependence of DC and small-signal device parameters on gate length has been examined, and scaling effects in InGaP-based pHEMT's are examined and compared to those for AlGaAs/InGaAs/GaAs pHEMTs. High-field transport in InGaP/InGaAs heterostructures is found to be similar to that of AlGaAs/InGaAs heterostructures. The lower ϵr of InGaP relative to AlGaAs is shown to be responsible for the early onset of short-channel effects in InGaP-based devices  相似文献

Hydrogenated microcrystalline silicon prepared at low temperatures by the glow discharge technique is examined here with respect to its role as a new thin-film photovoltaic absorber material. XRD and TEM characterisations reveal that microcrystalline silicon is a semiconductor with a very complex morphology. Microcrystalline p–i–n cells with open-circuit voltages of up to 560–580 mV could be prepared. “Micromorph” tandem solar cells show under outdoor conditions higher short-circuit currents due to the enhanced blue spectra of real sun light and therefore higher efficiencies than under AM1.5 solar simulator conditions. Furthermore, a weak air mass dependence of the short-circuit current density could be observed for such micromorph tandem solar cells. By applying the monolithic series connection based on laser patterning a first micromorph mini-module (total area of 23.6 cm2) with 9% cell conversion efficiency could be fabricated.  相似文献
